We established a simple, rapid method for gizzerosine analysis in fish meal. Gizzerosine was extracted from fish meal with 0.1?N HCl solution. Samples and standard gizzerosine solutions were absorbed onto a paper disc, which was then set on electrophoresis paper for 18?min, and the paper was dried. Gizzerosine was visualized with Pauly??s reagent, and the intensity of the colored spots was digitized and calculated by image processing method software. We achieved successful separation of gizzerosine from other Pauly??s reagent-positive components in fish meal extracts. The linearity of gizzerosine estimation using this method was within the range 30?C1000?ng (R 2?=?0.99). Gizzerosine was satisfactorily detected and completely separated from histamine and other Pauly??s reagent-positive compounds. This method does not require expensive instruments or tedious pretreatment to eliminate interfering compounds, such as histamine or histidine. It also uses less reagent compared with high-performance liquid chromatography. Moreover, it is a simple, rapid, sensitive, and reproducible method. It is suitable for monitoring gizzerosine in fish meal products that contain as little as 10?ppm gizzerosine. 相似文献

Hyperlipidemic ocular lesions are described for Watanabe heritable hyperlipidemic (WHHL) rabbits. Male WHHL rabbits 8 months old exhibited serum hyperlipidemia and ophthalmoscopically yellowish-white lesions along the corneoscleral junction and in the iris. Histopathologically, foamy macrophages aggregated in the stroma of the cornea, iris, and ciliary body were observed. These findings have been interpreted as lipid keratopathy. In addition, multiple clusters of a large number of foamy macrophages occurred throughout the choroid and sclera in association with the blood vessels. The lesions in the choroid and sclera could not be detected ophthalmoscopy, yet were much more prominent than those in the cornea, iris, and ciliary body, suggesting greater involvement and earlier onset of lipidosis at these sites associated with hyperlipidemia in WHHL rabbits. 相似文献

Reaction of Japanese beech (Fagus crenata) in an ionic liquid, 1-ethyl-3-methylimidazolium chloride ([C2mim][Cl]), which can dissolve cellulose, was investigated.
Although both lignin and polysaccharides such as cellulose and hemicelluloses can be liquefied at a treatment temperature
of around 100°C, the liquefaction of polysaccharides mainly occurs at the beginning of the treatment with [C2mim][Cl]. Cellulose
crystallinity in the wood was gradually broken down as the treatment continued. The solubilized polymers were depolymerized
to low molecular weight compounds. The results indicate that [C2mim][Cl] is an effective solvent and reagent for the liquefaction
of wood components and subsequent depolymerization of them.

Enzymatic saccharifications of the hydrolysate collected as the water-soluble (WS) portion after hot-compressed water (HCW)
treatment of Japanese beech with xylanase and β-xylosidase were performed to investigate the hydrolytic properties of these enzymes. The WS portions with different molecular
weight distributions and initial concentrations of xylooligosaccharides were prepared as substrates, one of which contained
mainly smaller xylooligosaccharides [degree of polymerization (DP) = 2 or 3], while the other contained larger xylooligosaccharides
(DP ≥ 4). The highest xylose recovery was obtained from both of the WS portions treated with β-xylosidase rather than with xylanase. This suggests that β-xylosidase could have a higher activity than xylanase toward larger xylooligosaccharides (DP ≥ 4) as well as smaller xylooligosaccharides
(DP = 2 or 3) recovered from HCW treatment. As a result, the use of β-xylosidase was found to be effective for saccharifi cation of the WS portion, even with a short reaction time for the HCW

Water relations in woody species are intimately related to xylem hydraulic properties. High CO(2) concentrations ([CO(2)]) generally decrease transpiration and stomatal conductance (g(s)), but there is little information about the effect of atmospheric [CO(2)] on xylem hydraulic properties. To determine the relationship between water flow and hydraulic structure at high [CO(2)], we investigated responses of sun and shade leaves of 4-year-old saplings of diffuse-porous Betula maximowicziana Regel and ring-porous Quercus mongolica Fisch. ex Ledeb. ssp. crispula (Blume) Menitsky grown on fertile brown forest soil or infertile volcanic ash soil and exposed to 500 micromol CO(2) mol(-1) for 3 years. Regardless of species and soil type, elevated [CO(2)] consistently decreased water flow (i.e., g(s) and leaf-specific hydraulic conductivity) and total vessel area of the petiole in sun leaves; however, it had no effect on these parameters in shade leaves, perhaps because g(s) of shade leaves was already low. We investigated the relative sensitivity of duckweed Lemna minor and six species of algae to seven herbicides, using an efficient high-throughput microplate-based toxicity assay. First, we assessed the sensitivity of L. minor to the seven herbicides, and then we compared its sensitivity to that of previously published data for six algal species based on EC50 values. For five herbicides, the most sensitive species differed: L. minor was most sensitive to cyclosulfamuron: Raphidocelis subcapitata was most sensitive to pretilachlor and esprocarb: Desmodesmus subspicatus was most sensitive to pyraclonil; and Navicula pelliculosa was most sensitive to pyrazoxyfen. Simetryn was evenly toxic to all species, whereas 2,4-D was evenly less toxic, with only small differences in species sensitivity. These results suggested that a single algal species cannot represent the sensitivity of the primary producer assemblage to a given herbicide. Therefore, to assess the ecological effects of herbicides, aquatic plant and multispecies algal toxicity data sets are essential. 相似文献

