Recent evidence has indicated the presence of IGF-I and IGF-I receptors in mammalian and teleost ovarian follicles. Since growth hormone (GH), which can be secreted from the pituitary concomitant with a gonadotropin as a response to gonadotropin-releasing hormone, generally acts to release IGF-I from tissues including the ovary, the effect of IGF-I itself on ovarian steroidogenesis and oocyte maturation was investigated in the model teleost, Fundulus heteroclitus. IGF-I was found to be without effect on ovarian follicle steroidogenesis, but initiated oocyte maturation in a dose-dependent manner even more rapidly and effectively than 17,20-dihydroxy-4-pregnene-3-one (DHP), the naturally occurring maturation-inducing steroid. IGF-II also induced oocyte maturation in a dose-dependent manner. IGF-I induction of oocyte maturation occurred in the absence of DHP production by the granulosa cells (which is normally stimulated by gonadotropin), and could be inhibited by cycloheximide but not actinomycin D, thus implicating the role of protein synthesis. These results suggest that GH-stimulated release of ovarian IGF-I may have an even more direct role than DHP on the reinitiation of oocyte maturation.  相似文献

Mature black sea bass, Centropristis striata L. (200–800 g), were captured in coastal South Carolina during the spawning season and administered hormones for ovulation induction and strip spawning. During both study years, control groups of females were incorporated into the study design and administered sham injections containing physiological saline solution. In 2004, females received a single intramuscular injection of human chorionic gonadotropin (hCG) (330 IU kg−1) (n=8) or two injections of hCG at 24‐h intervals (n=8). In 2005, females received a single injection of hCG (n=10) or an analogue of luteinizing hormone releasing hormone (LHRHa) (n=10). In 2004, all fish administered a single dose of hCG ovulated at least once. Six fish ovulated on two consecutive days and one fish ovulated on 3 days consecutively. In contrast, six of eight fish receiving two doses of hCG ovulated once, five ovulated on 2 days successively and three fish ovulated 3 days in succession. Of the fish that spawned, no differences were found in any reproductive parameters. In 2005, all fish administered hCG or LHRHa ovulated at least once. Three fish administered hCG ovulated twice, four fish ovulated on three consecutive days and one fish 4 days successively. All fish administered LHRHa spawned at least twice, six fish ovulated thrice and three fish ovulated 4 days, successively. A significant difference in fertility was found between hCG (75.6±11.4%) and LHRHa (55.6±27.4%). The results of this study indicate that both hCG and LHRHa are effective for ovulation induction in prespawning black sea bass.  相似文献

Iceland scallops, Chlamys islandica (O.F. Müller), were grown in hanging culture at 3 different depths (2, 12 and 40 m below the sea surface) in a northern Norwegian fjord. The growth of the soft parts of the body and the shell increment were monitored throughout the growing season (March until September). At the same time, temperature, salinity, water density, current speed, chlorophyll, particulate organic matter (POM) and particulate inorganic matter (PIM) were monitored at each of the experimental depths. Analysis of the environmental data together with the growth data showed that growth differences between the experimental depths were essentially the results of differences in nutritional conditions for the scallops, defined by the relationship between PIM and POM in the water column. The results also showed that growth in Iceland scallops can be increased significantly by suspending the animals in a more favourable nutritional environment than that found on the natural scallop beds.  相似文献

Piscine orthoreovirus (PRV) belongs to the Reoviridae family and is the only known fish virus related to the Orthoreovirus genus. The virus is the causative agent of heart and skeletal muscle inflammation (HSMI), an emerging disease in farmed Atlantic salmon (Salmo salar L.). PRV is ubiquitous in farmed Atlantic salmon and high loads of PRV in the heart are consistent findings in HSMI. The mechanism by which PRV infection causes disease remains largely unknown. In this study we investigated the presence of PRV in blood and erythrocytes using an experimental cohabitation challenge model. We found that in the early phases of infection, the PRV loads in blood were significantly higher than in any other organ. Most virus was found in the erythrocyte fraction, and in individual fish more than 50% of erythrocytes were PRV-positive, as determined by flow cytometry. PRV was condensed into large cytoplasmic inclusions resembling viral factories, as demonstrated by immunofluorescence and confocal microscopy. By electron microscopy we showed that these inclusions contained reovirus-like particles. The PRV particles and inclusions also had a striking resemblance to previously reported viral inclusions described as Erythrocytic inclusion body syndrome (EIBS). We conclude that the erythrocyte is a major target cell for PRV infection. These findings provide new information about HSMI pathogenesis, and show that PRV is an important factor of viral erythrocytic inclusions.  相似文献

  1. The paper ‘Biodiversity values of remnant freshwater floodplain lagoons in agricultural catchments: evidence for fish of the Wet Tropics bioregion, northern Australia’, published in Aquatic Conservation: Marine and Freshwater Ecosystems in 2015, has contributed in several ways to the integration of freshwater wetland science within new catchment management policies and practices for Great Barrier Reef (GBR) sustainability.
  2. The Tully–Murray biodiversity study developed novel protocols to sample larval, juvenile, and adult fish life‐history stages in floodplain lagoons using a combination of boat‐based backpack electrofishing and fyke netting. In addition, hydrological and hydrodynamic models were applied in a completely new way to quantify the timing, extent, and duration of water connectivity across floodplain streams, cane drains, and wetlands. Combining the two novel approaches enabled an analysis of lagoon fish assemblage patterns in relation to environmental gradients, especially floodplain hydrology, connectivity patterns, and measures related to agricultural land use.
  3. In demonstrating the importance of different levels of connectivity for different biodiversity outcomes in freshwater floodplain lagoons of the Tully–Murray catchment, the subject paper established that floodplain connectivity needs to be taken into consideration in wetland management practices.
  4. The timing of the subject publication was fortuitous. It coincided with the preparation of the evidence‐based 2017 Scientific Consensus Statement on land‐based water quality impacts on the GBR. As one of the few freshwater wetland ecology publications for the catchments of the GBR at that time, this paper played an important role in demonstrating freshwater wetland values, fish conservation options, and management imperatives to sustain wetland ecological health and services in GBR catchments.

