The objective of the present study was to investigate the influence of prostaglandin F(2alpha) (PGF (2alpha)) and nitric oxide (NO) on production of steroids and PGs by culturing bovine luteal cells obtained from ovaries on days 8-12 of the estrous cycle with a nitric oxide (NO) donor (Spermine NONOate), and a NO synthase inhibitor (N(G)-nitro-L-arginine methyl ester dihydrochloride: L-NAME). When the cells were exposed for 24 h to PGF(2alpha) (10(-7)-10(-5) M), production of progesterone (P(4)) increased significantly at all doses used (P<0.05). Moreover, PGF(2alpha) stimulated PGF(2alpha) production (P<0.01), depressed testosterone (T) production (P<0.05), but did not affect synthesis of prostaglandin E(2) (PGE(2)). Spermine NONOate decreased P(4) production to 66%, 47% and 34% of the control concentration after treatment with 10(-5) M, 10(-4) M and 10(-3) M, respectively, but did not affect T production, and increased PGF(2alpha) synthesis (P<0.05) and PGE(2) (P<0.01) at all doses used. L-NAME increased production of P(4) (P<0.01) but did not affect (P>0.05) secretion of T, PGF(2alpha) and PGE(2). Estradiol-17beta (E(2)) was detectable on the level of sensitivity of assay and was not significantly altered by any treatments. The overall results suggest that PGF(2alpha) and NO produced locally in bovine CL play roles in the regulation of the secretory function of the bovine CL as auto/paracrine factors. 相似文献

Laboratory experiments were carried out to investigate bioclogging on three types of online emitters using synthetic wastewater filtered at 10 μm (COD = 200 mg L?1). The three types of emitters were as follows: non-pressure-compensating emitters delivering 2 L h?1 (NPC2), non-pressure-compensating emitters delivering 4 L h?1 (NPC4) and pressure-compensating emitters delivering 2.2 L h?1 (PC). They were plugged on two parallel pipe diameters (16 and 20 mm). Emitter performance was followed by flow rate measurements. The results showed that PC emitters seemed to be more sensitive to clogging than NPC emitters. Biofilm was observed mainly on NPC flow paths and PC membranes; its presence was also confirmed by the assessment of different water quality parameters such as total organic carbon and pH. Experimental results compared with a model designed with computational fluid dynamics Software (ANSYS Fluent®) showed relations between biofilm development and velocity distribution on NPC flow paths. Information about disease management in winter wheat (Triticum aestiva) in eight European countries was collated and analysed by scientists and extension workers within the European Network for the Durable Exploitation of Crop Protection Strategies (ENDURE). This included information about specific disease thresholds, decision support systems, host varieties, disease prevalence and pathogen virulence. Major differences in disease prevalence and economic importance were observed. Septoria tritici blotch (Mycosphaerella graminicola) was recognized as the most yield reducing disease in countries with intensive wheat production, but also rust diseases (Puccinia striiformis and Puccinia triticina), powdery mildew (Blumeria graminis) and Fusarium head blight (Fusarium spp.) were seen as serious disease problems. Examples of current integrated pest management (IPM) strategies in different countries have been reported. Disease management and fungicide use patterns showed major differences, with an average input equivalent to 2.3 full dose rates (TFI) in the UK and a TFI of 0.6 in Denmark. These differences are most likely due to a combination of different cropping systems, climatic differences, disease prevalence, and socio-economic factors. The web based information platform www.eurowheat.org was used for dissemination of information and results including information on control thresholds, cultural practices which can influence disease attack, fungicide efficacy, fungicide resistance, and pathogen virulence, which are all elements supporting 1PM for disease control in wheat. The platform is open to all users. The target groups of EuroWheat information are researchers, advisors, breeders, and similar partners dealing with disease management in wheat. 相似文献

