Public perceptions of wildlife vary by species, context, and culture. Understanding these perceptions helps managers understand how their decisions may generate conflict among stakeholders. We mailed questionnaires to residents in the northern region of Hyogo prefecture in Japan to examine their willingness to tolerate wildlife species of greatest concern to government officials, including: (a) native species frequently causing agricultural damage (wild boar [Sus scrofa], sika deer [Cervus nippon], Japanese macaque [Macaca fuscata]), (b) vulnerable species (Asiatic black bear [Ursus thibetanus]), (c) endangered species (reintroduced oriental white stork [Ciconia boyciana]), and (d) non-native, invasive species (nutria [Myocastor coypus]). We used the Potential for Conflict Index (PCI2) to analyze our data, and found that respondents’ (n?=?868) acceptance was highest for the endangered oriental white stork and lowest for the non-native nutria. These perceptions also varied with sociodemographic characteristics, including a significant negative relationship between age and willingness to have boar, deer, macaque, bears, and nutria nearby.  相似文献

In 2005, severe leaf curling and yellowing were observed on tomato plants on Ishigaki Island. Because the symptoms were consistent with infection by a begomovirus, we used a polymerase chain reaction (PCR) with specific primers for begomovirus DNA-A and DNA satellite component (DNA-β) and detected products of the expected sizes from symptomatic tomato plant samples. DNA sequence analyses of the PCR products revealed that the symptomatic tomato plants were associated with Ageratum yellow vein virus (AYVV) infection. We confirmed AYVV transmission from the naturally infected weed host, Ageratum conyzoides, to healthy tomato plants by the insect vector Bemisia tabaci B biotype. This report is the first of AYVV occurrence in Japan.  相似文献

To reveal whether bats serve as an amplifying host for Yokose virus (YOKV), we conducted a serological survey and experimentally infected fruit bats with YOKV isolated from microbats in Japan. YOKV belongs to the Entebbe bat virus group of vector unknown group within the genus Flavivirus and family Flaviviridae. To detect antibodies against YOKV, we developed an enzyme-linked immunosorbent assay (ELISA) using biotinylated anti-bat IgG rabbit sera. Serological surveillance was conducted with samples collected in the Philippines and the sera supplied from Malaysia. One of the 36 samples from the Philippines (2.7%) and 5 of the 26 samples from Malaysia (19%) had detectable ELISA antibodies. In the experimental infections, no clinical signs of disease were observed. Moreover, no significant viral genome amplification was detected. These findings revealed that YOKV replicates poorly in the fruit bat, suggesting that fruit bats do not seem to serve as an amplifying host for YOKV.  相似文献

Na+/H+ antiporters are known to play a crucial role in pH and Na+ homeostasis. In the present study, we characterized the molecular structures and expression patterns of two Na+/H+ antiporters from the marine red alga Porphyra yezoensis (designated PySOS1 and PyNhaD). The full-length cDNAs of PySOS1 and PyNhaD were 5122 and 1804?bp, and contained open reading frames (ORFs) of 4773 and 1275?bp, respectively. The deduced amino acid sequences showed high similarity to SOS1 and NhaD from the higher plant Arabidopsis thaliana. PySOS1 and PyNhaD contained conserved sequences found in the cation?Cproton antiporter. Quantitative real-time PCR analysis revealed that both antiporter genes were expressed in both the gametophyte and sporophyte of P. yezoensis. In addition, mRNA expression of PySOS1 and PyNhaD was simultaneously upregulated by light irradiation, suggesting that coordinated activity between the two is important in pH and Na+ homeostasis under light conditions. Moreover, the expression levels of both genes were partially reduced by the photosynthetic inhibitors DCMU and DBMIB, suggesting that upregulation is linked to photosynthesis-related metabolism. These findings provide an initial step towards understanding Na+/H+ antiporters in marine red algae.  相似文献

