Cryopreservation process reduces lipids and phospholipids from buffalo bull spermatozoa. It was therefore hypothesized that supplementation of fatty acid to extender may improve the post‐thaw quality of buffalo semen. The objective was to evaluate the effect of arachidic acid supplementation in extender on post‐thaw quality of buffalo bull (Bubalus bubalis) spermatozoa. Semen was collected from three adult Nili‐Ravi buffalo bulls of similar age group with artificial vagina (42°C) for 3 weeks (replicate). Qualified semen ejaculates (n = 18) were split into four aliquots and diluted in tris–citric acid extender containing 0.0 (control), 5.0, 10.0 and 20.0 ng/ml at 37°C having approximately 50 × 106 spermatozoa/ml. Diluted semen was cooled to 4°C in 2 h and equilibrated for 4 h at 4°C. Cooled semen was filled in 0.5‐ml straws at 4°C, kept on liquid nitrogen vapours for 10 min and plunged in liquid nitrogen for storage. Thawing of frozen semen was performed after 24 h at 37°C for 30 s. Sperm progressive motility (%) was improved in a dose‐dependent manner by supplementing arachidic acid at 5.0, 10.0 and 20.0 ng/ml compared with control. Structural and functional integrity of sperm plasma membrane (%), number of acrosome‐intact live sperm (%) and sperm chromatin integrity (%) were better (p < 0.05) in extender having 5.0 ng/ml of arachidic acid compared with control. At 10.0 ng/ml, these values did not vary (p > 0.05) from those at 5.0 ng/ml. Further improvement in structural and functional integrity of sperm plasma membrane, number of acrosome‐intact live sperm and chromatin integrity was observed at 20.0 ng/ml of arachidic acid in extender. In conclusion, arachidic acid supplementation in extender improved the post‐thaw quality parameters of cryopreserved Nili‐Ravi buffalo bull spermatozoa. Among the arachidic acid concentrations studied, maximum improvement in post‐thaw semen quality parameters was observed at 20.0 ng/ml. 相似文献
Twenty-four multiparous Holstein cows were utilized in a completely randomized design to examine the effect of feeding field beans (FB, Vicia faba L. var. minor) as substitute for soybean meal (SBM) on lactation performance and metabolic response during the early lactation period. Cows were individually divided into two equal groups and fed for 16 weeks on one each of the two experimental diets. The controls were fed pelleted concentrate contained 150 g/kg dry matter (DM) of SBM as the main protein source, whereas the experimental concentrate contained 345 g/kg DM of FB. Oat hay was offered ad libitum to cows and water was freely available. Blood samples were assayed for their content of: urea, glucose, triglycerides, cholesterol, total protein, albumin, calcium and phosphorus, aspartate aminotransferase (AST) and alanine aminotransferase (ALT), nonesterified fatty acids (NEFA) and β-hydroxybutyrate (BHBA). Inclusion of FB had no detectable effects on DM intake (DMI), body weight (BW), or body condition score (BCS). Neither milk yield nor quality were influenced by dietary treatment, except for milk urea nitrogen (MU) that was reduced in cows fed the FB diet (P < 0.05). Clotting properties of milk were not affected adversely by added dietary FB. Concentration of blood urea (BU) was lower in cows fed the FB diet than in those fed the control SBM diet (P < 0.05). These findings indicate that feeding FB in a lactation diet supported lactation performance similar to cows fed traditional SBM-based diet, and the results may elicit great interest for countries where soybean utilization is adversely influenced by high supply costs. 相似文献
With climate change posing a serious threat to food security, there has been an increased interest in simulating its impact on cropping systems. Crop models are useful tools to evaluate strategies for adaptation to future climate; however, the simulation process may be infeasible when dealing with a large number of G × E × M combinations. We proposed that the number of simulations could significantly be reduced by clustering weather data and detecting major weather patterns. Using 5, 10 and 15 clusters (i.e., years representative of each weather pattern), we simulated phenology, cumulative transpiration, heat-shock-induced yield loss (heat loss) and grain yield of four Australian cultivars across the Australian wheatbelt over a 30-year period under current and future climates. A strong correlation (r2≈1) between the proposed method and the benchmark (i.e., simulation of all 30 years without clustering) for phenology suggested that average duration of crop growth phases could be predicted with substantially fewer simulations as accurately as when simulating all 30 seasons. With mean absolute error of 0.64 days for phenology when only five clusters were identified, this method had a deviation considerably lower than the reported deviations of calibrated crop models. Although the proposed method showed higher deviations for traits highly sensitive to temporal climatic variability such as cumulative transpiration, heat loss and grain yield when five clusters were used, significantly strong correlations were achieved when 10 or 15 clusters were identified. Furthermore, this method was highly accurate in reproducing site-level impact of climate change. Less than 7% of site × general circulation model (GCM) combinations (zero for phenology) showed incorrect predication of the direction (+/−) of climate change impact when only five clusters were identified while the accuracy further increased at the regional level and with more clusters. The proposed method proved promising in predicting selected traits of wheat crops and can reduce number of simulations required to predict crop responses to climate/management scenarios in model-aided ideotyping and climate impact studies. 相似文献
