Pasting characteristics of maize starch heat‐treated with six different water‐to‐ethanol ratios (%wt base 0:100, 10:90, 20:80, 30:70, 40:60, 50:50) were investigated; treated starches were called EW 0, 10, 20, 30, 40, and 50, respectively. Endotherms in DSC analysis shifted to a higher temperature as the water content in water‐ethanol mixture increased. The removed amount of fatty acids was much higher in treatments for EW 10, 20, and 30. The RVA peak viscosity of EW 10 and 20 were highest among the treated starches and setbacks were more than twice that of untreated starch. The characteristic change in the RVA viscogram corresponded to the amount of leached amylose from the granule. EW 30 displays similar properties as conventional heat‐moisture‐treated starch, but maintained a higher viscosity of ≈300 RVU throughout the heating process. In treatment with water‐ethanol mixtures, heat‐moisture treatment and defatting effects generated new types of modified starches. EW 40 and 50 had no clear pasting peak on RVA, and showed a viscosity at low temperature similar to granular cold water gelling.  相似文献

An 8-year 8-month-old castrated male Munchkin presented with vomiting, anorexia and hypoactivity. Computed tomography revealed excessive gas accumulation within the intestinal lumen and gas bubbles in the liver, spleen, and portal venous system, indicating hepatic portal venous gas. The cat died without any significant improvement, and mild splenomegaly was found at necropsy. Histologically, multiple gas vacuoles were diffusely observed in the liver and spleen. In the stomach, multiple gas vacuoles and scattered focal ulcers were detected within the mucosa. Multifocal hemorrhage was noted in the small and large intestines, whereas gas vacuoles were not present. Based on these findings, a gastric ulcer under high gas pressure may have provided an entry point for gas into the portal venous system.  相似文献

The target site of the novel fungicide quinofumelin was investigated in the rice blast fungus Pyricularia oryzae. Quinofumelin-induced mycelial growth inhibition was reversed by orotate but not by dihydroorotate. Recovery tests suggested that the target site of quinofumelin was dihydroorotate dehydrogenase (DHODH), which catalyzes the oxidation of dihydroorotate to orotate. Quinofumelin strongly inhibited P. oryzae class 2 DHODH (DHODH II) (IC50: 2.8 nM). The inhibitory activities of mycelial growth and DHODH II were strongly positively correlated, indicating that DHODH II inhibition by quinofumelin lead to antifungal activity. A P. oryzae DHODH II gene (PoPYR4) disruption mutant (ΔPopyr4), showing the same tendency as the quinofumelin-treated wild strain in recovery tests, was constructed, and disease symptoms were not observed in rice plants infected by ΔPopyr4. Thus, DHODH II, which plays an important role in pathogenicity and mycelial growth, is found to be the target site of quinofumelin.  相似文献

Developed by Mitsui Chemicals Agro, Inc. (Tokyo, Japan), quinofumelin is a novel fungicide with a distinct chemical structure including 3-(isoquinolin-1-yl) quinoline, demonstrating fungicidal activity against a variety of fungi, including rice blast and gray mold. We screened our compound library to identify curative compounds for rice blast and evaluated the effect of fungicide-resistant strains of gray mold. Our research demonstrated that quinofumelin has curative effects against rice blast and is not cross-resistant to existing fungicides. Accordingly, the use of quinofumelin can be considered a novel approach for disease control in agricultural production. In this report, the discovery of quinofumelin from the initial compound is described in detail.  相似文献

