Ethanol extracts from 15 kinds of marine algae collected from the coast of the Noto Peninsula in Japan were examined for their inhibitory effects on human salivary α-amylase. Four extracts significantly suppressed the enzyme activity. An inhibitor was purified from the extract of Sargassum patens . The compound was a new phloroglucinol derivative, 2-(4-(3,5-dihydroxyphenoxy)-3,5-dihydroxyphenoxy) benzene-1,3,5-triol (DDBT), which strongly suppressed the hydrolysis of amylopectin by human salivary and pancreatic α-amylases. The 50% inhibitory activity (IC(50)) for α-amylase inhibition of DDBT (3.2 μg/mL) was much lower than that of commercially available α-amylase inhibitors, acarbose (26.3 μg/mL), quercetagetin (764 μg/mL), and α-amylase inhibitor from Triticum aestivum (88.3 μg/mL). A kinetic study indicated that DDBT was a competitive α-amylase inhibitor with a K(i) of 1.8 μg/mL. DDBT also inhibited rat intestinal α-glucosidase with an IC(50) value of 25.4 μg/mL for sucrase activity and 114 μg/mL for maltase activity. These results suggest that DDBT, a potent inhibitor of carbohydrate-hydrolyzing enzymes, may be useful as a natural nutraceutical to prevent diabetes. 相似文献

Abstract A method which uses the pressure head to predict unsaturated hydraulic conductivity is presented to calculate the soil water flux in a field. Hydraulic conductivities in the primary drying and wetting processes were measured with core samples in the laboratory and the hysteresis between the hydraulic conductivity and pressure head was taken into account. Hydraulic gradients were measured every hour with tensiometers installed in the field. This method was applied to analyze the water movement at 94-cm depth in Hydric Hapludands. Downward or upward flow of water by summing soil water fluxes was examined using the water balance method. Amounts of downward flow determined by our method after heavy rain in a wet soil were slightly larger in the soybean plot but smaller in the bare plot than those obtained by the water balance method due to non-uniform infiltration. Water balance equation which used values of upward flow across a 94-cm depth estimated reasonably well the evapotranspiration from the soybean plot and the evaporation from the bare plot during a dry period. Effect of initial soil water conditions on downward flux by rainfall was well monitored by our method. 相似文献

Studies of nitrogen (N) use by plants have confirmed some winter N uptake; however, the mode of regulation of plant N use in winter is unknown. The regulation of N use by plants during winter may differ from that in the growing season, as plant growth strongly affects N use. We investigated the effects of winter buds on winter N use by Japanese red pine (Pinus densiflora), as a previous study demonstrated that N absorbed during winter contributes significantly to leaf growth in the following spring. We conducted a bud pruning experiment during winter to examine the effects of winter buds on winter N uptake and allocation among plant organs using 15N labeling. Over a three-week labeling period, the 15N content in roots increased to 0.20 ± 0.12 mg N g DW?1, which is equivalent to 1.8 ± 1.1 % of the total N content in the roots. However, this absorbed 15N rarely appeared in needles and buds. Bud pruning did not affect 15N uptake and allocation. On the other hand, significant total N retranslocation was found within the crowns of saplings without bud pruning, but N was not retranslocated in bud-pruned plants. The bud pruning experiment indicated that N was retranslocated from needles into winter buds. Since soil N availability changes dramatically and is unstable in many forest ecosystems, N contained in needles would be a more stable source of N than newly absorbed N. 相似文献

We investigated whether regular changes of the sire in a breeding farm of Hokkaido Native Horses (HKDs) enables the DNA‐level genetic variation of the produced animals to be maintained. The genotypes of 31 microsatellite markers were identified and analyzed in 207 animals produced in a breeding farm in which the sire was replaced every 3 to 5 years. The mean allele number indicating the degree of genetic variation was 5.97 and was similar to those reported previously. The mean observed heterozygosity was 0.74 and was higher than the expected heterozygosity, 0.69; FIS was ?0.07, indicating that the analyzed animals reflected frequent outbreeding and had maintained genetic variation. Based on genetic structural analysis, the number of genetic subpopulations of the animals was estimated to be as 6, and the majority (more than 50%) of each subpopulation corresponded to the progeny of one of the sires used in the breeding farm; these observations suggested that genetic variation in the analyzed animals reflected the genetic differences among sires. Pedigree records indicated that the average co‐ancestry coefficient between sires used in the breeding farm was 0.015 corresponding to second cousin. This level of kinship among sires is acceptable for producing HKDs that maintain genetic variation. 相似文献

F0F1, found in mitochondria or bacterial membranes, synthesizes adenosine 5'-triphosphate (ATP) coupling with an electrochemical proton gradient and also reversibly hydrolyzes ATP to form the gradient. An actin filament connected to a c subunit oligomer of F0 was able to rotate by using the energy of ATP hydrolysis. The rotary torque produced by the c subunit oligomer reached about 40 piconewton-nanometers, which is similar to that generated by the gamma subunit in the F1 motor. These results suggest that the gamma and c subunits rotate together during ATP hydrolysis and synthesis. Thus, coupled rotation may be essential for energy coupling between proton transport through F0 and ATP hydrolysis or synthesis in F1. 相似文献

We report the fabrication of transparent field-effect transistors using a single-crystalline thin-film transparent oxide semiconductor, InGaO3(ZnO)5, as an electron channel and amorphous hafnium oxide as a gate insulator. The device exhibits an on-to-off current ratio of approximately 106 and a field-effect mobility of approximately 80 square centimeters per volt per second at room temperature, with operation insensitive to visible light irradiation. The result provides a step toward the realization of transparent electronics for next-generation optoelectronics. 相似文献

The estimation of the reward an action will yield is critical in decision-making. To elucidate the role of the basal ganglia in this process, we recorded striatal neurons of monkeys who chose between left and right handle turns, based on the estimated reward probabilities of the actions. During a delay period before the choices, the activity of more than one-third of striatal projection neurons was selective to the values of one of the two actions. Fewer neurons were tuned to relative values or action choice. These results suggest representation of action values in the striatum, which can guide action selection in the basal ganglia circuit. 相似文献

The Antarctic ice sheet at Byrd Station has been core-drilled to bedrock; the vertical thickness of the ice is 2164 meters. Liquid water-indicative of pressure melting-was encountered at the bed. Heat flow through the base of the ice sheet is estimated at 1.8 microcalories per square centimeter per second. The minimum temperature was -28.8 degrees C at 800 meters; maximum ice density, 0.9206 at 1000 meters. Core studies reveal the existence of a chemically pure, structurally stratified sheet comprising bubbly ice to 900 meters that transforms to bubble-free deformed ice, with substantially vertically orientated c-axis structure, below 1200 meters. Below 1800 meters the deformed ice structure gives way to large annealed crystals. Several thin layers of dirt between 1300 and 1700 meters are tentatively identified as volcanic ash, and horizontally banded debris, including fragments of granite, is present in the basal ice. 相似文献
