Changes in the composition of fructooligosaccarides (FOSs) and related enzyme activities of burdock root during low-temperature storage (1 °C) were measured. Burdock root is composed of monosaccharides, such as glucose and fructose, disaccharides, such as sucrose, and FOS in the extracted carbohydrates. The composition was the highest in fructose, followed by sucrose, FOS, and glucose. The FOS composition was the highest in 1-kestose, followed by nystose and 1-fructofuranosyl nystose. The fructose and glucose contents in burdock root during storage at 1 °C decreased significantly, whereas the sucrose content increased sharply. In contrast, the 1-kestose, nystose, and 1-fructofuranosyl nystose contents significantly increased. The activities of sucrose:sucrose 1-fructosyl transferase (1-SST) and fructan:fructan 1-fructosyl transferase (1-FFT) increased sharply and then gradually decreased, and they were closely related to the changes in FOS content. However, the activity of fructan 1-exohydrolase (1-FEH) was constant. These results indicate that changes in the FOS composition and related enzyme activities of burdock root might be related to low temperature during long term storage.  相似文献

Pine wilt disease, caused by the pine wood nematode (PWN), Bursaphelenchus xylophilus, has become of worldwide quarantine concern in recent years. Here, we disclosed the surface coat (SC) proteins of the PWN which are thought to be one of the key components in pine wilt development. This is the first report that focused on the SC proteins and thoroughly identified those proteins of a plant-parasitic nematode using the proteomic approach. In this study, SC protein profiles were compared for PWNs grown on the fungus Botrytis cinerea and in host pine seedlings. The results demonstrated that the gross amount of PWN SC proteins drastically increased during infection of the host pine. Thirty-seven protein bands showed significant quantity differences between fungus-grown and host-origin PWNs, and were used for identification by matrix-assisted laser desorption ionization time of flight mass spectrometry analysis. These included several proteins that are presumed to be involved in the host immune response; for example, regulators of reactive oxygen species (ROS) and a ROS scavenger. These results might suggest that the PWN SC proteins are crucial in modulating or evading host immune response. Our data provide a new insight into the mechanism of pine wilt disease and the biological role of the SC proteins of plant-parasitic nematodes.  相似文献

In this study, effects of grazing‐experienced heifer presence on foraging behavior development at the feeding station (FS) scale for first‐grazing season calves were determined. A group of four calves grazing alone (C), and another comprising four calves (Wc) and three grazing‐experienced heifers (Wh), were alternately stocked every day for 2 h on the same pasture for 26 days. The foraging time budget for Wc was significantly longer than that for C (P < 0.05) on day 7, and was similar to that for Wh on all days. Bite rate per FS was significantly higher for Wc (15.5 bite/min) than for C (13.2 bite/min) from day 1 to 14 (33.4 vs. 29.0 bite/min, respectively; P < 0.01) and significantly lower than that for Wh on all days. The number of steps taken between adjacent FSs by Wc (4.7) was significantly lower than that for C (7.2) on days 1 and 14 (2.1 vs. 2.9 steps, respectively; P < 0.01) and was similar to Wh on all days. This suggests that grazing‐experienced heifers have positive effects on the foraging behavior development of new‐season grazing calves through 14 days after the start of stocking.  相似文献

We examined the water status of Hinoki cypress, Chamaecyparis obtusa (Siebold & Zucc.) Endl., trees after a severe typhoon to determine possible causes of the decline and dieback that can occur in what appear, at first, to be healthy trees in typhoon-damaged forest stands. We found that in apparently healthy trees in a storm-damaged stand, the water conducting area of the trunk cross section was greatly reduced compared with that of similarly sized trees in a nearby undamaged stand. Although leaf specific hydraulic resistance (Wl) from soil to leaf and from trunk to leaf was higher in trees from the storm-damaged than the undamaged stand, Wl values from soil to root were similar. Diurnal patterns in the rates of change in trunk diameter differed between trees in the damaged and the undamaged stand. We conclude that increased aboveground hydraulic resistance caused by a reduction in trunk water conducting area could be a major reason for the decline and dieback of apparently healthy trees in typhoon-damaged stands.  相似文献

