The Rac1 guanosine triphosphatase (GTPase) has been implicated in multiple cellular functions, including actin dynamics, proliferation, apoptosis, adhesion, and migration resulting from signaling by multiple receptors, including the B cell antigen receptor (BCR). We used conditional gene targeting to generate mice with specific Rac1 deficiency in the B cell lineage. In the absence of both Rac1 and the highly related Rac2, B cell development was almost completely blocked. Both GTPases were required to transduce BCR signals leading to proliferation, survival and up-regulation of BAFF-R, a receptor for BAFF, a key survival molecule required for B cell development and maintenance. 相似文献

Eco-tourism depending on wildlife is becoming increasingly profitable and landowners are beginning to favor game farming and ecotourism. In these areas, large-scale translocation of wildlife involves a diversity of species and large populations. The African buffalo (Syncerus caffer) is one of the major tourist attractions in Zambia. It accounts for 8.7% and 12.4% of the total animal species hunted in the Game Management Areas and the total hunting revenue earned in Zambia, respectively. It is ecologically an important animal species essential for the purpose of habitat control and facilitating the provision of suitable grazing pastures. However, the rearing of the African buffalo on game ranches has been hampered by its carrier state of the Southern Africa Terroritory (SAT) serotypes of foot and mouth disease virus (FMD). The African buffalo is also known to be a carrier of Theileria parva lawrencei, the causative agent of corridor disease (CD) that continues to have devastating effects on the livestock industry in Zambia. In addition, the importation of buffaloes from countries with populations endemic to bovine tuberculosis is highly restricted. Veterinary regulations in Zambia, strongly advocate against the translocation of buffaloes from protected areas to private ranches for disease control purposes thereby mounting a considerable constraint on the economic and ecological viability of the industry. It is hoped that this review will motivate the relevant government authorities in exploiting ways in which this animal species play a central role in eco-tourism. 相似文献

Biodegradability of chlorhexidine (CH), triclosan (TC), and benzalkonium chloride (CBA) has been tested in 18 surface water sampling points in the urban area of Buenos Aires. Sampling points were located in both the Reconquista and the Matanza-Riachuelo basins as well as in the La Plata River. High tolerance to the three disinfectants was found and indigenous strains capable of degrading CBA and TC were isolated. Neither tolerance nor biodegradation were correlated with sewage pollution. A strain that degrades CBA was identified as belonging to the genus Pseudomonas using the API20NE system and 16SRNA sequencing. In batch assays, the strain was capable of degrading 100, 200, and up to 500 mg L?1 of CBA in 10, 25, and 46 h respectively with specific growth rates (μ) of 0.56, 0.30, and 0.14 h?1. The efficiency of the process was between 99.5–98.0% in terms of compound removal and between 93.8–89.1% in terms of chemical oxygen demand (COD). The detoxification of the compound as a result of the biodegradation was assessed using Pseudokirchneriella subcapitata, Vibrio fischeri, and Lactuca sativa as test organisms. 相似文献

The soybean cyst nematode (SCN, Heterodera glycines Ichinohe) is a major pest of soybeans (Glycine max L. Merrill) in the central and southern United States. Soybean cyst nematode causes stunted top growth, root pruning and symptoms of mineral element deficiency in soybeans. The objective of this study was to determine the effect of two selections of SCN (I selected on PI209332 and IV selected on PI 89772) on macronutrient uptake, translocation, and tissue concentrations of soybean and to determine the elemental composition of cysts isolated from roots. Soybeans were grown in plastic tubes in the greenhouse where the middle one‐third of the Hodge fine sand (Typic Udipsamment) contained 0, 25,000, or 50,000 SCN eggs. After 35 days, plants were harvested and tissue nutrient element concentrations were determined. Plants infested with both SCN selections were smaller and had much less root volume than controls. Dry weight of each plant tissue decreased as SCN population was increased. Root concentration of potassium (K) and magnesium (Mg) was decreased, whereas root calcium (Ca) and phosphorus (P) concentrations were increased with SCN treatments. Leaf Mg and Ca concentrations increased with SCN treatment. Magnesium uptake per unit root volume was decreased, but Mg translocation (% of total plant content in aerial portion) was increased with SCN treatment. Calcium uptake per unit of root volume was increased, but translocation was unchanged by SCN treatment. The Ca and P concentration of cysts isolated from the soybean roots was high. This high concentration of Ca in cysts is interesting based on the greater root Ca concentration and uptake per unit of root volume in SCN infested plants. Since total uptake and root concentrations of both K and Mg were decreased by SCN treatment, infested soybeans might require very high levels of K and Mg fertilization. Purple corn ( Zea mays L.) has been used for centuries as a natural food colorant in South America and, more recently, in Asia and Europe. However, limited information is available on the factors affecting their anthocyanin concentration and profiles. In this study, 18 purple corn samples grown under different conditions in Peru were evaluated for quantitative and qualitative anthocyanin composition as well as total phenolics. High variability was observed on monomeric anthocyanin and phenolic contents with yields ranging from 290 to 1333 mg/100 g dry weight (DW) and from 950 to 3516 mg/100 g DW, respectively, while 30.5-47.1% of the total phenolics were anthocyanins. The major anthocyanins present were cyanidin-3-glucoside, pelargonidin-3-glucoside, peonidin-3-glucoside, cyanidin-3-maloylglucoside, pelargonidin-3-maloylglucoside, and peonidin-3-maloylglucoside, and 35.6-54.0% of the anthocyanins were acylated. Potassium sources/concentrations on the soil and seedling density did not significantly affect anthocyanin composition. The growing location affected anthocyanin levels and the percentage of anthocyanins to total phenolics ( p < 0.01) and should be taken into account when choosing a material for color production. 相似文献