We tested whether exposing rice plants to abiotic stress (salt or shade) during vegetative growth affects the chilling tolerance of reproductive organs, which is one of the most important traits for rice growing in a cool climate; we used two rice cultivars with different tolerance in two growing seasons. We divided the vegetative growth into three phases to clarify the most sensitive period: 7–22 days after transplanting (DAT), 23–38 DAT and 39–54 DAT. Chilling tolerance of the pre-stressed plants was based on the male sterility induced by low temperatures. Shade and salt stress during all three vegetative growth phases significantly reduced stomatal conductance. Shade decreased the specific leaf weight and the leaf sugar and starch contents, but salt had no significant effect, despite causing leaf damage. Low temperatures during the reproductive stage induced spikelet sterility in all plants, but the magnitude was greater in the salt- and shade-stressed plants of both cultivars, especially those stressed late during vegetative growth. The increased spikelet sterility caused by chilling was closely related to the reduction of the total spikelet number per panicle. This is the first study to show that salt and low light stress during vegetative growth increased the susceptibility of rice plants to chilling damage during panicle development.  相似文献

Nitrogen (N) retranslocation within tree canopies has been intensively studied and assumed to function as a one-way process (e.g., from older to newer leaves). However, recent studies have found that both N output and input occur in individual leaves, suggesting that 'gross' N retranslocation exists behind 'net' N retranslocation. In the present study, the amount and direction of gross N retranslocation within a canopy of deciduous oak Quercus serrata Thunb. ex. Murray saplings were investigated. Labeling was conducted with leaves of Q. serrata saplings cultivated under conditions of low-N (LN) or high-N (HN) fertility. Subsequently, N movement within the canopy was traced. Leaves at two different positions in the canopy (top and lateral) were labeled to determine the direction of gross N retranslocation. To detect seasonal differences, the leaf-labeling experiment was conducted twice during the early and late phases of the growing season. In addition, to compare the quantitative importance of gross N retranslocation and root N uptake, the latter was determined by labeling Q. serrata roots. The N-labeling experiment revealed gross N retranslocation among leaves, i.e., from top to lateral, lateral to top and lateral to lateral positions. Gross N retranslocation was quantitatively more important than root uptake, especially for plants cultivated at LN fertility. Season also affected the amount of gross N retranslocation, and these effects differed between LN and HN fertilities. These findings suggest that N allocation within a canopy is controlled dynamically by both gross N output and input. The mechanisms controlling gross N output and input likely function as key determinants of N allocation within a tree canopy.  相似文献

Rice blast is a devastating fungal disease resulting in major losses to rice crops. Owing to continuous acquisition of resistance by the causal fungus, several fungicide chemicals are no longer effective. Therefore, there is a need to identify natural components and develop new agents to control fungal pathogens. We previously demonstrated that the culture filtrate of Biscogniauxia sp. O821 inhibited infection behavior of Magnaporthe oryzae and subsequent blast lesion formation. In the present study, we isolated a new compound, (3aS,4aR,8aS,9aR)-3a-hydroxy-8a-methyl-3,5-dimethylenedecahydronaphto[2,3-b]furan-2(3H)-one (HDFO), from the culture filtrate of Biscogniauxia sp. O821 and determined its molecular weight as 248. The HDFO structure was determined by electrospray ionization-mass spectrometry and nuclear magnetic resonance spectroscopy after purification with column chromatography and high-performance liquid chromatography. The structure of this antifungal compound was similar to that of alantolactone and isoalantolactone. The growth inhibition zone against M. oryzae in presence of HDFO was observed at Rf 0.5–0.6 on a thin layer chromatography plate. HDFO inhibited conidial germination of M. oryzae in a dose-dependent manner (1–200 ppm). Furthermore, blast lesion formation was significantly suppressed by HDFO at over 5 ppm. These results suggest that HDFO from the culture filtrate of Biscogniauxia sp. O821 can protect rice from rice blast disease caused by M. oryzae. This is the first report that HDFO produced by Biscogniauxia sp. can serve as an antifungal compound against M. oryzae.  相似文献