Witchweed, Striga hermonthica (hereafter, referred to as “Striga”), is a major biotic constraint to cereal production in sub‐Saharan Africa. The parasitic plant is a socioeconomic problem that has forced some resource‐poor farmers to abandon their farms due to high infestation. This study was designed in order to elucidate farmers' perceptions of Striga control measures and to determine their potential adoption in two villages in western Kenya. Participatory rural appraisals and individual interviews were conducted in 2009 and 2010 in a sample of 128 and 120 households in Kaura and Kogweno‐Oriang villages in Homabay and Rachuonyo districts, respectively. The results revealed that crop production was the main occupation in most households. The farmers identified Striga as one of the major constraints to maize, sorghum, and finger millet production. According to the farmers, the most popular control measures were hand‐pulling, crop rotation, and intercropping, even though rotational systems might need a longer timeframe to reduce the soil seed bank of Striga. Although the level of Striga infestation and damage were increasing in the farmers' fields, the adoption of the control options was limited. The reason for the low adoption level of the control methods by the farmers is because they are “too risky” as there is no guarantee of a direct pay‐off in increased crop yield. Farmer‐led evaluation and adaptation of the various Striga control technologies in real‐life situations will facilitate the choice of appropriate options and facilitate their uptake.  相似文献

Differences between average allelic frequencies of genes that relate to traits suggest that it would be evidence of artificial selections. Sliding window approach is a useful method to identify genomic regions that have been differently selected between two breeds. The objective of this study was to identify the divergently selected regions between Japanese Black (JB) and Japanese Holstein (JH) cattle based on genotypic information obtained through a high‐density single nucleotide polymorphism (SNP) panel. After genotyping of 54 001 SNP markers on 100 animals (50 JB and 50 JH), 40 635 SNPs were suitable for the analysis. For each of these SNPs, the absolute difference between allelic frequencies of JB and JH was calculated. In the current study, 10 consecutive SNPs were defined as components of a window. For each window, the average difference in allelic frequency was calculated. This was termed sliding window average difference (SWAD). Among 40 055 windows, we focused on 39 windows with the largest SWAD. This was equivalent to 0.1% of all windows and the SWAD was more than 0.435. Some of these windows overlapped and were distributed in 11 regions. These regions were in good agreement with reported quantitative trait locus, therefore would be selection signatures and good candidates that harbor the causative mutations.  相似文献

Iron deficiency stress causes a severe reduction in plant growth. Although Fe deficiency causes an imbalance in divalent heavy metal nutrients, the mechanisms underlying the growth reduction caused by this imbalance remain unclear. We investigated Zn uptake and accumulation in maize under Fe-deficient conditions. Under Fe-deficient conditions, Zn uptake was 15-fold higher and Zn accumulation was 16-fold higher than that under normal nutrient conditions. The Zn content of maize leaves under Fe-deficient conditions was >0.4 mg g−1 dry weight, which was higher than the content of plants grown in a nutrient solution containing 50 µM ZnCl2. Plant growth under conditions of both Fe and Zn deficiency was significantly higher than that under only Fe-deficient conditions. Moreover, Fe deficiency increased the thiol content of the plant. These results indicate that Fe deficiency causes excess uptake and accumulation of Zn, and that the stress resulting from the Zn overload accelerates growth reduction in maize.  相似文献

Pyrroloquinoline quinone (PQQ) is believed to be an important factor for mammalian growth and development and has, therefore, been declared a vitamin by some researchers. However, this issue remains controversial, and from a nutritional viewpoint, accurate determination of PQQ levels in a variety of foods is very important. Here, we describe a simple, highly sensitive, and highly selective method for quantitative analysis of PQQ. Liquid foods or aqueous extracts of solid foods were analyzed using high-performance liquid chromatography (HPLC) combined with electrospray-ionization (ESI) tandem mass spectrometry (MS/MS). (15)N-labeled PQQ was added to the samples as an internal standard. Quantitative analyses of PQQ were performed by multiple reaction monitoring (MRM) with LC/MS/MS. Free PQQ was detected in almost all food samples in the range 0.19-7.02 ng per g fresh weight (for solid foods) or per mL (liquid foods). This method will enable the rapid and simple determination of PQQ levels in many samples.  相似文献

A bidirectional single-electron counting device is demonstrated. Individual electrons flowing in forward and reverse directions through a double quantum dot are detected with a quantum point contact acting as a charge sensor. A comprehensive statistical analysis in the frequency and time domains and of higher order moments of noise reveals antibunching correlation in single-electron transport through the device itself. The device can also be used to investigate current flow in the attoampere range, which cannot be measured by existing current meters.  相似文献
