Measurements of electrical conductivity at high pressure and temperature were taken on the lower mantle phase magnesiowustite with varying Fe3+ content. Although previous measurements at atmospheric pressure suggest Fe2+-Fe3+ hopping (small polaron) as the dominant conductivity mechanism, the present experiments show a change in charge transport mechanism with temperature. The lower temperature measurements are consistent with small polaron conduction, but at higher temperatures, which are more applicable to the lower mantle, a large polaron mechanism is suggested. Because these mechanisms have different temperature and compositional dependencies, this transition has important implications for extrapolation to mantle conditions.  相似文献

Bovine parainfluenza-3 virus (PI-3V) replicated in cultured bovine alveolar macrophages (BAM) in vitro. Cytopathic changes included spindle cell and giant cell formation, diffuse cell lysis, and cellular detachment progressing to total destruction of the monolayer. Direct immunofluorescent microscopy revealed viral antigen in greater than 90% of BAM on post-inoculation day (PID) 3. Virus titers in culture supernatants increased by a factor of 1.6 X 10(6) TCID50 by PID 6. Candida glabrata was phagocytized by a similar proportion of glass-adherent PI-3V-infected and sham-inoculated control BAM. However, when the number of glass-adherent cells available for assay was taken into account (normalization), the percentage of phagocytic PI-3V-infected BAM was significantly lower than that of the control cells at PID 5 and 7 (P less than 0.01). Phagosome-lysosome fusion assays had a marked reduction of fusion activity in PI-3V-infected BAM as compared with that of control BAM. Proportions of infected cells that showed fusion were approximately 50% of that of the control cells at PID 4 and 6 (P less than 0.05). Normalization of these values reduced the fusion rate of PI-3V-infected BAM to 34% of that of control BAM (P less than 0.01).  相似文献

Avian respiratory macrophages (ARM) were obtained from lungs and air sacs of 122 White Plymouth Rock chickens, ranging from 376 to 3800 g in weight. Procedures involved lavaging through the surgically prepared trachea with either a 15-g cannula or French #8 pediatric urinary catheters. Factors, in different combinations, investigated for their effects on the ARM yield, were: lavage fluids (0.85% physiologic saline, 0.1 M phosphate-buffered saline, Ca-Mg-free Hanks' solution, Eagle's minimum essential medium); additives (10 U heparin/ml, 0.1% EDTA, 12 mM lidocaine); lavage repetitions (from 3 to 10); fluid temperature (room and 41 C); and lavage time (fluid retention up to 35 min). None of the lavage methods emerged clearly as the best, with phosphate-buffered saline and 0.85% physiologic saline alone as good as when combined with additives. Although 10 lavages yielded more ARM, it appeared that the majority of ARM washed off into the early lavages. Chickens from a line selected for large body size had more ARM than those from a line selected for small body weight. Regardless of genetic line, however, the chickens yielded a very low number of ARM compared with mammalian species of the same or smaller weight. Most of the birds yielded only 200,000 to 300,000 ARM, with minimum yields being less than 10,000, the maximum being 2 million ARM. Either these results point to a deficiency in the defense system of the chicken's respiratory tract against bacteria, mycoplasma, fungi, and viruses, or mechanisms other than macrophages are primary in resistance to pathogens.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Informal economy of gold mining has contaminated some important river basins in Amazon. Follow-up studies on critical compartments showed some areas with high Hg levels in fish as well as in human hair samples. Average Hg in piscivorous fish in the Madeira river itself was 846 ppb (N=284) with a maximum of 3921 ppb. Mercury in fish from non polluted areas in this basin shows high variability, even for single species. A seasonal variation in Hg content was observed, with higher values at the end of the dry season. In the upper Tapajós basin comparable values were found for fish but with a definite decreasing trend downstream. Average value for piscivorous fish in the whole Tapajós basin is 482 ppb (N=122) with a maximum value of 3770 ppb. Hair Hg was higher in fishing villages in the Tapajós (average: 17 ppm; with N=432 and maximum value of 176 ppm) than in the Madeira (average: 9 ppm; N=169; maximum 71 ppm), and data from some areas of the Tapajós suggest a decrease with time. Mercury was much higher in urine of goldshop workers in Santarém (low Tapajós) than in Alta Floresta (high Tapajbs) and show a decreasing trend in both cases, probably related to the significant decline in gold mining activities during the study period (1986–1994).  相似文献

The aim of this study was to characterize the vocalizations produced during electroejaculation under general anaesthesia in pampas deer males and to determine whether the characteristics of those vocalizations differ in adult and young pampas deer males. Electroejaculation was applied to 13 adults (AM) and 13 young (YM) males under general anaesthesia. Vocalizations were digitally recorded, and the number and duration of vocalizations, the latency in relation to each voltage, the total time vocalizing, and the structure of the fundamental frequency (F0) [initial frequency (Fstart), maximal frequency (Fmax), minimal frequency (Fmin) and final frequency (Fend)] were analysed. No male vocalized with 0 V; the number of animals that vocalized increased at 2 and 3 V and increased again at 4, 5 and 6 V (p < 0.05). The latency time from the beginning of each series (each voltage) decreased until 4 V (p < 0.01). The number of vocalizations/voltage increased from 4 V (p < 0.05). The length of each vocalization and the total time during which animals vocalized were greater in YM than AM (p = 0.02 and p = 0.01, respectively). Similarly, the fundamental frequencies were higher in YM than AM (p ≤ 0.05). Overall, we concluded that the vocalizations emitted during electroejaculation in pampas deer under general anaesthesia are related to the voltage applied during the process. Young males vocalize more time, probably due to a greater sensibility to the electric stimulation. The differences in the characteristics of the vocalizations between adult and young males may be related to the anatomic differences in the neck of adult or young males.  相似文献

