Abstract

A group of spring wheat cultivars originating from Sweden, Finland, Norway, Germany, and the Netherlands was analysed for powdery mildew resistance. Using functional molecular markers, two alleles of the major resistance gene Pm3 were detected among the cultivars under the study. One of the alleles, Pm3d, was detected in the resistant cultivars ‘Vinjett’, ‘SW Estrad’, and ‘Zebra’, and in ‘Tjalve’, a cultivar of earlier release susceptible to the local population of powdery mildew. The second allele of Pm3 detected in the analysed group of cultivars was the allele Pm3f, rarely used in Europe. It was identified in the resistant cultivars ‘Satu’, ‘Helle’, and in the moderatively resistant cultivar ‘Mahti’. Pm3f was found to be effective against the local population of powdery mildew in Estonia, while Pm3d provided no protective effect. Besides the Pm3d allele on chromosome 1A, monosomic analysis of the cultivar ‘Vinjett’,which is almost immune to powdery mildew, identified two additional loci on chromosomes 5D and 7D, respectively, presumably responsible for the high resistance in this cultivar. In contrast to the earlier cultivars, six recently released cultivars (‘Vinjett’, ‘SW Estrad’, ‘Zebra’, ‘Satu’, ‘Helle’, ‘Meri’) demonstrated a high resistance to the powdery mildew fungus Blumeria graminis DC. f. sp. tritici both in the field and seedling tests, showing that the genetic basis of powdery mildew resistance in Nordic spring wheat has been improved noticeably in the last ten years.  相似文献

To analyze the accuracy of the Klason lignin method as applied for the determination of lignin contents in plant based-food products, Klason lignin preparations from curly kale, pears, whole wheat grains, and corn bran were chemically characterized. Characterization included routine ash and protein determinations and the extraction of fat/waxes as well as cutin/suberin depolymerization and extraction of the liberated monomers. Fat/wax and cutin/suberin amounts in the Klason lignin preparations were determined gravimetrically, and their compositions were analyzed by using GC-MS. Typical fat, wax, and cutin (and suberin) constituents such as saturated and unsaturated fatty acids, hydroxy and/or epoxy fatty acids, and phenolic acids were identified in all samples, whereas the detection of long-chain hydrocarbons, alcohols, and ketones, sterols, stanols, and dioic acids was dependent on the sample analyzed. Estimation of the contribution of non-lignin compounds to the Klason lignin contents reduced the noncorrected Klason lignin contents of the insoluble fibers from 28.7% (kale), 22.8% (pear), 14.8% (wheat), and 9.9% (corn) to maximum lignin contents of 6.5% (kale), 16.4% (pear), 4.9% (wheat), and 2.3% (corn). These data demonstrate that certain commonly used statements such as "cereal brans are highly lignified" need to be revised.  相似文献

As indicators of the early stage of the Maillard reaction in carrots, N-(furoylmethyl) amino acids (FMAAs) formed during acid hydrolysis of the corresponding Amadori products were analyzed using RP-HPLC with UV detection. N(ε)-FM-Lys (furosine), FM-Gly, FM-Ala, FM-Val, FM-Ile, FM-Leu, and FM-GABA were identified using synthesized standard material by means of mass spectrometry. Furthermore, N(ε)-carboxymethyllysine (CML) and pyrraline were analyzed as indicators for advanced stages of glycation. For commercial samples with high water content, the formation of Amadori compounds predominates, whereas the advanced stage of Maillard reaction plays only a minor part. Carrot juices, baby food, and tinned carrots showed quite low rates of amino acid modification up to 5%. For dehydrated carrots, significantly higher values for Amadori products were measured, corresponding to a lysine derivatization of up to 58% and nearly 100% derivatization of GABA. Drying experiments revealed great differences in reactivity between the amino acids studied. Whereas furosine reached constant values quite quickly, some FMAAs showed a continuous increase with heating time, indicating that selected FMAAs can be used as a hallmark for the early Maillard reaction to control processing conditions.  相似文献

Fusarium subglutinans causes maize ear rot and contaminates grain with the mycotoxin moniliformin. Previous DNA sequence analysis divided F. subglutinans from maize into two cryptic species, designated groups 1 and 2. Here, it was determined whether the two groups differ in the agriculturally important traits of virulence on maize and moniliformin production in planta. Thirty-seven strains from U.S. maize were assigned to groups 1 and 2 by DNA sequence analysis. In field tests, all strains were highly virulent on maize inbred B73 and four maize hybrids. In planta, 82% of group 1 strains and 25% of group 2 strains produced high levels (100-1500 microg/g) of moniliformin. All group 2 strains from more northern states produced little or no moniliformin (0-5 microg/g). These data indicate that moniliformin production is highly variable in F. subglutinans from U.S. maize and that production may not be required for the fungus to cause maize ear rot.  相似文献

