ABSTRACTThe aim of this study was to evaluate the defoliation patterns of individual tillers, efficiency of herbage utilisation, and forage production in continuously stocked Brachiaria humidicola cv. Comum swards during periods of the year of restricted pasture growth. The experiment was conducted at the Embrapa, Campo Grande-MS, Brazil, from April to October 2012. Treatments consisted of two grazing management strategies, defined by two ranges of sward height (10–15 and 20–25 cm) managed under continuous and variable stocking rates. The experiment was conducted as a completely randomised block design with three replications. During periods of resource constraints, grazing management strategies based on ‘steady state’ sward heights did not alter defoliation patterns, herbage utilisation efficiency (HUE), and forage production in Brachiaria humidicola swards under a continuous stocking method. A direct implication of our results is that grazing management strategies during the driest and coldest periods of the year in the tropics should be developed based on the conditions most suitable for better promoting faster pasture recovery and a return to production in the following spring; it is unlikely that any grazing management strategy would be successful in increasing herbage production and/or HUE in periods of restricted pasture growth. 相似文献

ABSTRACTThis study aimed to assess the physiological and biochemical responses of cotton plants to manganese (Mn2+) nutrition. Four cotton genotypes (G1 – TMG 47; G2 – FM 975 WS; G3 – TMG 11 WS and G4 – IMA 8405 GLT) were grown in nutrient solution under two Mn2+ concentrations (2 and 200 µmol L?1) for 10 days. No visible symptoms of Mn2+ toxicity were observed in the genotypes tested. All genotypes showed a marked increase in leaf chlorophylls, pheophytins, carotenoids, sucrose and total sugars concentration in response to high Mn2+ in a nutrient solution. However, the net photosynthetic rate, stomatal conductance, internal carbon dioxide concentration and transpiration decreased in genotypes G1 and G2 growing under 200 µmol L?1. Antioxidant enzymes such as superoxide dismutase (SOD), ascorbate peroxidase (APX) and glutathione reductase (GR) activities increased in genotypes G1, G3 and G4. Cotton genotypes showed an increased leaf antioxidant and sugar metabolism as a possible strategy to mitigate oxidative stress. The decrease in the net photosynthetic rate and stomatal conductance; the increased antioxidant enzymes activities (SOD, APX and GR); and the increase in leaf sucrose and total sugar concentration were the main physiological and biochemical responses in cotton plants to Mn2+ stress. 相似文献

Scanning electron microscopy (SEM) and chemical analysis were used to observe the cell wall changes that occur in cork with "mancha amarela", when compared to a standard cork. To mimic the microbial attack exhibited in cork with mancha amarela, the standard cork was treated enzymatically with commercial pectinase and hemicellulase preparations. The tissues treated with pectinase were comparable with those attacked with mancha amarela. Both were composed by deformed and wrinkly cells and exhibited cell wall separation at the middle lamella level, which suggests solubilization/removal of the pectic polysaccharides. The cork cell wall material, prepared as alcohol-insoluble residue, was fractionated by hot water (Pect(H)()2(O)) and hot dilute acid (Pect(acid)). The relatively large amount of hexuronic acid and the occurrence of Ara in the SPect(H)()2(O) and SPect(acid) allow to confirm, as far as we know, for the first time the presence of pectic polysaccharides in the cell walls of cork from Quercus suber L. They accounted for ca. 1.5% of the cork and may consist of polymers with long side chains of arabinosyl residues. These polymers have to be taken into account in any realistic model of the cork cell wall. Cork with mancha amarela contained a smaller amount of pectic polysaccharides (ca. 0.5%), which confirms that the cellular separation observed by SEM is related to the degradation/removal of the middle lamella pectic polysaccharides. 相似文献

A fragment (called frg#11, amino acids, aa 56-110) of the protein G (pG) of viral haemorrhagic septicaemia virus (VHSV) was designed after previous results showed it to be recognised by approximately 40% of the trout immunised to VHSV [Dis. Aquat. Organ. 34 (1999) 167]. frg#11 was then cloned, expressed, purified and used to study the production of antibodies to its epitopes in trout immunised to VHSV. Anti-frg#11 trout antibodies could be detected in serum from individual trout surviving VHSV exposure, immunised by injection with purified VHSV or DNA-immunised with its pG gene whereas it was not detected in non-infected and non-immunised trout. The trout serum antibodies which reacted more strongly by ELISA using solid-phase frg#11 (continuous or linear epitopes on the sequence of the pG) had the lowest VHSV-neutralising activity (epitopes which are pG conformation-dependent). Because antibodies recognising continuous as well as conformation-dependent epitopes of the pG seem to be involved in protective trout immunological responses to VHSV, the estimation of anti-frg#11 antibodies could help to the dissection of the complex trout antibody response to VHSV infections. In addition, these preliminary results suggest that the determination of anti-frg#11 antibodies might also be used to complement in vitro viral neutralising assays which seem to be restricted to pG conformation-dependent epitopes. 相似文献

