Efforts to restore productivity of pastures often employ agricultural management regimes involving either tillage or no-tillage options combined with various combinations of fertilizer application, herbicide use and the planting of a cash crop prior to the planting of forage grasses. Here we report on the emissions of CO2, N2O and NO from the initial phases (first 6 months) of three treatments in central Rondônia. The treatments were (1) control; (2) conventional tillage followed by planting of forage grass (Brachiaria brizantha) and fertilizer additions; (3) no-tillage/herbicide treatment followed by two plantings, the first being a cash crop of rice followed by forage grass. In treatment 3, the rice was fertilized. Relative to the control, tillage increased CO2 emission by 37% over the first 2 months, while the no-tillage/herbicide regime decreased CO2 emissions by 7% over the same period. The cumulative N2O emissions over the first 2 months from the tillage regime (0.94 kg N ha–1) were much higher than the N2O releases from either the no-tillage/herbicide regime (0.64 kg N ha–1) or the control treatment (0.04 kg N ha–1). The highest levels of N2O fluxes from both management regimes were observed following N fertilizations. The cumulative NO releases over the first 2 months were largest in the tillage treatment (0.98 kg N ha–1), intermediate in the no-tillage treatment (0.72 kg N ha–1), and smallest in the control treatment (0.12 kg N ha–1). For the first week following fertilization the percentage of fertilizer N lost as N2O plus NO was 1.0% for the tillage treatment and 3.0% for the no-tillage treatment.  相似文献

Genome-sequencing studies indicate that all humans carry many genetic variants predicted to cause loss of function (LoF) of protein-coding genes, suggesting unexpected redundancy in the human genome. Here we apply stringent filters to 2951 putative LoF variants obtained from 185 human genomes to determine their true prevalence and properties. We estimate that human genomes typically contain ~100 genuine LoF variants with ~20 genes completely inactivated. We identify rare and likely deleterious LoF alleles, including 26 known and 21 predicted severe disease-causing variants, as well as common LoF variants in nonessential genes. We describe functional and evolutionary differences between LoF-tolerant and recessive disease genes and a method for using these differences to prioritize candidate genes found in clinical sequencing studies.  相似文献

Objective

To assess the reaction of client-owned dogs to intravenous (IV) catheter placement after applying a local anaesthetic (EMLA) or placebo cream for either 30 or 60 minutes.

Study design

Prospective, randomized, blinded, placebo-controlled, clinical trial.

Animals

A total of 202 client-owned dogs of various breeds.

Methods

With owner consent, dogs were randomly allocated to one of four treatment groups: EMLA 60 minutes, EMLA 30 minutes, Placebo 60 minutes and Placebo 30 minutes. After the cream was applied for the allocated time, an IV catheter was placed and the behavioural reaction of the dog was scored. The reaction score was analysed using a Kruskal–Wallis test followed by Mann–Whitney U tests of the multiple pairwise comparisons, with Bonferroni correction.

Results

A large number of dogs, even in the placebo groups, did not react to IV catheter placement. However, the Kruskal–Wallis test showed an overall difference between treatment groups (χ2 = 11.029, df = 3, p = 0.012). The pairwise comparisons showed a lower overall reaction score in the EMLA 60 group than in the EMLA 30 and Placebo 60 groups (adjusted p = 0.018 and adjusted p = 0.044, respectively).

Conclusions and clinical relevance

This study shows that EMLA cream applied for 60 minutes reduces the behavioural reaction of dogs to IV catheter placement; therefore, this intervention can be advocated for routine use in veterinary medicine to enhance the welfare of dogs undergoing IV catheter placement.  相似文献

The RAPD-PCR technique has been used to generate DNA amplification profiles from a set of advanced generation inbred lines of white clover ( Trifolium repens L.). The genetic distances between lines have been estimated and have been compared with the extent of positive heterosis for dry matter yield in hybrid crosses. The analysis clearly separated the four major groups (H, J, R and S), with little within group variation. Heterosis, previously shown to be positively correlated with the extent of phenotypic variation between parental lines, was greatest between lines from groups showing the least genetic distance. The lack of correlation between heterosis and genetic divergence is discussed in the context of results from other systems. Additionally, the data allow a comparison of the extent of phenotypic variation for a trait under selective pressure with the degree of variation at the DNA level detected with the RAPD technique. The aim of this study was to compare blood copy, haematological and glucose values between cats experimentally infected with either Mycoplasma haemofelis (Group HF: 10 cats), ‘Candidatus M. haemominutum’ (Group HM: 3 cats) or ‘Candidatus M. turicensis’ (Group TU: 3 cats). Blood samples were collected regularly up to 85 days post-infection (DPI) for haemoplasma real-time quantitative PCR, haematology, Coombs’ testing and blood glucose measurement. Statistical analysis was performed using a general linear model (ANOVA) appropriate for a repeated measures experiment with significance set as P < 0.05. Cats in Group TU had significantly lower blood copy numbers than cats in Group HF (P < 0.001) and HM (P < 0.001). All Group HF cats developed anaemia (often severe), macrocytosis and evidence of erythrocyte-bound antibodies whereas Groups HM and TU cats did not. Group HF had significantly lower PCVs, haemoglobin concentrations and red blood cell counts, and significantly higher mean cell volumes, than Groups HM and TU. In Group HF, erythrocyte-bound antibodies reactive at 4 °C (both IgM and IgG) appeared between 8 and 22 DPI and persisted for two to four weeks, whereas those reactive at 37 °C (primarily IgG) appeared between 22 and 29 DPI and persisted for one to five weeks. In most cats antibodies appeared after the fall in haemoglobin started. Although Group TU had significantly lower glucose concentrations than Groups HF (P = 0.006) and HM (P = 0.027), mean blood glucose concentrations remained within the reference range in all groups. This study demonstrates that M. haemofelis infection, in contrast to ‘Candidatus M. haemominutum’ and ‘Candidatus M. turicensis’ infection, can result in a severe macrocytic anaemia and the development of cold and warm reactive erythrocyte-bound antibodies.  相似文献

Flexible tubular structures fabricated from solution electrospun fibers are finding increasing use in tissue engineering applications. However it is difficult to control the deposition of fibers due to the chaotic nature of the solution electrospinning jet. By using non-conductive polymer melts instead of polymer solutions the path and collection of the fiber becomes predictable. In this work we demonstrate the melt electrospinning of polycaprolactone in a direct writing mode onto a rotating cylinder. This allows the design and fabrication of tubes using 20 μm diameter fibers with controllable micropatterns and mechanical properties. A key design parameter is the fiber winding angle, where it allows control over scaffold pore morphology (e.g. size, shape, number and porosity). Furthermore, the establishment of a finite element model as a predictive design tool is validated against mechanical testing results of melt electrospun tubes to show that a lesser winding angle provides improved mechanical response to uniaxial tension and compression. In addition, we show that melt electrospun tubes support the growth of three different cell types in vitro and are therefore promising scaffolds for tissue engineering applications.  相似文献
