AIMS: To determine some of the risk factors for cows not observed in oestrus within 35–42 days of an unsuccessful artificial insemination (AI; phantom cows), and the reproductive outcomes and effect of treatment of phantom cows.

MATERIALS AND METHODS: Over 2 years, in dairy herds from the Waikato (n=10) and Canterbury (n=4) regions of New Zealand, pregnancy diagnosis was carried out 35–42 days after AI on cows that had been inseminated in the first 3 weeks after the start of mating (PSM) but had not been seen returning to oestrus. Risk factors for phantom cows were analysed using a generalised linear mixed effect model.

In Year 1, all phantom cows were left untreated. In Year 2, phantom cows were categorised as having a corpus luteum (CL) (CL+ n=120), or having ovarian follicles ≥10 (n=101) or <10 (n=40)?mm in diameter. Cows with a CL were treated with cloprostenol or untreated and placed with bulls. Cows with no CL received intravaginal progesterone (P4) for 7 days, with injection of gonadotrophin-releasing hormone (GnRH) on Days 0 and 9, and cloprostenol on Day 7 followed by AI. Pregnancy diagnosis of all cows took place 100–120 days after PSM and interval to conception and final pregnancy rate determined.

RESULTS: Overall, of cows inseminated in the first 3 weeks after PSM that did not return to oestrus, 610/6,734 (9.1%) were phantom cows. From the final multivariable analysis, treatment for anoestrus, BCS ≤4.0 at mating, being 2 or >6 years of age, and pure-bred, and decreasing interval between calving and mating, until 98 days post calving, were associated with increased odds of being a phantom cow. Compared to all other groups of cows, phantom cows had a longer interval to conception (p<0.001) and a lower final pregnancy rate (p<0.001).

Treatment of CL+ cows or cows with follicles ≥10?mm did not affect reproductive outcomes (p>0.3). For cows with follicles <10?mm treatment decreased the final percentage not pregnant (3/27; 11%; p=0.01) and interval to conception (21 days; p=0.02) compared with controls (7/13; 54% and 37 days, respectively).

CONCLUSIONS AND CLINICAL RELEVANCE: Risk factors for phantom cows were identified that could be manipulated to reduce the number of phantom cows in a herd, in particular increasing BCS. Treatment of the majority of phantom cows did not improve reproductive performance.  相似文献

Verticillium longisporum is one of the major pathogens of oilseed rape (Brassica napus; genome AACC, 2n = 38) in Europe. Current European cultivars possess only a low level of resistance against V. longisporum, meaning that heavy infection can cause major yield losses. The aim of this study was to identify quantitative trait loci (QTL) for resistance against V. longisporum as a starting point for marker-assisted breeding of resistant cultivars. Resistance QTL were localized in a segregating oilseed rape population of 163 doubled haploid (DH) lines derived by microspore culture from the F1 of a cross between two B. napus breeding lines, one of which exhibited V. longisporum resistance derived by pedigree selection from a resynthesized B. napus genotype. A genetic map was constructed comprising 165 restriction fragment length polymorphism, 94 amplified fragment length polymorphism and 45 simple sequence repeats (SSR) markers covering a total of 1,739 cM on 19 linkage groups. Seedlings of the DH lines and parents were inoculated with V. longisporum isolates in four greenhouse experiments performed in Sweden during autumn 1999. In three of the experiments the DH lines were inoculated with a mixture of five isolates, while in the fourth experiment only one of the isolates was used. The intention was to simulate four different environments with variable disease pressure, while still maintaining uniform conditions in each environment to enable reliable disease scoring. The disease index (DI) was calculated by scoring symptoms on a total of 21 inoculated plants per line in comparison to 21 noninoculated plants per line. Using the composite interval mapping procedure a total of four different chromosome regions could be identified that showed significant QTL for resistance in more than one environment. Two major QTL regions were identified on the C-genome linkage groups N14 and N15, respectively; each of these QTL consistently exhibited significant effects on resistance in multiple environments. The presence of flanking markers for the respective QTL was associated with a significant reduction in DI in the inoculated DH lines.  相似文献

Enzyme-linked immunosorbent assay examination of sera from pigs vaccinated with T. spiralis nativa infective larvae and/or challenged with T. spiralis spiralis larvae using a T. spiralis spiralis excretory-secretory antigen showed a significant cross-reaction between the two species of Trichinella. Eight of 12 pigs vaccinated with a high dose of T. spiralis nativa reacted positively 28 days postvaccination while the remaining four pigs had high but negative ELISA optical density readings. Five of six pigs challenged with the homologous species reacted positively 28 days postchallenge but the sixth pig remained negative despite having a muscle infection of 5.6 larvae/g of musculature.  相似文献

Studies were carried out to find methods for obtaining optimum yields of bovine ephemeral fever (BEF) virus and for concentrating the virus in order to develop inactivated virus vaccines. Cells from the SVP cell line, which was derived from the pig kidney PS cell line, were most satisfactory for growing and assaying BEF virus. BHK 21 and Vero cells also gave similar yields of virus but were not as useful for virus assay. A plaque assay in SVP cells, in which there was 0.1 μg of actinomycin D per ml of overlay, produced reproducible clear plaques and was slightly more sensitive than assays in BHK 21 cell roller tubes. High multiplicities of infection (MOI), around 1 PFU/cell, produced low yields of infectious virus, whereas decreasing the MOI approximately 100-fold led to an increase in virus yield of up to four logs. BEF virus could be concentrated using zinc acetate or ammonium sulphate but not polyethylene glycol 6000. Ammonium sulphate proved most suitable and produced an easily handled precipitate with up to 100% recovery of virus infectively, and 100-fold concentration was possible. This concentrated virus could be rapidly desalted by gel filtration through Sephadex G-75. The virus could be further purified by sucrose density gradient ultracentrifugation provided the gradient contained a protein stabilizer of 0.1% bovine serum albumin. Inactivation kinetics with 0.025% β-propiolactone was similar to that reported for other rhabdoviruses.  相似文献