Chromosome and morphological variations of embryogenic calli-derived plants of gynogenic haploid, diploid, triploid and tetraploid asparagus (Asparagus officinalis L.) were investigated. Artificial tetraploids were produced using colchicine treatment of seeds of diploid cultivar, ‘Poultom’. ‘Haidel’ (2X) was crossed with the artificial tetraploids, from which one gynogenic haploid, one diploid, 6 triploid, 3 mixoploids were obtained. Embryogenic calli were first obtained from crown buds, subsequently induced to form somatic embryos, and after 30 days, induced to germinate. Chromosome variation in embryogenic calli-derived plants increased with increasing duration of subculture, particularly when low ploidy levels of plants such as haploid and diploid were used as explants. Approximately 80% of haploid-derived plants showed morphological variations such as dwarfness and abnormal morphological characteristics, although no differences were observed in cladodes and stem characteristics between other polyploid-derived plants and their parents. The data presented here would supply important fundamental information for commercial mass-propagation using somatic embryogenesis. Nutritional status and feeding habits of immature female bighand thornyhead Sebastolobus macrochir were assessed based on Fulton’s condition factor K, hepatosomatic index (HSI), and stomach observations. Fish were obtained from bottom trawl samples taken seasonally during 2006/2007 in the Tohoku area, off the Pacific coast of northern Honshu, Japan. Results of the generalized linear model (GLM) indicated that fish condition deteriorated with depth, and small fish had a lower K, HSI, and percent stomach content index (SCI) than larger specimens. The empty stomach rate was higher in deeper waters in northern Tohoku (except for small fish from spring to autumn) and in southern Tohoku (spring/summer). Changes in the index of relative importance (%IRI) showed that the main prey items changed from nutritionally high to low prey items with depth. This suggests that deeper waters, where fish with the lowest condition values occur, are a poorer quality habitat for immature bighand thornyhead and that small fish are subordinate to larger fish. The effect on growth, due to variations in nutritional status and feeding habits, produced by the distributional changes in bighand thornyhead is also discussed.  相似文献

Two anaesthetics, clove oil and methane sulphonate (MS-222), were examined for their effects on the olfactory nerve response of masu salmon ( Oncorhynchus masou Brevoort) and rainbow trout ( O. mykiss Walbaum). Exposing both species to clove oil for 3 min at concentrations of 50 and 100 mg L−1, or for 10 min at 50 mg L−1, did not significantly reduce their olfactory response. Directly applying clove oil anaesthesia to the olfactory epithelium significantly reduced olfactory response though after 20 min, olfactory response recovered to 70% and 52% of pre-treatment levels in masu salmon and rainbow trout respectively. Compared with the post-anaesthetic recovery of responses after clove oil (50 mg L−1), buffered MS-222 (100 mg L−1) with NaHCO3 (100 mg L−1), and unbuffered MS-222 (100 mg L−1) treatment for 3 min, the response after MS-222 treatment declined gradually and significantly, but not after clove oil and MS-222+NaHCO3 treatments. Clove oil appears to be an effective and relatively safe anaesthetic for salmonids with little long-term impact on their olfactory response, which plays a crucial role in their life history.  相似文献

Seven kinds of angiotensin I-converting enzyme (ACE) inhibitory peptides were isolated from the hydrolysates of wakame (Undaria pinnatifida) by Protease S "Amano" (from Bacillus stearothermophilus) by using three-step high-performance liquid chromatography (HPLC) on a reverse-phase column. These peptides were identified by amino acid composition analysis, sequence analysis, and liquid chromatography-mass spectrometry (LC-MS), as Val-Tyr (IC(50) = 35.2 microM), Ile-Tyr (6.1 microM), Ala-Trp (18.8 microM), Phe-Tyr (42.3 microM), Val-Trp (3.3 microM), Ile-Trp (1.5 microM), and Leu-Trp (23.6 microM). These peptides have resistance against gastrointestinal proteases in vitro. Each peptide was determined to have an antihypertensive effect after a single oral administration in spontaneously hypertensive rats (SHR). Among them, the blood pressure significantly decreased by Val-Tyr, Ile-Tyr, Phe-Tyr, and Ile-Trp in a dose of 1 mg/kg of body weight (BW). The present study showed that antihypertensive effect in the hydrolysates of wakame by Protease S "Amano" was attributed to these peptides.  相似文献

Levels of two moleculer types of gonadotropin-releasing hormone (GnRH), salmon GnRH (sGnRH) and chicken GnRH–II (cGnRH–II) in the various brain regions and pituitary gland of sockeye salmon (Oncorhynchus nerka) and chum salmon (O. keta) during smoltification and spawning migration, respectively, were measured using specific time-resolved fluoroimmunoassay (TR-FIA) systems. Changes in sGnRH levels in different brain regions tended to be specifically synchronized with serum thyroid hormone or pituitary gonadotropin (GTH) levels during smoltification and spawning migration, respectively. In contrast, cGnRH–II levels did not show such synchronized changes. SGnRH and cGnRH–II in various brain regions might have different roles during smoltification and spawning migration of salmonid fishes.  相似文献