The influence of the addition of metal chelators on oxidative stability was studied in a milk drink and in a mayonnaise system containing highly polyunsaturated lipids. Milk drinks containing 5% (w/w) of specific structured lipid were supplemented with lactoferrin (6-24 muM) and stored at 2 degrees C for up to 9 weeks. Mayonnaise samples with 16% fish oil and 64% rapeseed oil (w/w) were supplemented with either lactoferrin (8-32 muM), phytic acid (16-124 muM), or EDTA (16-64 muM) and were stored at 20 degrees C for up to 4 weeks. The effect of the metal chelators was evaluated by determination of peroxide values, secondary volatile oxidation products, and sensory analysis. Lactoferrin reduced the oxidation when added in concentrations of 12 muM in the milk drink and 8 muM in the mayonnaise, whereas it was a prooxidant at higher concentrations in both systems. In mayonnaise, EDTA was an effective metal chelator even at 16 muM, whereas phytic acid did not exert a distinct protective effect against oxidation. The differences in the equimolar effects of the metal chelators are proposed to be due to differences in their binding constants to iron and their different stabilities toward heat and low pH.  相似文献

The bioavailability of acylated vs nonacylated anthocyanins and the effect of cooking and dose on the comparative bioavailability were investigated in a clinical feeding study using purple carrots as the anthocyanin source. Treatments were purple carrots as follows: 250 g raw (463 micromol of anthocyanins: 400 micromol acylated, 63 micromol nonacylated), 250 g cooked (357 micromol of anthocyanins: 308.5 micromol acylated, 48.5 micromol nonacylated), and 500 g cooked (714 micromol of anthocyanins: 617 micromol acylated, 97 micromol nonacylated). Four of the five carrot anthocyanins were found intact in plasma by 30 min after carrot consumption and peaked between 1.5 and 2.5 h. Acylation of anthocyanins resulted in an 11-14-fold decrease in anthocyanin recovery in urine and an 8-10-fold decrease in anthocyanin recovery in plasma. Cooking increased the recovery of nonacylated anthocyanins but not acylated anthocyanins. Large dose size significantly reduced recovery of both acylated and nonacylated anthocyanins, suggesting saturation of absorption mechanisms.  相似文献

Antioxidant capacity of hydrophilic and lipophilic extracts from eight broccoli genotypes was compared using the oxygen radical absorbance capacity (ORAC) assay. Each genotype was analyzed for carotenoid, tocopherol, ascorbic acid, and flavonoid content. Results indicate that the antioxidant capacity of hydrophilic extracts ranged from 65.8 to 121.6 micromol trolox equivalents (TE)/g of tissue, and the capacity of lipophilic extracts ranged from 3.9 to 17.5 micromol TE/g. Ascorbic acid and flavonoid content of the hydrophilic extracts did not explain the total variation in antioxidant capacity of those extracts, suggesting either the presence of other antioxidant components that have yet to be identified or that the known antioxidants are producing synergistic effects. The carotenoids did correlate with antioxidant capacity of the lipophilic extracts and accounted for the majority of the variability in that fraction. The variability in hydrophilic and lipophilic antioxidant capacity found among these genotypes suggests that potential efficacy from antioxidants will vary considerably from genotype to genotype.  相似文献

A new approach in soil remediation washing techniques is the use of l-cysteine based on the formation of organic complexes. In this study, the applicability of l-cysteine for the mobilisation of different mercury species from contaminated soils was evaluated. Soils were treated with l-cysteine solutions with S?CHg molar ratios of 1, 2, 10, 20, 100 and 200. In 24?h batch experiments, leachates with water could mobilise 1% of Hg. The addition of l-cysteine led to an increase of Hg mobilisation of 42% for soils with inorganically bound Hg. In column experiments, the maximum Hg removal rate was 75%. For soils with organically bound Hg or HgS, only 1?C5% of Hg was mobilised. Thus, the extraction of Hg from soils with l-cysteine is highly dependent on the Hg-binding form. Hg speciation analyses of leachates indicate that Hg?Cl-cysteine complexes are labile complexes which can be easily transformed. Soil samples speciation analysis revealed that reduction to elemental mercury takes place at low S?CHg ratios (1 to 10), assumingly by microbial activity. At higher S?CHg ratios of 10 and 100, precipitation of stable Hg?CS complexes could be observed. These species transformation processes are limitations for considering l-cysteine leaching as a remediation strategy.  相似文献
