The purpose of this study was to investigate the response of horses to confinement and isolation in a stable (indoor individual housing) for the first time using behavioral indices, heart rate, and salivary cortisol concentration. Six naive 2-year-old Australian Stock Horse fillies were examined at 4-hour intervals over 24 hours in an outdoor group paddock followed by 24 hours in indoor individual housing. Behavioral observations and scores and heart rates were recorded and saliva samples were taken at each interval. During stabling, all horses became agitated and demonstrated increased vocalization and movement. Behavioral scores were significantly higher in the indoor individual housing (P < .001). No significant difference in heart rates between the two environments was detected. Mean salivary cortisol did not increase significantly (2 ng/mL ± 1.4 ng/mL in outdoor group paddock vs 2.5 mL ± 1.2 ng/mL in indoor individual housing). No diurnal rhythm in salivary cortisol was evident in either the outdoor group paddock or indoor individual housing. The results of this study highlight that a combination of behavioral and physiological measures allow better understanding of stress, where one measurement may be misleading. First time stabling of horses elicited marked behavioral responses indicative of stress that were not reflected in increased heart rates or salivary cortisol concentrations. The lack of a diurnal cortisol rhythm and the comparatively high basal cortisol concentrations found in the outdoor group paddock environment may imply that the fillies were already stressed; therefore, stabling did not cause further aberrations detectable by salivary cortisol analysis. 相似文献

Linear models were developed and evaluated for the estimation of daily (24 h) and total lactation milk yield of dairy sheep from single morning (am) or evening (pm) milking records. A total of 22,908 individual test-day milk records of 5110 ewes of the Chios breed, raised in 46 flocks, were used. Depending on the model, different daily milk yield estimates were derived for each ewe, accounting for lactation number, stage of lactation, season of previous lambing and interval between successive milkings. Daily milk yield was also estimated from doubling the single am or pm record. Actual and estimated yields were compared using root mean square errors (RMSEs), mean absolute differences, an accuracy parameter defined as the ratio of the actual yield variance over the sum of the variances of actual yield and absolute difference, and the product moment correlation between estimated and actual yield. Results were validated on independent datasets. Linear models resulted in less biased and more accurate estimates of the daily milk yield than simply doubling the am or pm record. Root mean square errors decreased by 7-37% and the mean absolute difference was reduced by 1-4% of the average daily yield. Higher accuracy and correlation were obtained from linear model application than doubling. Total lactation milk yield was predicted based on actual daily yield and compared to predictions based on estimated daily yield from am or pm records, as well as directly on single milking records. Root mean square errors obtained when daily yield had been estimated with linear models were 26-35% lower compared to doubling the am or pm yield and 0-13% lower compared to estimating the total lactation yield directly from single milking records. Linear model application also resulted in lower mean absolute difference and higher accuracy and correlation than doubling the am or pm record. Recording the yield of a single milking (am or pm) instead of both can benefit milk recording by reducing its cost and increasing farmer participation. A general bio-economic model for beef cattle production was used to define breeding objectives for Charolais cattle to be utilized in a variety of linked production systems. Economic weights were calculated for 16 traits (some with both direct and maternal components) in three production systems (pure-breeding and terminal crossing with beef or dairy cows) and two marketing strategies (sale or fattening of weaned surplus calves). Economic weights for the total breeding objective were calculated as weighted averages, where weights were numbers of cows expected to be mated with Charolais bulls in each production system and marketing strategy. Results suggest that the direct component of calving performance and cow longevity were of primary economic importance in all systems. Conception rate of cows and weaning weight reached about 50% of the standardized economic weight of calving performance in purebred systems with sale of weaned calves, whereas in purebred systems with fattening the economic importance of the