Flavan-3-ol monomers and oligomers from dimers to decamers were quantitatively determined by HPLC-FLD in leaves of four Theobroma cacao varieties. Cacao flush leaves had higher procyanidin content than mature leaves. Moniliophthora perniciosa basidiospore infection results in an increase in cacao leaf procyanidins in the inoculated leaves, as well as in the uninoculated, adjacent, older leaves on an actively growing branch, resulting in both a local and systemic increase in procyanidin content in response to infection. Procyanidin oligomers increased significantly in leaves of the less susceptible genotype Scavina 6 upon infection, and total procyanidin levels were higher than in Cacao Comum a more disease susceptible genotype. Selection of genotypes with high constitutive and/or inducible content of procyanidins in young flush leaves may be a strategy to develop witches’ broom disease resistant cultivars. 相似文献

Tetranychus urticae Koch has recently developed resistance to chlorfenapyr in Australia and Japan, but no attempt has yet been made to describe the biochemical mechanisms involved in chlorfenapyr resistance. In this study a laboratory-selected chlorfenapyr-resistant strain was investigated. Resistance to chlorfenapyr was associated with a strong increase in esterase activity and P450 mono-oxygenase (MO) activity but a decrease in 3,3',5,5'-tetramethylbenzidine (TMBZ) peroxidation activity. Differences in esterase activities between susceptible and resistant strains increased with increasing carbon number of the aliphatic side-chain of the nitrophenol substrate. A 4.4-fold increase in the O-deethylation of 7-ethoxy-4-trifluoromethyl coumarin (7-EFC) mediated by P450 MOs was detected. Remarkably, the resistant strain showed only half of the total TMBZ peroxidation activity found in the susceptible strain. The activity of these enzymes was further determined on different crosses and back-crosses of both strains. Results indicated that activities correlated with chlorfenapyr susceptibility and could be considered as biochemical markers. Esterase isozymes of both strains and their crosses were separated with isoelectric focusing (IEF) and visualised after activity staining. It was clear that two distinct zones of enhanced esterase activity were present in the chlorfenapyr-resistant strain (EST 11, pI = 4.88 and EST 16, pI = 4.71). EST 11 was identified with inhibitors as a carboxylesterase. The relative presence and intensity of these esterase zones changed in the different crosses and could be seen as a marker for chlorfenapyr resistance. Glutathione-S-transferase and glucose-6-phosphate dehydrogenase activities were not significantly different between strains. A twofold decrease in TMBZ peroxidase activity in the resistant strain could reflect decreased activation of chlorfenapyr. On the basis of these results the involvement of P450 MOs and esterases in the activation and detoxification of chlorfenapyr in T. urticae is challenged and discussed. 相似文献

Fusarium culmorum is a phytopathogenic, toxigenic fungus causing seedling diseases, foot rot and head blight of cereals. For estimating competition effects in mixtures of two single-spore isolates, two winter rye single crosses were tested with either four isolates individually or four 1 : 1 mixtures of the same isolates in six field environments. Two isolates (FC46, FC64) were highly aggressive deoxynivalenol (DON) and 3-acetyl DON-producers, the other two (FC30, FC71) were medium aggressive nivalenol-producers. Rye heads were inoculated during flowering with conidia of pairs of isolates expressing similar (FC46 + FC64, FC30 + FC71) or contrary (FC46 + FC71, FC30 + FC64) levels of aggressiveness and similar or different concentrations and chemotypes of mycotoxins, respectively. Head blight rating and yield components relative to the non-inoculated plots were recorded as aggressiveness traits. Additionally, mycotoxin concentrations were measured in the rye grain. Random pathogen samples were re-isolated from heads at the onset of symptom development and analysed by molecular markers (RAPD–PCR) in one environment. Aggressiveness of the isolate mixtures was significantly lower than that of the isolates applied individually on both rye genotypes. Similarly, mycotoxin concentrations were significantly lower in the mixtures in seven out of eleven comparisons. Among the re-isolates, the component genotypes of a mixture significantly deviated from the inoculated 1 : 1 ratio when a particular isolate (FC46) was present in the mixture. This isolate displayed a superior competitive ability irrespective of the aggressiveness or mycotoxin profile of the mixing partner illustrating that pathogenic fitness is caused by additional factors that have not, as yet, been identified. 相似文献