Hypothalamus-independent negative feedback mechanism of the pituitary-thyroid axis was studied by electron microscopic observation of thyroid follicular cells of encephalectomized or hypophysectomized fetal rats under hypothyroidism induced by propylthiouracil (PTU). Fetal hypophysectomy or encephalectomy was performed on day 19 of gestation. Mother rats were given either a daily amount of 40 mg PTU or saline alone on the day of operation and on the next day. On day 21, their fetuses were obtained. As a result, PTU induced marked goiters even in encephalectomized fetuses, the goiters being comparable in weight to those induced in littermate intact fetuses. Electron microscopically, the follicular lumen was narrowed, microvilli were lengthened with a marked increase in number, and rER was extremely expanded. Follicular cells often confined colloid droplets and lysosomes. Fetal hypophysectomy, even in PTU-treatment, caused a reduction in the thyroid weight, accompanied by flattening of follicular cells, shortening of microvilli, widening of follicular lumen, and shrinkage of rER. These findings would support the view that the negative feedback mechanism of the pituitary-thyroid axis can operate without the hypothalamus in fetal rats.  相似文献

The caliber of the umbilical arteries was measured in fetal rats on day 14 to 21 of gestation. The fetuses were rapidly frozen in an acetone-dry ice mixture. The caudal half of each whole-body frozen fetus was gradually shaved with a knife from the ventral side to expose the umbilical arteries for measurement of their calibers. Observation reveals that, on day 15 of gestation, the umbilical arteries are asymmetrical in that the right artery is more dilated than the left. It is further seen that the left umbilical artery is closed on day 17 in contrast to the right which is increasingly more dilated toward term. When indomethacin, a synthetic compound having an action similar to that of glucocorticoids, was administered to pregnant rats 3 hr prior to sacrifice on day 21 of gestation, the right fetal umbilical artery was significantly constricted. The reason for this phenomenon is unknown, but is discussed in association with the manner of closure of the ductus arteriosus.  相似文献

An esterase or esterases in acetone powder preparations of mouse liver microsomes hydrolyze the cyclopropanecarboxylate ester linkage of pyrethroid insecticide chemicals derived from primary alcohols. The rate of cleavage of (+)-trans-chrysanthemates with various alcohol moieties decreases in the following order: 5-propargyl-2-furylmethyl; 5-benzyl-3-furylmethyl (bioresmethrin); 3-phenoxybenzyl; tetrahydrophthalimidomethyl esters. The hydrolysis rate of benzylfurylmethyl esters with various acid moieties decreases in the order: (+)- or (?)-trans-chrysanthemate; (+)-trans-ethanochrysanthemate; tetramethylcyclopropanecarboxylate; (+)- or (?)-cis-chrysanthemate or (+)-cis-ethanochrysanthemate. The trans-isomers of chrysanthemates and ethanochrysanthemates are hydrolyzed from 2.6- to more than 50-fold more rapidly than the corresponding cis-isomers. This enzyme system does not hydrolyze secondary alcohol esters, i.e., allethronyl (+)-trans- and (+)-cis-chrysanthemates.On intraperitoneal administration to mice, the (+)-trans-chrysanthemate and -ethanochrysanthemate of benzylfurylmethanol are of very low toxicity relative to the corresponding (+)-cis-isomers and the tetramethylcyclopropanecarboxylate. S,S,S-tributyl phosphorotrithioate (DEF) pretreatment increases the toxicity of these five compounds by 2.6- to more than 188-fold, with the exception of bioresmethrin whose toxicity is not altered. When the toxicity is increased, it is probably the result of esterase inhibition since DEF strongly inhibits the esterase activity of fresh liver microsomes while the mixed-function oxidase system remains active. The oxidase system metabolizes the chrysanthemates more rapidly than the ethanochrysanthemates of benzylfuryl-methanol. Depending upon the pyrethroid involved, the esterase or the mixed-function oxidase system, or both may be responsible for limiting the toxicity of these pyrethroids to mice.  相似文献