By advancing the understanding of factors driving biodiversity patterns, and the importance of connectivity and ecohydrological processes in freshwater floodplain wetlands of the GBR catchment, the Tully–Murray study helped to drive new policy directives for the protection and restoration of catchment, floodplain, and estuary functions, and connectivity, now embedded in the Reef 2050 Long‐Term Sustainability Plan 2018, an overarching strategy for managing the GBR over the next 35 years, and complementary Queensland environmental legislation.

Purpose

This study aimed to compare the effectiveness of chemical-enhanced soil washing (with chelating agents, humic substances and inorganic acids) and soil stabilisation by inorganic industrial by-products (coal fly ash, acid mine drainage sludge and zero-valent iron) and organic resource (lignite) for timber treatment site remediation.

Materials and methods

Both remediation options were assessed in terms of extraction/leaching kinetics and residual leachability (toxicity characteristic leaching procedure, TCLP) of the major risk drivers, i.e. Cu and As.

Results and discussion

In chemical-enhanced soil washing, chelating agents only minimised the Cu leachability. Humic substances were ineffective while inorganic acids reduced the As leachability to the detriment of the soil quality. For the waste-stabilised soil, the short-term leaching potential (72 h) and long-term TCLP leachability (9 months) revealed that Fe-/Al-/Ca-rich AMD sludge and coal fly ash sequestered As through adsorption and (co-)precipitation, while carbonaceous lignite stabilised Cu with oxygen-containing functional groups. The short-term and long-term leaching of Cu and As into the soil solution was negligible in the presence of the waste materials. However, the waste-stabilised soil did not maintain sufficient Cu stability in the TCLP tests, in which acetate buffer induced significant mineral dissolution of the waste materials.

Conclusions

These results suggest that chelant-enhanced washing (significant reduction of Cu leachability) may be augmented with subsequent stabilisation with inorganic waste materials (effective control of As leachability), thus minimising the environmental risks of both Cu (heavy metal) and As (metalloid) while preserving the reuse value of the soil. Additional tests under field-relevant conditions are required to provide a holistic performance evaluation.  相似文献

Turbulence can greatly influence reaction and heat transfer rates in fluids. The topology of the three-dimensional interface between mixing fluids directly determines the location and degree of reaction. The time-resolved measurement of the three-dimensional concentration field in a transitional gas jet is reported. A thin sheet of laser light was swept through the flow volume by a rotating mirror in a time brief enough that motion of the gas was minimal. The light sheet illuminated different parallel planes within the flow, and light scattered from particles seeding the jet was imaged onto a detector. The series of two-dimensional measurements made during one scan of the flow volume constituted a full three-dimensional mapping of structures within the flow. Computer graphics software was used to reconstruct and visualize three-dimensional surfaces of constant concentration and the magnitude of the concentration gradient vector over such surfaces.  相似文献

Pollen of Brassica exhibited strong self-incompatibility. It did not germinate on, adhere to, or extend into the stigmatic tissues of the female parent plant. In contrast, pollen that had been treated with cycloheximide germinated on and penetrated into self-stigmas to the same degree as untreated pollen placed on crossstigmas (compatible). Germ tubes from cycloheximide-treated pollen did not continue growth through stylar tissues and therefore were unable to effect fertilization.  相似文献

Using inorganic carbon measurements from an international survey effort in the 1990s and a tracer-based separation technique, we estimate a global oceanic anthropogenic carbon dioxide (CO2) sink for the period from 1800 to 1994 of 118 +/- 19 petagrams of carbon. The oceanic sink accounts for approximately 48% of the total fossil-fuel and cement-manufacturing emissions, implying that the terrestrial biosphere was a net source of CO2 to the atmosphere of about 39 +/- 28 petagrams of carbon for this period. The current fraction of total anthropogenic CO2 emissions stored in the ocean appears to be about one-third of the long-term potential.  相似文献

Physicochemistry and structural studies of two types of japonica rice, low amylose Calmochi-101 (CM101) and intermediate amylose M-202 (M202), were conducted to determine similarities and differences between the rices perhaps attributable to amylose content differences. The rheological behavior of the gelation and pasting processes of flours and starches was determined with high accuracy and precision using a controlled stress rheometer. Fat and protein, although minor constituents of milled rice, were shown to have significant effects on the physicochemical and pasting properties of starches and flours. Removal of protein and lipids with aqueous alkaline or detergent solutions caused lower pasting temperatures and higher overall viscosity in both starches, compared with their respective flours. There was less viscosity difference between M202 flour and its starch when isolated by enzymatic hydrolysis of protein. The protease did not reduce internally bound lipids, suggesting that fats help to determine pasting properties of rice flours and their respective starches. Structural integrity differences in individual granules of waxy and nonwaxy rice flours, starches, and whole raw, soaked, and cooked milled grain were revealed by fracture analysis and scanning electron microscopy. Calmochi 101 and M202 did not differ in weight-averaged molar mass (Mw) and root-mean-square radii (Rz) between flours and starches, as determined by high-performance size exclusion chromatography (HPSEC) and multiple-angle laser light scattering (MALLS) (Park, I.; Ibanez, A. M.; Shoemaker, C. F. Starch 2007, 59, 69-77).  相似文献