An ethyl acetate extract of a culture filtrate (ECF) from an unidentified fungal isolate O821 was evaluated for antifungal activity against the rice pathogen Magnaporthe oryzae. The O821-ECF significantly inhibited spore germination, appressorium formation, and mycelial growth of M. oryzae, and its antifungal activity was heat-stable. It also significantly suppressed the number and size of blast lesions. In an analysis of the ITS sequence of this isolate, it shared similarities with species of the fungus Biscogniauxia. These results suggest that isolate O821 of the genus Biscogniauxia produces a heat-stable antifungal compound(s) in its culture filtrate. 相似文献

Yellow mosaic disease, caused by wheat yellow mosaic virus (WYMV), is one of the most serious diseases of winter wheat in Japan and China. A single major QTL for WYMV resistance in the Japanese wheat variety 'Yumechikara', designated Q.Ymym, has been mapped on a 43.6 cM linkage block between the two markers Xcfd233 and Xgwm349 on chromosome 2D. We were able to obtain two recombinants within the block, which facilitated reducing the size of the linkage block. The pseudomolecule sequence of 'Chinese Spring' (CS) indicated that the original Q.Ymym region of 43.6 cM corresponded to 68.5 Mb and the narrowed Q.Ymym region represents a size of 27.3 Mb. The sequence features of the Q.Ymym region were unique in comparison with CS sequences, which may have led to the low recombination rate within the block. The Q.Ymym haplotype block was detected in other WYMV-resistant varieties but not in the susceptible varieties used in this study. The unique sequence structure of the Q.Ymym region allowed the development of co-dominant markers for use in marker-assisted selection. 相似文献

In northern Japan, juvenile chum salmon Oncorhynchus keta (Walbaum) are released from hatcheries to enhance the fishery resource. Infections with ectoparasitic protozoans, particularly the flagellate Ichthyobodo salmonis and the ciliate Trichodina truttae, occasionally cause severe mortality among hatchery‐reared juveniles. This study examined the susceptibility of the two parasites to wide‐ranging UV irradiation (experiment 1) and then investigated whether UV disinfection of the rearing water using a commercial device was useful for preventing infections among juveniles in a small‐scale rearing system over a 28‐day period (experiment 2). In experiment 1, parasite mortality reached 100% with UV irradiation doses of ≥9.60 × 105 μW s/cm2 for I. salmonis and ≥8.40 × 105 μW s/cm2 for T. truttae. In experiment 2, disinfection of the rearing water at a UV irradiation dose of 2.2 × 106 μW s/cm2 succeeded in complete prevention of both parasites in the juvenile salmon. These results elucidate the minimum dose of UV irradiation for inactivation of I. salmonis and T. truttae, and demonstrate the usefulness of water disinfection using a commercial UV irradiation device to prevent infections by these parasites in hatchery‐reared juvenile chum salmon. 相似文献

Terminal molting probability (TMP) of snow crab Chionoecetes opilio was estimated using instar- and state-structured model incorporating the terminal molt in the waters off the Pacific coast
of northern Honshu, Japan. TMP is defined as the probability that terminal molt occurs within a year in an instar and is an
important parameter for stock assessment. TMP was estimated from the model and the rate of terminal molted crabs (RTM) for
instars, which means carapace widths over 30 mm. Carapace widths at RTM of 0.5 were 81 mm for males and 66 mm for females.
TMP was estimated as 0.23 and 0.29 for these carapace widths, respectively. Carapace widths at TMP of 0.5 were 94 mm for males
and 71 mm for females, and for these carapace widths 57% of male and 50% of female crabs were considered as old-shell which
had survived 1 year or more from terminal molt. Changing the fishing or natural mortality by ±0.1, the maximum difference
of TMP was less than 0.1 from default estimates. 相似文献
