OBJECTIVE: To evaluate semiquantitative and quantitative assays for microalbuminuria and determination of the urine albumin-creatinine (UAC) ratio in detection of systemic disease in dogs without overt proteinuria. DESIGN: Prospective study. ANIMALS: 408 dogs. PROCEDURES: Urine samples that had been obtained from dogs for which a complete medical record was available and in which results of a dipstick test for urine protein were negative were evaluated. Urine protein-creatinine ratios (cutoff values, 0.5 and 0.1), semiquantitative and quantitative microalbuminuria values (cutoff value, 1 mg/dL), and UAC ratios (cutoff values, 100 and 200 mg/g) were determined. Clinical diagnoses rendered within 3 months of enrollment in the study were recorded. Sensitivity and specificity were determined with disease status serving as the standard. Associations with clinical diagnosis, sex, age, BUN and serum creatinine concentrations, blood pressure, results of bacterial culture of urine, temperature, pyuria, hematuria, and bacteriuria were evaluated by use of logistic regression analysis. RESULTS: 48 dogs were healthy, and 360 had at least 1 disease. Significant associations were detected between age, presence of disease, presence of neoplastic disease, BUN and serum creatinine concentrations, and hematuria and results of 1 or both of the microalbuminuria assays. CONCLUSIONS AND CLINICAL RELEVANCE: Microalbuminuria was associated with underlying disease. The sensitivity and specificity of the semiquantitative microalbuminuria test for detection of systemic disease were superior to those of other tests. Microalbuminuria testing in conjunction with other screening procedures may increase diagnosis of subclinical disease, but a prospective study in which the predictive values of screening tests are evaluated, with and without microalbuminuria determination, is needed.  相似文献

Yellow starthistle (Centaurea solstitialis L.) is a nonnative pest of rangelands that decreases forage quality and yield. Mowing may control starthistle effectively and complement herbicide use in an integrated pest management strategy, but little research has investigated its effects on nontarget vegetation. We monitored biomass and seedbank size of annual and perennial species, in addition to starthistle, in response to 3 yr of mowing treatments, either mowing alone or in combination with solarization tarps or thatch removal. All mowing treatments were very effective at reducing starthistle biomass and seedbank: mowing alone reduced biomass 92 ± 2%, mowing with thatch removal 91 ± 1%, and mowing with solarization 95 ± 1%. Compared to seedbank sizes in the control plots, yellow starthistle seedbank decreased by 100% (mowing alone), 92% (mowing + thatch removal), and 100% (mowing with solarization) after 3 yr of treatment. Mowing also significantly improved perennial species' biomass. Annual species' biomass varied on a year-to-year basis but was not significantly affected by any treatment. Seedbank sizes of annuals and perennials also did not differ according to mowing treatment. This research indicates that late-season mowing can effectively reduce starthistle biomass without adverse effects on other vegetation and that mowing alone is sufficient to reduce starthistle seedbank size without additional methods of decreasing seed rain.  相似文献

Target region amplification polymorphism (TRAP) is a relatively new PCR-based technique that detects large numbers of loci in a single reaction without extensive pre-PCR processing of samples. The aim of this study was to integrate TRAP markers in an EST-derived SSR linkage map of a RIL mapping population from the cross of the durum wheat cultivars Ciccio and Svevo, for a more general purpose of establishing a high-throughput system for genetic map saturation. Primer combinations producing PCR products with at least 4–5 polymorphic bands were selected and analyzed across the mapping population. The PCR reactions produced a total of 2,881 fragments with an average of 52 peaks per reaction. A total of 142 new TRAP markers were mapped and found to be randomly distributed in the genome. The total length of the map was 2,043.0 cM, with an average chromosome length of 145.9 cM. Homoeologous group one had the highest number of TRAP markers (38 loci) and the longest map length (407.9 cM) for a total of 87 markers, while the homoeologous group five had the lowest TRAP marker number (5 loci) and the shortest map length (232.5 cM). The distribution of markers among the seven homoeologous groups was random. The results indicate that TRAP is highly efficient in genetic mapping, generating a large number of markers scattered across the genome. This closes many existing gaps in marker coverage and may join otherwise separate linkage groups.  相似文献

Mediterranean ecosystems are characterized by large arid areas where the patchy distribution of trees offers little protection against harsh climate conditions for seedling establishment. Climate change is predicted to result in an increase in these arid regions, with pronounced effects on vegetation. Production of seedlings with developed ectomycorrhizas is a promising strategy for minimizing the initial transplant shock, thereby increasing plant survival and growth during the first, most critical years of a plantation. One important species in the Mediterranean basin is Quercus suber (cork oak), which occurs, together with other evergreen oak species, in an agro-silvo-pastoral system that represents an example of sustainable land use in Europe. In this study, a Pisolithus tinctorius isolate was used for ectomycorrhizal colonization of cork oak nursery seedlings, and the effects on aboveground plant growth and leaf structural and physiological parameters were investigated. Ectomycorrhizal development resulted in a significant increase in leaf area, dry weight, nitrogen content, and photosynthetic pigments, and mycorrhizal plants showed a higher photosynthetic capacity and water use efficiency. Nursery-inoculated plants established in the field showed increased survival and growth during the first year after transplant. These results indicate a potential for further enhancing the use of mycorrhizal inoculation as a cultivation practice in forest nurseries. Considering the difficulty of soil restoration under limiting environmental conditions, nursery inoculation with ectomycorrhizal fungi can be an important advantage for improving the quality of seedling stock and its performance after out-planting in the field, benefiting the regeneration of arid regions and the reintroduction of inocula of ectomycorrhizal fungi into these areas.  相似文献

Teladorsagia circumcincta is one of the most economically important gastrointestinal nematode parasites of sheep in cool temperate regions, to which sheep show genetically-varying resistance to infection. This is a very common parasite and viable sheep production requires the extensive use of anthelmintic drugs. However, the emergence of drug-resistant parasites has stimulated the search for alternative control strategies to curb production losses. Lambs become infected soon after weaning and begin to control parasite burden within 8–10 weeks of continual infection. This control is an acquired characteristic mediated by the development of parasite-specific antibodies. This paper describes the immunology associated with resistance and susceptibility, focussing on differential T cell activation that regulates the production of specific effector mechanisms. It continues by summarizing the methods used to identify genes that could be exploited as molecular markers of selection for resistance. In particular it focusses on the link between understanding the molecular immunology of infection and the identification of candidate genes for selection.  相似文献
