Postmortem proteolysis is an important determinant of beef tenderness. Caspase 3 is a protease that functions in apoptosis and has been shown to degrade myofibrillar proteins. Our objective was to evaluate whether caspase 3 activity is related to beef tenderness and muscle growth, and whether caspase 3 is activated in postmortem beef muscle. In experiment 1, longissimus thoracis (LT) and sternomandibularis muscle samples were obtained at 0, 0.25, 1, 3, 24, 72, and 240 h postmortem from 5 steers. In experiment 2, a group of 40 beef cattle was slaughtered at the University of Wyoming Meat Lab with 10 steers of different tenderness and growth characteristics chosen for the analysis of caspase 3 activity in the LT. In experiment 3, 10 steers with different tenderness but matched growth characteristics were chosen for analyses. In experiment 1, no significant activation (P = 0.70) of caspase 3 activity was detected; only a decreased activity at 72 (P = 0.05) and 240 h (P = 0.02) postmortem was observed. Western blot analysis of both muscle samples showed only the pro-caspase 3 form and failed to detect the activated enzyme. In experiment 2, caspase 3 activity in the LT immediately postmortem was greater (P = 0.05) for the cattle with increased Warner-Bratzler shear force values. No difference in caspase 3 activity was detected for experiment 3. Our results demonstrate that caspase 3 activity is not activated, with its activity decreasing with time postmortem, and caspase 3 activity is not associated with Warner-Bratzler shear force at slaughter. Interferon (IFN)-stimulated gene (ISG) 56 family (composed of ISG54, ISG56, ISG58, and ISG60) plays important roles in defense against viral infection in mammalian cells. Numerous studies have been conducted on ISG54, ISG56, and ISG60; however, little is known on ISG58. In the present study, the upstream sequence of porcine ISG58 gene was first characterized as functional promoter by luciferase reporter assay, and then two directly adjacent IFN-stimulated response elements (ISREs), one at ?206 to ?194 (ISRE-I) and a second one, directly upstream of this element at ?219 to ?207?bp (ISRE-II), were identified using the bioinformatics method. The subsequent site-directed deletion and transient transfection experiments showed the candidate ISREs are functional. ISRE-I works better than ISRE-II and synergistic cooperation exists between two ISREs. Additionally, the effect of porcine ISG58 on activation of NF-κB was analyzed using the dual-luciferase reporter assay. 通过实地考察和植物标本的采集与鉴定,并结合相关文献资料的查阅,对新疆西昆仑山禾本科植物物种多样性特点进行了研究。结果表明,新疆西昆仑山的禾本科植物共计37属137种,分别占新疆禾本科植物属、种总数的52.11%和42.55%,属种丰富度比较高;其中发现该区新记录植物6种。本区禾本科植物特有现象不明显,属的组成以仅含1种的属(17属17种)和少种属(13属35种)为主,共计30属52种,分别占属、种总数的81.08%和37.96%;中型属(4属32种)和较大属(3属53种)共计7属85种,分别占属、种总数的18.92%和62.04%,在种级水平占据明显优势,是该区系的主体。区系地理成分有6个分布类型和4个变型,以温带分布及其变型为主。植物生活型以多年生草本(116种,占84.67%)为主,其次为一年生草本(21种,占15.33%)。生态型以中生植物(63种,占45.99%)和旱生植物(61种,占44.53%)为主,显示其与草原、草甸的密切关系,另有少量湿生植物(13种,占9.49%)。说明该地区比较适合禾本科多年生的旱生和中生植物生长,且该区系的禾本科植物地理成分类型与该区所处地理位置为温带较吻合。 相似文献

Spirocerca lupi is a nematode mainly affecting dogs but has been found in other animals, particularly carnivores. Anatomical areas of typical and aberrant migration vary. This report describes four cases of Spirocerca lupi causing neurological symptoms, similar to thoracolumbar disc syndrome, as a result of aberrant migration of the nematode into the spinal canal. In two of the cases, the nematode could be demonstrated intraoperatively. The third was discovered on post-mortem examination, while the fourth case displayed compelling evidence of Spirocerca lupi involvement. Surgical removal of the Spirocerca lupi nematode would be the treatment of choice in cases of spinal migration, with therapeutic and preventive treatment with doramectin instituted to treat remote sites and prevent re-infection. In areas endemic for spirocercosis, Spirocerca lupi should be considered as an important differential diagnosis in cases that are presented with clinical signs suggestive of a spinal cord lesion. 相似文献

采用呼吸抑制剂测定了结缕草休眠与打破休眠种子在发芽过程中的各个呼吸代谢途径-糖酵解呼吸途径(EMP)、三羧酸循环呼吸途径(TCA)、磷酸戊糖呼吸途径(PPP)的呼吸速率及其占总呼吸速率的比例,结果表明,休眠种子的总呼吸速率显著低于打破休眠种子的总呼吸速率,造成这种差异的原因是磷酸戊糖途径及三羧酸循环途径的呼吸速率低,这是休眠结缕草种子不能发芽的原因之一。 相似文献