direct component of cow conception rate, losses at calving, mature weight of cows, weaning weight, and fattening traits were of equal importance (each approximately about 20% that of calving performance). In terminal crossing systems, weaning weight was important when calves were sold at weaning, and fattening traits were important for systems selling fattened animals. It has been reported that the posterior pituitary (PP) gland contains a potent, unknown prolactin (PRL)-releasing factor (PRF) in rats. PRFs are assumed to be produced in neurones located within the hypothalamus, and to be peptidergic in nature. However, little is known about PRFs in domestic animals. To characterize the PRF in the PP of domestic animals, the present study examined the PRL-releasing activity of an acidic extract from bovine PP (bPP) in vitro and in vivo in cattle. First, the PRL-releasing effect of bPP extract was compared with that of PRL-releasing peptide (PrRP), and thyrotropin-releasing hormone (TRH) from cultured bovine anterior pituitary cells. The extract significantly increased PRL concentrations in the culture medium, at doses of 0.002 and 0.02 eq./ml (one eq. is the PP extract from one animal), compared with the control (p < 0.05). PrRP failed to stimulate the release of PRL. TRH significantly increased PRL concentrations in the culture medium, at doses from 10(-9) to 10(-7) M, compared with the control (p < 0.05). The rate of increase in the PRL concentration, by 0.02 eq./ml bPP extract, was significantly greater than that in TRH (p < 0.05). Secondly, plasma PRL responses to the intravenous (i.v.) injection of bPP extract (0.5 eq./head), PrRP [3.59 mug/kg body weight (BW)], TRH (1 mug/kg BW), and a dopamine receptor antagonist (sulpiride, 0.1 mg/kg BW), were examined in calves. PrRP failed to stimulate PRL release; however, plasma PRL increased immediately following the injection of bPP extract, TRH and sulpiride. The PRL-releasing effect of i.v. injections of TRH and sulpiride was more potent than that of bPP extract. Finally, plasma PRL responses to the intra-hypothalamic injection of bPP extract were examined in calves. The intra-hypothalamic infusion (arcuate nucleus) of 0.0625 eq./head of bPP extract strongly stimulated PRL release in calves (p < 0.05). Five animals in a colony of cynomolgus monkeys (Macaca fascicularis) died or were euthanatized because of alveolar echinococcosis, during a period of 5 years. The remainder of the colony was screened for possible infection with Echinococcus multilocularis, using serology and ultrasonography. A total of 46 animals out of a group of 55 were examined. The presence of anti-Em2 antibodies analyzed with enzyme-linked immunosorbent assay was demonstrated in 3 monkeys. In 2 of these 3 monkeys, multilocular structures compatible with metacestodal cysts in the liver were identified, using ultrasonography. The presence of alveolar echinococcosis was subsequently confirmed at postmortem examination in 1 animal. The other animals are still alive. Two other monkeys were negative in the serological examination but had cystic structures in the liver, which were identified as bile duct cysts at postmortem examination in 1 animal. The other monkey is still alive. These findings suggest that serology for antibodies against the Em2 antigen may represent a useful method in identifying animals that might be infected with E. multilocularis and are therefore at risk of developing fatal alveolar echinococcosis. 相似文献

The in vitro susceptibility to penicillin G, erythromycin and clindamycin was determined by the disc diffusion test and by E‐test for a total of 47 streptococcal strains (three Streptococcus uberis, 36 Streptococcus agalactiae, eight Streptococcus dysgalactiae spp. dysgalactiae) isolated from bovine intramammary infections in Argentina. Moreover, resistance phenotypes of erythromycin‐resistant streptococcal isolates was characterized. MIC90 of penicillin G, erythromycin and clindamycin for S. agalactiae were 0.75, 8.0 and 12.0 μg/ml respectively. Resistance to erythromycin and clindamycin was detected in 13 (27.6%) and 12 (25.5%) isolates respectively. No isolate was resistant to penicillin G. Resistance against macrolides, lincosamides and streptogramin B (MLSB) represented by the constitutive MLSB phenotype was present in 11 (23.4%) erythromycin‐resistant isolates and two isolates (4.3%) expressed the M phenotype. The inducible MLSB phenotype was not identified. Results suggest that beta‐lactams are the first‐line antibiotics when treating streptococcal udder infections; however, the continuous monitoring of the antibiotic resistance is essential, as the emergence of resistant strains has become a growing concern on the therapy of bovine mastitis. 相似文献
