BACKGROUND

Tebufenozide is widely used to control populations of the smaller tea tortrix, Adoxophyes honmai. However, A. honmai has evolved resistance such that straightforward pesticide application is an untenable long-term approach for population control. Evaluating the fitness cost of resistance is key to devising a management strategy that slows the evolution of resistance.

RESULTS

We used three approaches to assess the life-history cost of tebufenozide resistance with two strains of A. honmai: a tebufenozide-resistant strain recently collected from the field in Japan and a susceptible strain that has been maintained in the laboratory for decades. First, we found that the resistant strain with standing genetic variation did not decline in resistance in the absence of insecticide over four generations. Second, we found that genetic lines that spanned a range of resistance profiles did not show a negative correlation between their LD50, the dosage at which 50 % of individuals died, and life-history traits that are correlates of fitness. Third, we found that the resistant strain did not manifest life-history costs under food limitation. Our crossing experiments indicate that the allele at an ecdysone receptor locus known to confer resistance explained much of the variance in resistance profiles across genetic lines.

CONCLUSION

Our results indicate that the point mutation in the ecdysone receptor, which is widespread in tea plantations in Japan, does not carry a fitness cost in the tested laboratory conditions. The absence of a cost of resistance and the mode of inheritance have implications for which strategies may be effective in future resistance management efforts. © 2023 The Authors. The molecular pathways involved in retrograde signal transduction at synapses and the function of retrograde communication are poorly understood. Here, we demonstrate that postsynaptic calcium 2+ ion (Ca2+) influx through glutamate receptors and subsequent postsynaptic vesicle fusion trigger a robust induction of presynaptic miniature release after high-frequency stimulation at Drosophila neuromuscular junctions. An isoform of the synaptotagmin family, synaptotagmin 4 (Syt 4), serves as a postsynaptic Ca2+ sensor to release retrograde signals that stimulate enhanced presynaptic function through activation of the cyclic adenosine monophosphate (cAMP)-cAMP-dependent protein kinase pathway. Postsynaptic Ca2+ influx also stimulates local synaptic differentiation and growth through Syt 4-mediated retrograde signals in a synapse-specific manner.  相似文献

Forest emissions of biogenic volatile organic compounds (BVOCs), such as isoprene and other terpenes, play a role in the production of tropospheric ozone and aerosols. In a northern Michigan forest, the direct measurement of total OH reactivity, which is the inverse of the OH lifetime, was significantly greater than expected. The difference between measured and expected OH reactivity, called the missing OH reactivity, increased with temperature, as did emission rates for terpenes and other BVOCs. These measurements are consistent with the hypothesis that unknown reactive BVOCs, perhaps terpenes, provide the missing OH reactivity.  相似文献

The purpose of this study was to determine if polymerase chain reaction (PCR) could be used to detect FeLV proviral DNA in bone marrow samples of cats with varying suspicion of latent infection. Blood and bone marrow samples from 50 cats and bone marrow from one fetus were collected, including 16 cats with diseases suspected to be FeLV-associated. Serum enzyme-linked immunosorbent assay (ELISA), blood and bone marrow immunofluorescent antibody test (IFA), and blood and bone marrow PCR were performed on each cat, and IFA and PCR on bone marrow of the fetus. Forty-one cats were FeLV negative. Five cats and one fetus were persistently infected with FeLV. Four cats had discordant test results. No cats were positive on bone marrow PCR only. It appears persistent or latent FeLV infection is not always present in conditions classically associated with FeLV.  相似文献

Equine farms are increasing in number in Northeast and Mid-Atlantic states. These farms may influence environmental quality owing to manure collection, storage, spreading, and disposal practices. A manure management survey was mailed to 700 New Jersey equine farms during the winter of 2009-2010 (these farms were located in two watersheds). Two hundred forty-two surveys were returned; 10% were training or performance farms, whereas the remainder (90%) were breeding, boarding, or pleasure farms. There were differences between the two watersheds in turnout time, pasture acreage, use of drag harrows, and income. Respondents answered questions about diet and environmental management on farms. Seventy-eight percent had bunk management plans. Forty-seven percent disposed of waste feed in a manure pile, 35% left it on fields or paddocks, and 18% put it in a dumpster. Twenty-six percent engage a professional for feeding advice (21% veterinarians, 2% extension, 3% nutrition consultants). Most respondents did not have a regular feed management plan. Sixty-two percent of all farms spread manure on-farm. Of those who spread, 29% have >8.09 hectares (20 acres) for spreading. Ninety-six percent of respondents stored manure >100 ft from water or wetlands. Comparison of cross-tabulations indicates a strong relationship (P < .01) between rotational grazing and the use of a sacrifice or exercise area. Eighty-eight percent of respondents using sacrifice areas also rotationally graze. Results indicate future outreach programs should focus on feed management, manure storage location, rotational grazing, and the use of sacrifice areas as part of a pasture management program.  相似文献

