Vibrio vulnificus is a Gram-negative pathogenic bacterium that causes serious infections in humans and requires iron for growth. A clinical isolate, V. vulnificus M2799, secretes a catecholate siderophore, vulnibactin, that captures ferric ions from the environment. In the ferric-utilization system in V. vulnificus M2799, an isochorismate synthase (ICS) and an outer membrane receptor, VuuA, are required under low-iron conditions, but alternative proteins FatB and VuuB can function as a periplasmic-binding protein and a ferric-chelate reductase, respectively. The vulnibactin-export system is assembled from TolCV1 and several RND proteins, including VV1_1681. In heme acquisition, HupA and HvtA serve as specific outer membrane receptors and HupB is a sole periplasmic-binding protein, unlike FatB in the ferric-vulnibactin utilization system. We propose that ferric-siderophore periplasmic-binding proteins and ferric-chelate reductases are potential targets for drug discovery in infectious diseases. 相似文献

We investigated the expression of gustducin in chicken taste buds using molecular biological, biochemical and immunohistochemical techniques. Expression of a gustducin‐like sequence was detected by RT‐PCR in the tissues containing taste buds, and corresponded to the predicted gustducin gene in the chicken. Expression of this sequence was not detected in the brain, heart, liver, pancreas, intestine, kidney and testis of the chicken. The expressed sequence had a high specificity for oral tissues that contained taste buds. These results suggest that the detected sequence was the chicken gustducin gene. Next, we generated a polyclonal antiserum against the chicken gustducin protein to observe its localization in the oral tissues. The results revealed that the chicken gustducin was specifically expressed in the taste buds. It is suggested that the chicken has a gustatory system mediated by gustducin, and chicken gustducin is a reliable marker for taste buds or taste cells. This is the first molecular biological, biochemical and immunohistochemical demonstration of the presence of gustducin in the chicken. 相似文献

Trichodectes pinguis, referred to commonly as the bear-biting louse, has been reported in several bear species. However, graphical (blurred or coarse) and genetic information on the louse is limited. In this study, we identified T. pinguis collected from Japanese black bears in the Aomori Prefecture, Japan. We confirmed 12S rDNA sequences derived from the collected T. pinguis and performed molecular phylogenetic analysis based on 12S rDNA. The analysis revealed the parasitic louse to be T. pinguis. Interestingly, the body size of T. pinguis found in this study was smaller than the previous recorded body size of them in Japan and Turkey. To better understand the biting louse infesting bears, morphometric and genetic information from other bear hosts needs to be accumulated. 相似文献

Symptoms of Welsh onion leaf blight, caused by Stemphylium vesicarium, are divided into two types, i.e., brown oval lesions and yellow mottle lesions. Yellow mottle lesions exert considerable
economic damage on Welsh onion in northern Japan. In this study, we investigated the life cycle of the pathogen in terms of
seasonal fluctuation of spore dispersal and its relationship with development of disease, formation period of pseudothecia
and overwintering of the pathogen based on field surveys, spore trapping and fungal isolation. Conidia were trapped throughout
the cropping season except before mid June, when no ascospores were trapped. Brown oval lesions, which contained a large number
of conidia, usually occurred in July followed by yellow mottle lesions with an increasing number of conidia trapped. These
observations suggest that conidia released from brown oval lesions play an important role as a secondary inoculum source of
the disease, leading to the development of yellow mottle lesions. Pseudothecia on leaves were first observed at the end of
the cropping season or immediately after harvest (late October). The pathogen overwintered in the form of pseudothecia produced
on leaves with or without symptoms. Ascospores failed to be trap in the field during the interval between before and beginning
of the cropping season in April–May. However, pot experiments demonstrated that ascospores were released from leaf debris
in November and rapidly increased in number after snow melt. From this circumstantial evidence, we hypothesize that ascospores
are the primary inoculum source of Welsh onion leaf blight. 相似文献

Feeding sites for wild Yeso sika deer around Lake Akan, Japan, were established. Effects on the number of deer using the feeding sites, the prevention of bark stripping damage, the amount of feeding, and eating time in a 5‐year period (1999–2003) were evaluated. The number of deer using feeding sites increased with years during the feeding period. The damaged tree ratio after the initiation of feeding markedly decreased compared with 16.5% before the initiation of feeding. After the start of feeding, there were no trees with damage the entire circumference. According to tree species, the number of damaged trees of Ulmus laciniata Mayr as a percentage of all investigated trees was high (5.2%). The total amount of beet pulp feeding increased with the feeding year, showing 4.5‐fold increase. At feeding sites in deer culling, eating behavior was observed during the night. The preventive effects on bark stripping damage continued during the 5‐year feeding period. However, with the course of feeding years, the number of deer using feeding sites and the level of feeding increased. 相似文献

A male crossbred calf developed a limp and pain upon deep pressure on the right hind limb and the right forelimb. The radiographic findings of affected limbs and pathological findings of bone biopsy were similar to those observed in canine panosteitis. The anthocyanin profiles and varieties/breeding line differences of anthocyanin concentrations in common/tartary buckwheat sprouts have been studied. Four anthocyanins, cyanidin 3-O-glucoside, cyanidin 3-O-rutinoside, cyanidin 3-O-galactoside, and cyanidin 3-O-galactopyranosyl-rhamnoside, were isolated from the sprouts of common buckwheat, were separated using high-performance liquid chromatography (HPLC), and were identified using reversed-phase liquid chromatography (LC)/electrospray ionization-mass spectrometry (ESI-MS)/MS techniques. In tartary buckwheat sprouts, two anthocyanins (cyanidin 3-O-glucoside and cyanidin 3-O-rutinoside) were identified. Among 19 common/tartary buckwheat varieties/breeding lines, Hokkai T10 contained the highest amounts of anthocyanins. Cyanidin 3-O-glucoside and cyanidin 3-O-rutinoside concentrations in 6-10 days after seeding sprouts of Hokkai T10 ranged from 0.16 to 0.20 mg/g dry wt and from 5.55 to 6.57 mg/g dry wt, respectively. In addition, dark-grown sprouts of Hokkai T10 accumulated 0.091 and 2.77 mg/g dry wt of cyanidin 3-O-glucoside and cyanidin 3-O-rutinoside whereas other varieties/breeding lines accumulated trace amounts of anthocyanins. Given its anthocyanin-rich red cotyledons, Hokkai T10 is a promising line for use as "Moyashi" type sprouts and is strongly recommended as a new functional food, rich in dietary anthocyanins. 相似文献

The present immunohistological study was conducted to investigate the localization of fatty acid translocase CD36 (FAT/CD36) in the skeletal muscle (Biceps femoris) fibers of bovine, swine and ovine. The results showed that CD36 was mainly localized in type I muscle fiber of these animals. In contrast, FAT/CD36 localization in type II fiber was insignificant in the types of muscle in the present experiments, suggesting that type II fiber of bovine, swine and ovine might lack fatty acid translocase FAT/CD36. 相似文献
