There is widespread concern and debate about the state of global marine resources and the ecosystems supporting them, notably global fisheries, as catches now generally stagnate or decline. Many fisheries are not assessed by standard stock assessment methods including many in the world's most biodiverse areas. Though simpler methods using widely available catch data are available, these are often discounted largely because data on fishing effort that contributed to the changes in catches are mostly not considered. We analyse spatial and temporal patterns of global fishing effort and its relationship with catch to assess the status of the world's fisheries. The study reveals that fleets now fish all of the world's oceans and have increased in power by an average of 10‐fold (25‐fold for Asia) since the 1950s. Significantly, for the equivalent fishing power expended, landings from global fisheries are now half what they were a half‐century ago, indicating profound changes to supporting marine environments. - 1. Freshwater ecosystems and their associated biota are among the most endangered in the world. This, combined with escalating human pressure on water resources, demands that urgent measures be taken to conserve freshwater ecosystems and the services they provide. Systematic conservation planning provides a strategic and scientifically defensible framework for doing this.
- 2. Pioneered in the terrestrial realm, there has been some scepticism associated with the applicability of systematic approaches to freshwater conservation planning. Recent studies, however, indicate that it is possible to apply overarching systematic conservation planning goals to the freshwater realm although the specific methods for achieving these will differ, particularly in relation to the strong connectivity inherent to most freshwater systems.
- 3. Progress has been made in establishing surrogates that depict freshwater biodiversity and ecological integrity, developing complementarity‐based algorithms that incorporate directional connectivity, and designing freshwater conservation area networks that take cognizance of both connectivity and implementation practicalities.
- 4. Key research priorities include increased impetus on planning for non‐riverine freshwater systems; evaluating the effectiveness of freshwater biodiversity surrogates; establishing scientifically defensible conservation targets; developing complementarity‐based algorithms that simultaneously consider connectivity issues for both lentic and lotic water bodies; developing integrated conservation plans across freshwater, terrestrial and marine realms; incorporating uncertainty and dynamic threats into freshwater conservation planning; collection and collation of scale‐appropriate primary data; and building an evidence‐base to support improved implementation of freshwater conservation plans.

- 1. SECORE (SExual COral REproduction) Project is an initiative of public aquariums and research institutions to produce and exchange sexual coral recruits for the sustainable management of ex situ populations. Here we present the results of the initial three years (2002–2004).
- 2. Primary polyps (n=501) of corals (Acropora tenuis, Agaricia humilis, Favia fragum) were transported from Rotterdam Zoo to Cologne, Burgers', Hagenbeck and London Zoos, where development of juveniles was monitored for 10 months. All polyps were produced at Rotterdam Zoo from laboratory colonies (A. humilis, F. fragum), and from larvae generated from field collected gametes at Akajima, Okinawa, Japan (A. tenuis). Additionally, planulae of A. tenuis (n=1440) were transported from Rotterdam Zoo to Burgers' Zoo and to London Zoo to obtain primary polyps.
- 3. Larval settlement (A. tenuis) was observed to be 3.00 ± 2.57% (mean ± SD; n=1480) in 2002 and 17.36 ± 6.01% (mean ± SD; n=1480) in 2003, significantly lower compared to settlement at Rotterdam Zoo (57.84 ± 11.01% in 2003; mean ± SD, n=1480). High post‐transport survival rates of 95.18 ± 4.86% (mean ± SD; n=501) were observed in primary polyps of all species.
- 4. Juvenile survival (t=10 months; A. tenuis: 18.4–86.2%; A. humilis: 0–19.7%; F. fragum: 13.3–72.7%) differed significantly between institutions. Mean colony sizes (measured 10 months after transportation) were, in all cases, similar or higher to those reported from literature.
- 5. The results demonstrate the potential of this method to serve as an economical and sustainable alternative to existing mostly exploitative techniques for aquarium stocking. The use of sexual recruits provides an effective and low cost alternative, which is, in principle, applicable to all coral species.
- 6. The project was extended from 9 to 28 institutions across Europe, the USA and Japan in 2004.

Degenerative disorders of motor neurons include a range of progressive fatal diseases such as amyotrophic lateral sclerosis (ALS), spinal-bulbar muscular atrophy (SBMA), and spinal muscular atrophy (SMA). Although the causative genetic alterations are known for some cases, the molecular basis of many SMA and SBMA-like syndromes and most ALS cases is unknown. Here we show that missense point mutations in the cytoplasmic dynein heavy chain result in progressive motor neuron degeneration in heterozygous mice, and in homozygotes this is accompanied by the formation of Lewy-like inclusion bodies, thus resembling key features of human pathology. Intraluminal vesicles of multivesicular endosomes are either sorted for cargo degradation into lysosomes or secreted as exosomes into the extracellular milieu. The mechanisms underlying the sorting of membrane into the different populations of intraluminal vesicles are unknown. Here, we find that cargo is segregated into distinct subdomains on the endosomal membrane and that the transfer of exosome-associated domains into the lumen of the endosome did not depend on the function of the ESCRT (endosomal sorting complex required for transport) machinery, but required the sphingolipid ceramide. Purified exosomes were enriched in ceramide, and the release of exosomes was reduced after the inhibition of neutral sphingomyelinases. RNA polymerase II (Pol II) transcribes genes that encode proteins and noncoding small nuclear RNAs (snRNAs). The carboxyl-terminal repeat domain (CTD) of the largest subunit of mammalian RNA Pol II, comprising tandem repeats of the heptapeptide consensus Tyr1-Ser2-Pro3-Thr4-Ser5-Pro6-Ser7, is required for expression of both gene types. We show that mutation of serine-7 to alanine causes a specific defect in snRNA gene expression. We also present evidence that phosphorylation of serine-7 facilitates interaction with the snRNA gene-specific Integrator complex. These findings assign a biological function to this amino acid and highlight a gene type-specific requirement for a residue within the CTD heptapeptide, supporting the existence of a CTD code. 相似文献

A farmers' knowledge, attitude and practice (KAP) survey was conducted in the highlands of Tigray, northern Ethiopia, to better understand rodent damage and rodent management from the farmers' perspective. Farmers (n = 191) from Dogu'a Temben district, were interviewed using a semi-structured questionnaire. The large majority of the farmers stated that rodents are the main pests in crop fields (92.1%) and storage (88.5%). The farmers (64.2%) reported they experienced 100–500 kg ha−1 damage in crop fields, which is equivalent to 8.9–44.7% loss in annual production. There was some overlap between the most common crops grown in the highlands and the most common crops susceptible to rodent attack. Farmers identified barley as the crop most susceptible to rodent attack (76.4%) and the booting stage as the crop developmental stage with the highest rodent abundance and damage. Rodenticide application was the most commonly practiced management strategy in crop fields (51.8%); in storage, farmers mainly keep domestic cats around granaries (80.6%). We recommend a reduction in reliance on chemical rodenticide in crop fields and a shift to a more sustainable rodent management approach to reduce rodent numbers and damage. 相似文献

Interpretation of the result of a diagnostic test depends not only on the actual test result(s) but also on information external to this result, namely the test's sensitivity and specificity. This external information (also called prior information) must be combined with the data to yield the so-called updated, posterior estimates of the true prevalence and the test characteristics. The Bayesian approach offers a natural, intuitive framework in which to carry out this estimation process. The influence of the prior information on the final result may not be ignored.
