Kiwifruit (Actinidia deliciosa and Actinidia chinensis) is allergenic to sensitive patients, and, under Japanese regulations, it is one of the food items that are recommended to be declared on food labeling as much as possible. To develop PCR-based methods for the detection of trace amounts of kiwifruit in foods, two primer pairs targeting the ITS-1 region of the Actinidia spp. were designed using PCR simulation software. On the basis of the known distribution of a major kiwifruit allergen (actinidin) within the Actinidia spp., as well as of reports on clinical and immunological cross-reactivities, one of the primer pairs was designed to detect all Actinidia spp. and the other to detect commercially grown Actinidia spp. (i.e., kiwifruit, Actinidia arguta, and their interspecific hybrids) except for Actinidia polygama. The specificity of the developed methods using the designed primer pairs was verified by performing PCR experiments on 8 Actinidia spp. and 26 other plants including fruits. The methods were considered to be specific enough to yield target-size products only from the target Actinidia spp. and to detect no target-size products from nontarget species. The methods were sensitive enough to detect 5-50 fg of Actinidia spp. DNA spiked in 50 ng of salmon testis DNA used as a carrier (1-10 ppm of kiwifruit DNA) and 1700 ppm (w/w) of fresh kiwifruit puree spiked in a commercial plain yogurt (corresponding to ca. 10 ppm of kiwifruit protein). A quantitative method for four theaflavins and two methylated theaflavin derivatives in black tea leaves was developed by solid-phase extraction and a high-performance liquid chromatographic method with photodiode array detection. The theaflavins in black tea leaves were extracted three times with 40 vol 50% aqueous ethanol (mg dry tea powder/mL) containing 2% ascorbic acid. The ethanol extracts were diluted 4-fold with distilled water. All diluted extracts were directly applied to the solid-phase C18 cartridge column without concentration. The fraction of theaflavins was obtained by 40% ethanol extraction after rinsing with water followed with 15% ethanol extraction. An aliquot of theaflavins after concentration was injected onto an ODS C18 reversed-phase column, and four theaflavins and two methylated theaflavins were sufficiently separated by a linear gradient system using distilled water and acetonitrile with 0.5% acetic acid. This analytical method is sensitive for the determination of a small amount of methylated theaflavins, since various interfering substances produced during the fermentation process were eliminated in advance by solid-phase extraction. Using this analytical method, we also demonstrated that methylated theaflavins were easily produced during the manufacture of black tea. 相似文献
In a 6-week feeding experiment, chum salmon, Oncorhynchus keta, swim-up fry, were fed fish meal diets supplemented with silkworm pupae powder (5%), dried beef liver (5%), krill meal (5%) or earthworm powder (5%) at the expense of fish meal, or substituting glucose (13%) for dextrin. Fish fed the diet with earthworm powder showed the best growth performance (675% weight gain in 6 weeks) and feed efficiency (117%). The growth rate and feed efficiency of the krill meal group were also significantly better than those of the control group (without supplement or substitution). Substitution of glucose and supplementation of silkworm pupae or beef liver failed to improve the growth rate, but significantly improved feed efficiency. Food consumption of fish fed the diet supplemented with earthworms was lowest, and none of the dietary treatments appreciably increased food intake of the fish over that of the control group. The dietary group receiving earthworm powder also showed significantly higher body fat content than the remaining dietary groups. No significant differences were noted in body protein and ash contents among all dietary treatments. 相似文献
To assess the stress level of cheetahs reared in Japan and to identify the prime components of the climatic conditions that affect their thermal stress, fecal corticosterone was monitored for 8 months from May to the following January. A total of 203 fecal samples were gathered in the morning from seven adult cheetahs that were kept at a zoological garden in Wakayama, Japan. Cheetahs were on exhibit singly or together with a harmonious conspecific during the day, but housed singly at night. Although the monthly fluctuation in corticosterone concentrations was not significant, the concentrations were relatively low during the summer season. Individual differences among cheetahs and the interaction effect between individual and month on the corticosterone concentrations were significant. Whereas the corticosterone concentrations negatively correlated with air temperature, they were positively correlated with the amount of rainfall. The highest air temperature and the amount of rainfall were extracted as the prime factors affecting corticosterone concentrations. These results suggest that cheetahs reared in Japan are somewhat subjected to thermal stress, particularly on cooler and/or rainy days. 相似文献
ABSTRACT: Stocking of exogenous, hatchery-reared white-spotted charr Salvelinus leucomaenis has been conducted throughout much of their range in Honshu Island, Japan, to increase angling opportunities. Although the native charr populations are thought to have declined because of hybridization with introduced fish, their distribution and genetic status have been uncertain. Fine population structures of charr in the upper Tone River drainage were examined using mitochondrial DNA and microsatellite analyses so as to clarify the presence of native populations. One common mtDNA haplotype was detected in all populations in the Ohashi River and Watarase River, and four and one tributary populations were monomorphic for such haplotypes, respectively. However, several haplotypes, considered to have originated from stocked hatchery fish, were observed in the stocked and the remaining populations. Judging from the genetic integrity over a fine geographic scale, the former were considered as indicative of native populations and the latter as admixtures with hatchery fish. Comparisons of genetic diversity, deviations from the Hardy–Weinberg equilibrium, principal component analysis, and relatedness estimations based on microsatellite DNA can also provide evidence for distinguishing native populations from those influenced by hatchery fish. 相似文献

In the North Nile Delta of Egypt, the impacts of overplanting paddy rice on water delivery performance have not been discussed quantitatively. Further, the amount of water that could have been saved if farmers would follow the planned area is unknown. In this study, water delivery performance was assessed by comparison of actual paddy rice planting and the government’s planned conditions. For both conditions, performance indicators relating to adequacy, equity, and dependability were analyzed across six locations in conjunction with the branch canal water level in 2013 and 2014. Based on the difference between the actual water supply and planned water demand, the amount of water that could have been saved for downstream uses was calculated. The average adequacy for the investigation period was good at one location, fair at 2 locations, and poor at 3 locations in both years. Further, adequacy under both actual and planned conditions was poor in late July at all locations. The planned adequacy and dependability downstream and equity among locations improved compared to the actual condition in both years. Under the condition that paddy rice area is the upper limit planned by the government, about 12.3% and 9.6% of water could be potentially saved in each year. The difference between actual and planned water delivery performance is caused by the branch canal’s low water level. Control of overplanting paddy rice and coordination of water distribution among water user associations would improve stable water level in the canal and, eventually, water delivery performance.
ABSTRACT: The effect of dietary taurine on juvenile Japanese flounder was determined by feeding three taurine-supplemented experimental diets (TAU) and a commercial diet (CD) to evaluate a practical diet for juvenile Japanese flounder. Juvenile Japanese flounder were reared on the three experimental diets supplemented with taurine at 0, 0.5, 1.0% and CD. These diets were fed to juvenile Japanese flounder of an initial mean body weight of 0.2 g for 6 weeks at 20°C and the taurine contents of the whole body and tissues were analyzed. The final average body weight of juvenile Japanese flounder fed the 1.0% TAU was significantly higher than that of the other groups. Taurine contents in the whole body and tissues increased with the increase in dietary taurine level. These results indicate that juvenile Japanese flounder require at least 15 mg/g taurine in the diet, even though a combined mix of fish, krill and squid meal was the main protein source in the experimental diets. 相似文献