A common single-nucleotide polymorphism in the brain-derived neurotrophic factor (BDNF) gene, a methionine (Met) substitution for valine (Val) at codon 66 (Val66Met), is associated with alterations in brain anatomy and memory, but its relevance to clinical disorders is unclear. We generated a variant BDNF mouse (BDNF(Met/Met)) that reproduces the phenotypic hallmarks in humans with the variant allele. BDNF(Met) was expressed in brain at normal levels, but its secretion from neurons was defective. When placed in stressful settings, BDNF(Met/Met) mice exhibited increased anxiety-related behaviors that were not normalized by the antidepressant, fluoxetine. A variant BDNF may thus play a key role in genetic predispositions to anxiety and depressive disorders.  相似文献

At least 16 fragments were detected in images of comet C/1999 S4 (LINEAR) taken on 5 August 2000 with the Hubble Space Telescope (HST) and on 6 August with the Very Large Telescope (VLT). Photometric analysis of the fragments indicates that the largest ones have effective spherical diameters of about 100 meters, which implies that the total mass in the observed fragments was about 2 x 10(9) kilograms. The comet's dust tail, which was the most prominent optical feature in August, was produced during a major fragmentation event, whose activity peaked on UT 22.8 +/- 0.2 July 2000. The mass of small particles (diameters less than about 230 micrometers) in the tail was about 4 x 10(8) kilograms, which is comparable to the mass contained in a large fragment and to the total mass lost from water sublimation after 21 July 2000 (about 3 x 10(8) kilograms). HST spectroscopic observations during 5 and 6 July 2000 demonstrate that the nucleus contained little carbon monoxide ice (ratio of carbon monoxide to water is less than or equal to 0.4%), which suggests that this volatile species did not play a role in the fragmentation of C/1999 S4 (LINEAR).  相似文献

During the first few days after weaning, pigs often experience BW loss as they adapt to eating solid food. During this time period, they are also known to drink excessively and develop abnormal oral behavior such as belly nosing. The excessive drinking may stem from the piglets' attempt to satiate hunger through gut fill from a familiar ingestive source. Gut fill through water intake may affect the establishment of feeding behavior. Using drinker devices other than the standard nipple drinker may ease the piglets' transition at weaning by facilitating the initiation of feeding and preventing the development of behavioral problems such as excessive drinking and belly nosing. In this experiment, we examined the effect of drinker type on water and food intake, growth rates, and belly nosing in newly weaned piglets. Eighteen pens of 15 piglets each (270 piglets total) were weaned at 18.1 +/- 0.1 d of age and housed in pens containing 1 of 3 drinker devices (standard nipple, push-lever bowl, and float bowl). Piglets' water and feed intake, water use, BW, and behavior were examined on a pen basis through 2 wk after weaning. Piglets with nipple drinkers wasted more water than the other piglets (P < 0.001; float, 295 +/- 70 mL x pig(-1) . d(-1); nipple, 1,114 +/- 63 mL x pig(-1) . d(-1); and push-lever, 186 +/- 63 mL x pig(-1) . d(-1)), whereas piglets with float bowls consumed less water than the other piglets (P < 0.001; float, 475 +/- 81 mL . pig(-1) x d(-1); nipple, 870 +/- 76 mL x pig(-1) . d(-1); push-lever, 774 +/- 76 mL x pig(-1) . d(-1)). Drinker type affected feeding behavior (P = 0.02); piglets with push-lever bowls spent less time at the feeder than the other piglets, although no difference was detected for feed intake (P = 0.64) or overall ADG (P = 0.16). Piglets with push-lever bowls also tended to perform less piglet-directed nosing behavior than piglets with the float bowl (P = 0.04). Piglets appear to use more water during the first 2 d after weaning with certain drinker devices. However, piglets do not appear to attain satiety through water consumption because most of the water used during the first few days after weaning is wasted. This excessive drinking and water wastage can be abated through the use of push-lever drinkers without negative implications for feed intake or growth rates.  相似文献

Colletotrichum coccodes is the causal agent of the potato blemish disease black dot. Two PCR primer sets were designed to sequences of the ribosomal internal transcribed spacer (ITS1 and ITS2) regions for use in a nested PCR. The genus-specific outer primers (Cc1F1/Cc2R1) were designed to regions common to Colletotrichum spp., and the species-specific nested primers (Cc1NF1/Cc2NR1) were designed to sequences unique to C . coccodes . The primer sets amplified single products of 447 bp (Cc1F1/Cc2R1) and 349 bp (Cc1NF1/Cc2NR1) with DNA extracted from 33 European and North American isolates of C. coccodes. The specificity of primers Cc1NF1/Cc2NR1 was confirmed by the absence of amplified product with DNA of other species representing the six phylogenetic groups of the genus Colletotrichum and 46 other eukaryotic and prokaryotic plant pathogenic species. A rapid procedure for the direct extraction of DNA from soil and potato tubers was used to verify the PCR assay for detecting C. coccodes in environmental samples. The limit of sensitivity of PCR for the specific detection of C. coccodes when inoculum was added to soils was 3·0 spores per g, or the equivalent of 0·06 microsclerotia per g soil, the lowest level of inoculum tested. Colletotrichum coccodes was also detected by PCR in naturally infested soil and from both potato peel and peel extract from infected and apparently healthy tubers. Specific primers and a TaqMan fluorogenic probe were designed to perform quantitative real-time (TaqMan) PCR to obtain the same levels of sensitivity for detection of C. coccodes in soil and tubers during a first-round PCR as with conventional nested PCR and gel electrophoresis. This rapid and quantitative PCR diagnostic assay allows an accurate estimation of tuber and soil contamination by C. coccodes .  相似文献