Crop response to deficit irrigation is an important consideration for establishing irrigation management strategies when water supplies are limited. This study evaluated the response of native spearmint to water deficits applied using overhead sprinklers in eastern Washington, US. Five levels of irrigation were applied ranging from full irrigation (100%) to 5% of weekly averaged full crop water needs. Soil water monitoring with soil water balance was used to estimate soil water deficits for irrigation scheduling and soil water use. Mint oil yields, oil components, dry matter production, and the water-use efficiency of the spearmint were assessed. There was significant reduction in fresh mint hay (harvested biomass) yield with increasing water deficit. However, spearmint oil yields remained generally uniform across irrigation treatments at the first cutting but decreased at the driest plots during the second harvest due to a loss of plant stand. The wet harvest index and water-use efficiency improved significantly for both harvests with increasing water deficit. Hay yield, oil yield, wet harvest index, and water-use efficiency are pooled across sides and replicate blocks to provide trends with changes in maximum evapotranspiration. The three major monoterpenes show changes suggesting less mature oil yields. The study demonstrates the feasibility of sustaining native spearmint yields under managed deficit irrigations for deficits not lower than 0.5 ETc.  相似文献

Water temperature strongly affects aquatic ectotherms, as even slight temperature changes can have dramatic effects on physiological rates. Water bodies receiving industrial thermal discharges can undergo dramatic spatial and temporal changes in water temperature. To quantify effects on aquatic ectotherms, thermal habitat quality (bioenergetic growth rate potential; GRP) for zebra mussel, Dreissena polymorpha (Pallas), rusty crayfish, Orconectes rusticus (Girard), walleye, Sander vitreus (Mitchill) and smallmouth bass, Micropterus dolomieu (Lacepède) was estimated near two power plant thermal discharges on the Ohio River, USA, from 2010 to 2012 using bioenergetics models. These results were then compared with GRP under increased base temperatures representing climate warming. Growth rate potential for all species was low near the discharges during summer and highest in winter, with increasing prey consumption minimising the negative effects of increased temperatures. In their immediate vicinity, thermal discharges had a more adverse effect on GRP than plausible climate warming but primarily affected GRP over a small spatial area, particularly within 400 m downstream from the power plants. Examining thermal habitat suitability will become increasingly important as rising energy demand and climate change collectively affect aquatic organisms and their habitats.  相似文献

Chemical treatment of natural fibers is a well-defined means of mechanical property improvement in natural fiberreinforced composites. An understanding of mechanical and thermal properties in these media is essential for evaluating heat transfer, thermal degradation, and overall performance of these composites over their product lifetime. However, very little information is available illustrating the effect of such treatment on the thermal properties of kenaf composites. Also, no study to date has reported the thermal conductivity of individual kenaf fibers. This study reports the effects of fiber treatment (in 6 % NaOH) on thermal transport in unidirectionally oriented kenaf-epoxy composites and individual kenaf fibers. The effective thermal conductivities and thermal diffusivities of chemically treated fiber composites show a general increase over untreated fiber composites (0.210 to 0.232 W/m/K at 28 °C, 0.206 to 0.234 W/m/K at 200 °C). This improvement may be attributed to improved interfacial contact between the fibers and epoxy matrix shown in microstructural images after chemical treatment. The thermal conductivity of individual fibers was evaluated at room temperature using two techniques. Results from both techniques showed slight increases after chemical treatment (0.58±0.53 to 1.0±0.13 W/m/K and 1.2±0.54 to 1.6±0.28 W/m/K) but lacked statistical significance. Any improvement in surface crystallinity after chemical treatment does not appear to affect overall fiber thermal conductivity. A better understanding of thermal transport in kenaf fibers and composites enables better estimation of the performance of these composites in different applications. Moreover, the thermal conductivities of individual fibers are useful in understanding the fiber’s contribution to conduction in different fiber reinforcement configurations.  相似文献