The design and tests of slow-release formulations of sulfometuron (SFM), an anionic sulfonylurea herbicide, are described. The formulations are based on incorporation of the herbicide in octadecyltrimethylammonium (ODTMA) micelles, which adsorb on a clay mineral, montmorillonite. An optimization of herbicide/micelle clay ratios yielded high adsorption of SFM (95%), and at a 1% (w/w) water suspension only 0.5% of the adsorbed SFM was released at times varying from hours to 9 days. An analytical test in Seville soil showed that under excessive irrigation (400 mm) 100% of the commercial formulation leached, whereas the micelle-clay formulations showed only 50-65% elution. A plant bioassay in Rehovot soil showed that the commercial dispersible granule formulation (Oust, 75% ai sulfometuron methyl) yielded only 23% root elongation inhibition at the top 5 cm of the soil, whereas complete inhibition was achieved with the micelle-clay formulation. The detected concentration of SFM for the micelle-clay formulation at a depth of 15-20 cm was half of that detected for the commercial one, indicating a reduction in leaching when applying the micelle-clay formulation. A 10-fold reduction in the applied dose of SFM in the micelle-clay formulations resulted in good herbicidal activity of 60-87% inhibition. Adsorption-desorption studies of norflurazon on 17 soils of very different characteristics have been performed using a batch equilibration method and correlated to its mobility, activity, and persistence in soils. The influence of different soil properties and components on norflurazon adsorption was determined. The significant variables were organic matter (OM) content and iron and aluminum oxides, which accounted for 85 and 11% of the variability, respectively. Norflurazon desorption from soils was hysteretic in all cases, being more irreversible at the lowest herbicide concentrations adsorbed. The percentage of norflurazon eluted from columns of selected soils reached almost 100% in soils with sand content >80% and OM <1%, but in the soil which gave the highest sorption, herbicide residues were not detected at depths >16 cm. The herbicidal activity of norflurazon was followed by measuring its bleaching effect on soybean plants, and the herbicide concentration required to give 50% chlorophyll inhibition (CI(50)) was calculated. CI(50) was achieved on a sandy soil with 0.08 mg x kg(-)(1), whereas 1.98 mg x kg(-)(1) was necessary for the soil that presented maximum norflurazon adsorption. 相似文献

This study investigated the in vitro and in vivo efficacy of trichlorfon against Dawestrema cycloancistrium, as well as its physiological effects on arapaima. Naturally parasitized arapaima gill arches were exposed in vitro to 100, 250, 500 and 750 mg/L of trichlorfon and a control group (only distilled water), in triplicate. Parasites were monitored, and mortality was used to determine the median effective concentration (EC50). The 750 mg/L concentration demonstrated 100% in vitro efficacy against D. cycloancistrium after 60 min, while the intermediate (500 mg/L) and the lowest (100 and 250 mg/L) tested concentrations were completely efficient after 90 and 130 min, respectively. The EC50-1h of trichlorfon for D. cycloancistrium was determined at 171.73 mg/L. Parasitized arapaima juveniles were exposed to a control group and 150 mg/L of trichlorfon in triplicate. Fish were exposed to two therapeutic baths for 60 min with 24-h intervals between treatments. Therapeutic baths with 150 mg/L of trichlorfon were 92.99% effective against D. cycloancistrium and did not bring about haematological alterations (erythrogram, white blood cell count, thrombogram, plasma glucose and total proteins). Therefore, 150 mg/L of trichlorfon can be used in therapeutic baths to control and treat D. cycloancistrium infestations with no physiological impairments for arapaima. 相似文献

The influence of 457 bacteria isolated from soil on spore germination by Aspergillus flavus was studied by light and scanning microscopy. Bacteria were found to be in physical association with the fungus. The bacteria were tested for antagonism against 11 fungal phytopathogens. Environmental concerns and rapidly decreasing phosphorus (P) resources caused a renewed interest in improving soil P tests for a more efficient P fertilization. This led to the development of better P fertilizer recommendation systems for major arable crops and grass. Nevertheless, these P fertilizer recommendation systems seem to fail for intensive vegetable crops, with often a very short growing season and limited rooting system. This leads to low P use efficiencies in the horticultural sector. In order to address this problem we set up a study to answer following questions: (1) which soil P test predicts the plant available P content for intensive vegetable crops the best and (2) can new insights, such as combining different soil P tests, improve P fertilizer recommendations for intensive vegetable crops? To this end, bulk samples of 41 soils with very different P status (based on ammonium lactate extractable P) were collected. The plant available P content of these soils was determined using six commonly used soil P tests (P‐CaCl2, P‐water, P‐Olsen, P‐acetate, P‐lactate, and P‐oxalate) and a P fertilizer pot experiment with endive (a very P sensitive vegetable crop) was conducted. Six pots of each soil were planted with endive. Three of these pots received no P fertilization (0P) and three pots received ammonium polyphosphate equivalent to 24 kg P ha?1 (24P). All other factors were kept constant. Relative crop yield of the 0P fertilized plants compared to the 24P fertilized plants was determined. Plotting these relative yields against the P status of the soil per soil P test allowed to fit a Mitscherlich curve through the data. Also the combination of two different soil P tests to predict the relative yield with a Mitscherlich equation was evaluated. The coefficients of variation of the soil P tests, the R2 values and the relative standard errors of the parameter estimates revealed that P‐acetate and P‐water predicted the relative yield of the 0P plants the best and that combining two different soil P tests gave no extra predictive power. This finding may form the basis for the development of a new P fertilizer recommendation system for intensive vegetable crops, leading to an improved P use efficiency in horticulture. In order to develop this new system more data relating soil P test values with RY of intensive vegetable crops should be collected. 相似文献
