A liquid chromatographic method for determining glyphosate (GLYPH) and its major metabolite aminomethylphosphonic acid (AMPA) in various environmental substrates is described. Ion-exchange column chromatography is coupled with post-column ninhydrin derivatization and absorbance detection at 570 nm. Use of a valve-switching technique allowed quantitation of both analytes in a single chromatographic run and eliminated slow-eluting, coextracted interferences. The method was successfully used to quantitate GLYPH and AMPA in organic and mineral soils, stream sediments, and foliage of 2 hardwood brush species. Mean recovery efficiencies for GLYPH as determined from fortified blank field samples were as follows: bottom sediment 84%, suspended sediment 66%, organic soils 79%, mineral soils 73%, alder leaf litter 81%, salmonberry leaf litter 84%, and artificial deposit collectors 87%. Precision for GLYPH determination was good with less than 14% coefficient of variation on mean recovery for all substrates. Limits of detection were lowest for sediments (0.01 microgram/g dry mass) and highest for foliage substrates (0.10 microgram/g dry mass). Anthocyanins and phenolics of 10 blue honeysuckle (Lonicera caerulea L.) genotypes were characterized and quantified by HPLC-DAD. Peak assignments were confirmed by low-resolution electrospray mass spectrometry. Six anthocyanins were detected with the major peak identified as cyanidin 3-glucoside. Five additional anthocyanins were characterized as cyanidin 3,5-diglucoside, cyanidin 3-rutinoside, pelargonidin 3-glucoside, peonidin 3-glucoside, and peonidin 3-rutinoside. Four polyphenolics were identified as chlorogenic acid, neochlorogenic acid, quercetin 3-rutinoside, and quercetin 3-glucoside. Two additional unidentified phenolics were characterized as flavonol and hydroxycinnamic derivatives based on UV-vis spectra. Hydroxycinnamate levels ranged from 30.4 to 156.2 mg/100 g, whereas the flavonol content ranged from 12.6 to 32.8 mg/100 g. The L. caerulea subspecies boczkarnikovae contained the highest amounts of hydroxycinnamic derivatives and flavonols. 相似文献

OBJECTIVES: We describe the methods used to develop and score a 17-item 'screener' designed to estimate intake of fruit and vegetables, percentage energy from fat and fibre. The ability of this screener and a food-frequency questionnaire (FFQ) to measure these exposures is evaluated. DESIGN: Using US national food consumption data, stepwise multiple regression was used to identify the foods to be included on the instrument; multiple regression analysis was used to develop scoring algorithms. The performance of the screener was evaluated in three different studies. Estimates of intakes measured by the screener and the FFQ were compared with true usual intake based on a measurement error model. SETTING: US adult population. SUBJECTS: For development of instrument, n=9323 adults. For testing of instrument, adult men and women in three studies completing multiple 24-hour dietary recalls, FFQ and screeners, n=484, 462 and 416, respectively. RESULTS: Median recalled intakes for examined exposures were generally estimated closely by the screener. In the various validation studies, the correlations between screener estimates and estimated true intake were 0.5-0.8. In general, the performances of the screener and the full FFQ were similar; estimates of attenuation were lower for screeners than for full FFQs. CONCLUSIONS: When coupled with appropriate reference data, the screener approach described may yield useful estimates of intake, for both surveillance and epidemiological purposes. 相似文献

Grassland management aimed at enhancing carbon (C) in soil is an important tool in mitigation of rising atmospheric CO2, yet little is known of how grassland soil C changes with livestock stocking rate (SR). We relate soil organic and inorganic C mass (t ha−1 to 60 cm depth) with cattle stocking over periods of 7–27 year for 32 paddocks distributed across nine community pastures in the mixed-grass prairie of Saskatchewan, Canada. Initial analysis comparing Akaike information criterion models showed that cattle SR explained a greater proportion of variance in soil C, particularly soil organic C, than rainfall. Soil organic C mass increased with cattle SR (R2 = .293; p = .001), even when the latter was normalized to account for differences in vegetation composition and growing conditions among pastures. Normalized SR varied from 0.49 to 2.30 times recommended levels, over which SOC increased from 24.7 to 57.4 t ha−1. Increases in soil organic C under greater stocking coincided with increased abundance of introduced vegetation, particularly the rhizomatous grass Poa pratensis. Inorganic soil C accounted for 34.6% of total soil C, being particularly large below 30 cm soil depth, but did not vary with stocking rate. These findings indicate that both organic and inorganic C are important pools of C in northern temperate grassland soils, with soil organic C positively associated with long-term cattle SR. Further studies are recommended to understand more fully the mechanisms regulating grazing impacts on soil C mass in northern temperate grasslands. 相似文献

The purpose of the present work was to examine whether partial acid hydrolysis (PAH) of a high‐amylose maize starch (ae‐VII) would enhance the effects of hydrothermal treatments to produce granular resistant starch (RS) that is stable to further heat treatment at atmospheric pressure. PAH ae‐VII starches were prepared by heating 35% (w/v) suspensions with 1% (w/w) HCl at 25°C for 6, 30, and 78 hr. Native and PAH starches were then treated by annealing (ANN) or heat‐moisture treatment (HMT). ANN was done at 70% moisture at 50, 60, or 70°C for 24 hr, and HMT was done at 30% moisture at 100, 120, or 140°C for 80 min. RS that survives boiling during analysis was determined by a modification of the AOAC method for determining total dietary fiber. RS was also determined by the Englyst method. Little change in the gelatinization enthalpy was found for ae‐VII starch after PAH, ANN, or HMT as individual treatments. After PAH, either ANN or HMT led to decreased gelatinization enthalpy. HMT and ANN alone increased boiling‐stable RS but decreased total RS. After PAH of ae‐VII, either ANN or HMT tended to increase the yield of boiling‐stable granular RS, with the greatest yield (≤63.2%) observed for HMT. 相似文献

OBJECTIVE: To determine the reliability and validity of scales measuring low-fat milk consumption self-efficacy and norms during school lunch among a cohort of 5th graders. DESIGN: Two hundred and seventy-five students completed lunch food records and a psychosocial questionnaire measuring self-efficacy and norms for school lunch low-fat milk consumption during the fall and spring semesters of the 1998-1999 academic year. Test-retest reliability was assessed in participants who also completed the questionnaire in the spring semester (n = 262). Principal component analyses identified and confirmatory factor analyses confirmed latent variables. Bivariate correlations measured construct validity. SETTING: Houston-area middle school. SUBJECTS: Fifth graders (n = 275) from one middle school in southeast Texas. RESULTS: Two scales measuring psychosocial influences of low-fat milk consumption were identified and proved reliable in this population: milk self-efficacy and milk norms. Milk self-efficacy and norms were positively correlated with milk consumption and negatively correlated with consumption of sweetened beverages. CONCLUSIONS: These questionnaires can be used in similar interventions to measure the impact of self-efficacy and norms for drinking low-fat milk during school lunch. 相似文献

分析了辐照牛肉中辐解产物2-十二烷基环丁酮在不同的吸收剂量(0.5~10.0kGy)、贮藏时间(0-40d)及贮藏温度(-20℃~4℃)下的变化规律。采用硅胶层析柱-气相色谱/质谱联用技术萃取分离并检测。结果表明,2-十二烷基环丁酮的浓度与吸收剂量呈线性正相关,随着贮藏时间延长其含量减少,贮藏温度对其没有显著影响。利用2-十二烷基环丁酮可鉴定牛肉是否经过辐照,在已知贮藏温度和时间的条件下,可评估辐照牛肉的大约吸收剂量范围。 相似文献