The genetic structure of 4 Colombian Creole cattle breeds, namely, Coste?o con Cuernos, Blanco Orejinegro (BON), Romosinuano (ROMO), and Sanmartinero (SM), was studied with an analysis of the available pedigree data. The comparison between the effective number of founders (f(e)) and the effective number of ancestors (f(a)) revealed a decrease in the genetic variation that was rather important for the ROMO and San Martinero breeds, which had the lowest f(a)/f(e) ratios (0.34 and 0.53, respectively). All breeds showed similar values for the number of equivalent generations traced, ranging from 3.1 in BON to 4.8 in ROMO. These 2 populations also had the lowest and the highest population sizes, respectively. The lowest average inbreeding coefficient considering the whole pedigree was obtained by BON (0.18%), whereas the highest was attained by ROMO (1.22%). Finally, the percentage of individuals with an inbreeding level greater than 6.25% in the reference population was high, indicating that the existing conservation management strategies could be improved to successfully maintain the genetic variability of these populations. 相似文献

Efficient syntheses of (Z,E)-5,7-dodecadienol, a pheromone component of the Siberian moth, Dendrolimus superans sibiricus, and (E,Z)-10,12-hexadecadienol, a pheromone component of various Lepidoptera pheromones, were accomplished by cis reduction of the corresponding enynols with activated zinc. The most energetic reagent was zinc galvanized with copper and silver (Zn/Cu/Ag) that achieved rapid and high-yield reduction in methanol-water. The stereoselectivity of semi-hydrogenation was > or =98%. A process whereby zinc dust was continuously activated throughout the reduction with an acid was also satisfactory (95-98% cis). Field evaluation of the 1:1 mixture of (Z,E)-5,7-dodecadienol and (Z,E)-5,7-dodecadienal with the Siberian moth in Russia showed that the rubber septa pretreated with compound and stored at -80 degrees C were as effective as freshly treated septa. Moth responses to septa aged in open air indicated that lure effectiveness declined significantly after 2 weeks of aging. Thus, if rubber septa are used as pheromone dispensers in Siberian moth traps monitoring, they should be replaced biweekly with fresh septa for optimal trap effectiveness. 相似文献

Here, we addressed biodegradation vs. volatilization processes, and also bioavailability limitations during biopile remediation of soil initially contaminated by more than 5,000 mg/kg of hydrocarbons. In order to select bioremediation strategies, we first conducted a biotreatability study, which included geochemical, textural, and microbiological characterization of the soil matrix. Next, we implemented five bioremediation approaches onsite in real-scale biopiles. In order to monitor hydrocarbon depletion and to distinguish between biological and non-biological processes, we analyzed chemical biomarkers by means of gas chromatography?Cmass spectrometry. In addition, a comprehensive study of soil grain size and its implications on bioavailability were studied. Furthermore, the evolution of microbial populations was also examined. Two of the strategies implemented in the biopiles (the combination of a slow-release fertilizer and a surfactant, and the use of an oleophilic fertilizer respectively) reduced the soil hydrocarbon content to under 500 mg/kg in 5 months. Additional results from this study indicate that volatilization was the predominant degradation process for light hydrocarbons (below 12 carbon atoms), whereas heavier compounds were mainly biodegraded. However, even in the most favorable situation, a residual concentration of hydrocarbons linked to the finer fraction of the soil was found. 相似文献