The current study evaluates the effects of early (8th week) and late (16th week of age) weaning of male goat kids on their body growth, testicular growth, sexual behavior, plasma testosterone concentration, and pubertal age. Early (n = 6) and late (n = 7) weaned Beetal bucks were weekly monitored from 18th to 38th week for their body weight, scrotal circumference, testicular volume, testicular echogenicity (via ultrasonography), sexual activities, and plasma testosterone concentration. In comparison to early-weaned, late-weaned bucks showed a marked increase (p < .05) in body weight (11.4 ± 0.8 vs. 13.7 ± 0.6kg), testicular volume (44.1 ± 7.2 vs. 79.8 ± 18.7cm3), scrotal circumference (10.7 ± 0.6 vs. 12.8 ± 0.7cm), and testicular echogenicity (28.3 ± 2.7 vs. 38.3 ± 2.1) from 18th, 28th, 21st, and 24th week onward, respectively. Sexual activities started earlier in late- than early-weaned bucks (22nd vs. 25th week, respectively). Moreover, the sexual behavior index was better (p < .05) after the 34th week in late than early-weaned bucks. The plasma concentration of testosterone (at 39 weeks of age) was relatively more and the onset of puberty was 2–3 weeks earlier (p < .05) in late than early-weaned bucks. In conclusion, age-based early weaning of male kids impairs their testicular growth, sexual behavior, and age at puberty compared to conventional weaning. 相似文献
This study investigated variation in the keratin-associated proteins gene, KRTAP6-3, in 5 Pakistani sheep breeds/crosses using polymerase chain reaction-single strand confirmation polymorphism (PCR-SSCP) analysis. Different banding patterns were revealed, including previously described patterns and a novel pattern (named variant H). The amplified PCR product of the novel banding pattern was directly sequenced, and a synonymous nucleotide variation c.51T>C was revealed. Among the wool traits assessed, a strong correlation (r = 0.929; P < 0.001) was observed between fibre diameter standard deviation (FDSD) and coefficient of variation of fibre diameter (CVFD), between FDSD and medullation (r = 0.720; P < 0.001), between FDSD and medullation standard deviation (MeSD) (r = 0.734; P < 0.001), between MeSD and coefficient of variance of medullation (CVMed), (r = 0.903, P < 0.001), and between CVFD and medullation (r = 0.660), CVFD and MeSD (r = 0.786; P < 0.001), CVFD and CVMed (r = 0.701; P < 0.001) and medullation and MeSD (r = 0.771; P < 0.001). Variant B was found to be associated (P = 0.018) with CVFD; the presence of B being associated with a higher CVFD, than in its absence (41.08 ± 3.98 versus 36.34 ± 3.08). Variant C was associated with CVMed (P = 0.040), where sheep with C had a lower CVMed than sheep where it was absent. Variation in KRTAP6-3 was found to affect fibre diameter related traits of wool.

Soliva sessilis (Burweed) is a winter annual weed found in wheat crop of Pakistan. It is thought to have an allelopathic effect on wheat crop. In this regard, an experiment was conducted in 2019 in the department of Weed Science and Botany at the University of Agriculture Peshawar, to assess the allelopathic potential of S. sessilis on various wheat varieties grown in Peshawar, Pakistan. Physiologically matured roots and leaves of S. sessilis were dried, ground and stored. Three common wheat varieties were selected for the research including Atta Habib, Pirsabaq-15, and Shahkar-13. The treatments of the experiment comprised of the aqueous extracts of the roots and leaves of S. sessilis, hot water extracts, methanol extracts, litter, and mulches. A mass of 5?g of S. sessilis was taken for the different extracts for 24?h, however, the methanol extract was kept for 48?h. The effect of the treatments was highly significant on wheat seed germination, radicle length, plumule length, fresh biomass as well as dry biomasses of the three tested wheat varieties. The variety Atta Habib was discovered as the most susceptible variety to the treatments of aqueous extracts, hot water extracts, and leaves’ litter. In contrary, the variety Pirsabaq-15 exhibited a remarkable increase in the various data collection parameters. The leaves of S. sessilis were observed more toxic as compared to the roots. The mulch of S. sessilis indicated stimulatory effect on the wheat varieties. For the interaction effect between treatments and varieties, the leaves and roots aqueous extract and their litter showed inhibitory effect. In conclusion, the leaves and roots of S. sessilis are potentiality allelopathic to wheat crop plants and further study is needed to assess the types of these allelochemicals present in the leaves and roots of the weed.
Seedling vigor is the basic component of the transplanted rice, which depends on its growing environment and proper age. Thus the influence of nursery management and seedling age was evaluated after transplanting 10, 20, 30, and 40-days older seedlings raised by using seed rate (high and low) and nitrogen (N) (with and without) in rice growing season 2008 and 2009. Study revealed that 10-days older nursery seedlings, irrespective to nursery-bed treatments, showed accelerated growth start after transplanting in main field while at later stages significant interaction was observed. Transplanting of 20-days older fertilized seedlings grown at low seeding density at nursery-bed resulted in similar growth performance as in younger. Maximum net-benefit (45111, 44402 Rs. ha?1) was recorded with transplanting 10-days older nursery seedlings while the fertilized seedlings grown at lower seeding density recorded highest net-benefit (37908, 36780 Rs. ha?1) in both the years. 相似文献
