ABSTRACT: Scrapie in sheep and goats has been known for more than 250 years and belongs nowadays to the so-called prion diseases that also include e.g. bovine spongiform encephalopathy in cattle (BSE) and Creutzfeldt-Jakob disease in humans. According to the prion hypothesis, the pathological isoform (PrPSc) of the cellular prion protein (PrPc) comprises the essential, if not exclusive, component of the transmissible agent. Currently, two types of scrapie disease are known - classical and atypical/Nor98 scrapie. In the present study we examine 24 cases of classical and 25 cases of atypical/Nor98 scrapie with the sensitive PET blot method and validate the results with conventional immunohistochemistry. The sequential detection of PrPSc aggregates in the CNS of classical scrapie sheep implies that after neuroinvasion a spread from spinal cord and obex to the cerebellum, diencephalon and frontal cortex via the rostral brainstem takes place. We categorize the spread of PrPSc into four stages: the CNS entry stage, the brainstem stage, the cruciate sulcus stage and finally the basal ganglia stage. Such a sequential development of PrPSc was not detectable upon analysis of the present atypical/Nor98 scrapie cases. PrPSc distribution in one case of atypical/Nor98 scrapie in a presumably early disease phase suggests that the spread of PrPSc aggregates starts in the di- or telencephalon. In addition to the spontaneous generation of PrPSc, an uptake of the infectious agent into the brain, that bypasses the brainstem and starts its accumulation in the thalamus, needs to be taken into consideration for atypical/Nor98 scrapie.  相似文献

In a 4‐week experiment, 15 cannulated rainbow trout were fed three diets based on fish meal (FM), Saccharomyces cerevisiae yeast (SC) and Wickerhamomyces anomalus and S. cerevisiae yeast mix (WA). Fish were fed daily, and blood samples were collected on day 7 of each week at 0, 3, 6, 12 and 24 hr after feeding. In the final week, fish were exposed to a 1‐min netting stressor. All essential and non‐essential plasma amino acid levels except methionine were similar between fish fed diets FM, SC and WA. Plasma methionine and sarcosine were significantly higher in fish fed diets SC and WA, possibly due to the crystalline methionine level, form or feeding regime. Hydroxy‐proline and 3‐methyl‐histidine were higher in fish fed diet FM, which can be explained by the higher levels present in fish meal compared with yeast. In stressed fish, there were no dietary effects on plasma amino acid levels, but significant increases in taurine and cystathionine were found in stressed compared with unstressed fish. These results demonstrate that yeast‐based diets produce similar plasma amino acid profiles to fish meal and suggest that yeast may be a suitable fish meal replacement in diets for rainbow trout.  相似文献

Standing dead trees (snags) play important roles in forest ecology by storing carbon as well as providing habitats for many species. Moreover, snags preserved for hundreds of years can provide useful data to extend tree-ring chronologies used for climatological and ecological studies beyond the lifespans of living trees. Here we examined the growth patterns of Scots pine snags from the central Scandinavian Mountains, in relation to still living trees. Using changes point analyses, we showed that a majority (74%) of the snags displayed significant negative growth changes prior (on average 17 years) to death. Change points around the same years were also seen in living trees, but they recovered their growth. The average growth reduction of snags showing negative growth changes before death was 46%. In most cases the final growth change points coincided with very cold summers, or to a lesser degree to period of cool summers. It was suggested that pines ending up as snags were less resilient than the trees which continued living, and thus not able to recover after cold summer events. Since the snag growth reductions prior to death were partly unrelated to climate, care should be taken when using such data in dendroclimatological studies.  相似文献