Weaned wether goats (n = 144) approximately 6 months of age were placed in a 2 x 3 factorial design experiment for 5 months to test the main effects and interaction of two levels of nutrition (growth + maintenance, NUT1; twice growth + maintenance, NUT2) and three levels of Haemonchus contortus burden (0, 500 and 2000 larvae administered every 2 weeks: W0, W500 W2000, respectively) on weight, feed intake, level of infection and packed cell volume (PCV). The rationale for the experimental design was based on the lack of information concerning the interaction between nutritional status and worm burden. Results indicated significant effects of worm burden levels on PCV, faecal egg contents (eggs per gram of feces (EPG)), actual worm numbers, feed intake and efficiency of feed utilization. Nutrition x worm burden interactions were also significant for PCV and EPG. However, the differences detected for PCV and actual worm numbers did not translate into large or consistent differences in body weight. Goats on NUT2, after an initial period, showed little difference in body weight, irrespective of worm burden. Within the NUT1 level, W0 kids weighed more than W500 or W2000 kids throughout the study. Although not statistically significant, this constitutes a trend towards an interaction between nutrition and worm burden. In both nutrition levels, there were no body weight differences between W500 and W2000 until the last 14 days. Feed intake was depressed in the first 3 months of the experiment for infected animals, but was subsequently followed by a compensatory reaction. Lower establishment rates, based on actual worm counts, were observed for the higher infection level, but in both infection levels establishment rates tended to decrease with time. Nutrition was found to be more important to counteract the consequences of a parasitic infection than to counteract the establishment of that same infection. 相似文献

The karyotype, the histological structure of the testis and the synaptonemal complex (SC) of the mammalian species Tayassu tajacu , Tayassu pecari and of an interspecific male hybrid captured in nature were analysed. The specimens of T. tajacu (2n=30) and T. pecari (2n=26) exhibited seminiferous tubules with germ cells in all spermatogenesis stages. In the SC studies both species had a regular structure, easily identified in the autosomes and in the sex chromosomes. The hybrid (2n=28) had seminiferous tubules with almost all germinal cells in the spermatogonium stage and only a few cells in the primary spermatocyte stage. Gross abnormalities in SC were observed. A few lateral elements showed regular or partially regular synapsis, other lateral elements were synapsed as multivalents, and most axial elements remained unsynapsed. The results suggest that the karyotypes of the parental species have sufficient differentiation to avoid chromosome synapsis. Alternatively, the hypothesis of the existence of genetic incompatibilities between the parental species is discussed. 相似文献

This study proposes to evaluate rapid methodologies to estimate oxidative stability and biodegradability for mineral, vegetable and synthetic oil samples, using quick and simple experiments. The synthetic oil sample was obtained from castor oil, so the vegetable oil chosen for this evaluation was a sample of the crude castor oil source. The oxidative stability tests described enabled the comparison of synthetic lubricant samples and showed that their stability was lower than that of petroleum-based oil. However, the physico-chemical properties, such pour points and viscosity index, were improved and potentially interesting for lubricant applications. The biodegradability experiments were carried out using a model of bio-kinetics. These studies proved that synthetic lubricant samples were easily degradable (similar to crude castor oil) and showed half-life significantly lower than those of the mineral oil samples. 相似文献

Genotoxic and antigenotoxic effects of acerola fruit at two stages of ripeness were investigated using mice blood cells. The results show that no ripeness stage of acerola extracts presented any genotoxic potential to damage DNA (Comet assay) or cytotoxicity (MTT assay). When antigenotoxic activity was analyzed, unripe fruit presented higher DNA protection than ripe fruit (red color) extract. The antioxidant capacity of substances also showed that unripe samples inhibit the free radical DPPH more significantly than the ripe ones. The results about determination of compounds made using HPLC showed that unripe acerola presents higher levels of vitamin C as compared to ripe acerola. Thus, vitamin C and the complex mixture of nutrients of Malpighia glabra L., and especially its ripeness stages, influenced the interaction of the fruit extract with the DNA. Acerola is usually consumed when ripe (red fruit), although it is the green fruit (unripe) that has higher potential as beneficial to DNA, protecting it against oxidative stress. 相似文献
