1. Whole body protein synthesis was measured in chick embryos cultured in vitro. On day 7 of incubation chick embryos were cultured for 60 min in synthetic serum‐free medium containing 4‐[3H]phenylalanine. Specific radioactivities in free and protein‐bound phenylaline in the whole embryo were measured, starting 2 min after commencement of the culture process.
2. The values for fractional synthesis rate (FSR) estimated in vitro at 20, 30, 45 and 60 min during the embryo culture agreed well, ranging from 35 to 40%/d, suggesting that the method would serve as a useful model for studying the effect of growth promoters in chick embryos.
3. Bovine insulin in the synthetic medium did not affect FSR of protein in chick embryos cultured in vitro. 相似文献

The electrochemical behavior of a single molecule can be observed by trapping a small volume of a dilute solution of the electroactive species between an ultramicroelectrode tip with a diameter of approximately 15 nanometers and a conductive substrate. A scanning electrochemical microscope was used to adjust the tip-substrate distance ( approximately 10 nanometers), and the oxidation of [(trimethylammonio)methyl] ferrocene (Cp(2)FeTMA(+)) to Cp(2)FeTMA(2+) was carried out. The response was stochastic, and anodic current peaks were observed as the molecule moved into and out of the electrode-substrate gap. The self-assembly of monodisperse gold and silver colloid particles into monolayers on polymer-coated substrates yields macroscopic surfaces that are highly active for surface-enhanced Raman scattering (SERS). Particles are bound to the substrate through multiple bonds between the colloidal metal and functional groups on the polymer such as cyanide (CN), amine (NH(2)), and thiol (SH). Surface evolution, which can be followed in real time by ultraviolet-visible spectroscopy and SERS, can be controlled to yield high reproducibility on both the nanometer and the centimeter scales. On conducting substrates, colloid monolayers are electrochemically addressable and behave like a collection of closely spaced microelectrodes. These favorable properties and the ease of monolayer construction suggest a widespread use for metal colloid-based substrates. 相似文献

Much attention is focused today on predicting how plants will respond to anticipated changes in atmospheric composition and climate, and in particular to increases in CO(2) concentration. Here we review the long-term global fluctuations in atmospheric CO(2) concentration as a framework for understanding how current trends in atmospheric CO(2) concentration fit into a selective, evolutionary context. The relationship between fine-scale spatial patterns of forage abundance and the feeding patterns of large ungulates is not well known. We compared these patterns for areas grazed in winter by elk and bison in a sagebrush-grassland landscape in northern Yellowstone National Park. At a fine scale, the spatial distribution of mapped feeding stations in 30 m × 30 m sites was found to be random where there were no large patches devoid of vegetation. In areas similar to the mapped sites, the underlying spatial distribution pattern of biomass was also determined to be random. At a broad scale, forage biomass differed among communities across the northern range but forage quality did not. These results suggest that ungulates are feeding randomly within forage patches (fine scale) but may select feeding sites based upon forage abundance at broader, landscape scales. Lamb copper status is commonly assessed by measuring copper concentrations in four liver samples collected from lines of lambs sent to meat slaughtering premises. High liver copper concentrations were found in lambs examined in April and May. Five farms with two or more lamb liver copper concentrations greater than 3000 micromol/kg on a wet matter basis and two farms with adequate copper concentrations but less than 3000 micromol/kg in the autumn of 1992 were selected for a more detailed investigation into the factors affecting their lamb copper status in 1993. High liver copper status was associated with low pasture molybdenum, grazing paddocks recently topdressed with copper sulphate, supplementing with mineralised drenches and copperised salt licks, and the high copper content in chicory. Lamb liver copper concentrations were significantly (p<0.05) higher in the autumn than in the summer on two of three farms. 相似文献