BACKROUND: In Tetranychus urticae Koch, acetylcholinesterase insensitivity is often involved in organophosphate (OP) and carbamate (CARB) resistance. By combining toxicological, biochemical and molecular data from three reference laboratory and three OP selected strains (OP strains), the AChE1 mutations associated with resistance in T. urticae were characterised. RESULTS: The resistance ratios of the OP strains varied from 9 to 43 for pirimiphos‐methyl, from 78 to 586 for chlorpyrifos, from 8 to 333 for methomyl and from 137 to 4164 for dimethoate. The insecticide concentration needed to inhibit 50% of the AChE1 activity was, in the OP strains, at least 2.7, 55, 58 and 31 times higher for the OP pirimiphos‐methyl, chlorpyrifos oxon, paraoxon and omethoate respectively, and 87 times higher for the CARB carbaryl. By comparing the AChE1 sequence, four amino acid substitutions were detected in the OP strains: (1) F331W (Torpedo numbering) in all the three OP strains; (2) T280A found in the three OP strains but not in all clones; (3) G328A, found in two OP strains; (4) A201S found in only one OP strain. CONCLUSIONS: Four AChE1 mutations were found in resistant strains of T. urticae, and three of them, F331W, G328A and A201S, are possibly involved in resistance to OP and CARB insecticides. Among them, F331W is probably the most important and the most common in T. urticae. It can be easily detected by the diagnostic PCR‐RLFP assay developed in this study. Natural products (NPs) have long been used as pesticides and have broadly served as a source of inspiration for a great many commercial synthetic organic fungicides, herbicides and insecticides that are in the market today. In light of the continuing need for new tools to address an ever‐changing array of fungal, weed and insect pests, NPs continue to be a source of models and templates for the development of new pest control agents. Interestingly, an examination of the literature suggests that NP models exist for many of the pest control agents that were discovered by other means, suggesting that, had circumstances been different, these NPs could have served as inspiration for the discovery of a great many more of today's pest control agents. Here, an attempt is made to answer questions regarding the existence of an NP model for existing classes of pesticides and what is needed for the discovery of new NPs and NP models for pest control agents. © 2014 Society of Chemical Industry 相似文献

There is little information known about the energy requirements of cats in temperature climates. Energy requirement of domestic short‐haired cats was determined using three groups of mixed gender – old kept outside (approximately 9.9 years of age; 4.8 kg; n = 9), young kept outside (approximately 3.1 years of age; 3.9 kg; n = 8) or young kept inside (approximately 3.1 years of age; 3.9 kg; n = 8). Cats were housed individually for 5 weeks during summer (18.5 ± 0.5 °C) and winter (8.5 ± 0.4 °C) and were fed a commercially available maintenance diet ad libitum. In both periods, energy expenditure was determined from the rates of 2H and 18O elimination for blood H2O over a 12 day period, from a doubly labelled water bolus 2H2O (0.7 g/kg BW) and H218O (0.13 g/kg BW) administered intravenously. During the summer period, macronutrient digestibility was determined. Older cats had a reduction (p < 0.05) in apparent digestibility of dry matter (approximately 9%), energy (approximately 8%) and protein (6%). There was a significant effect of age and season on energy intake and energy expenditure. While lean mass was affected by age and season, there was no effect of age or season on energy expenditure when expressed as a proportion of lean mass. Possible seasonal differences in nutrient digestibility may explain these results. 相似文献

Regions of rainfall gradients at desert margins are areas where eco-geomorphic changes and degradation in response to global warming and grazing are most intensive. Assessment and understanding of the consequences of global warming and human disturbance of these vulnerable ecosystems requires detailed information regarding the spatial variation of soil and vegetation properties and their dependence on rainfall. Such detailed information is still of limited availability. This study aimed to rectify this shortcoming by analyzing data on the chemical and spectral properties of soil samples collected along the Judean Desert climatic gradient. Six clusters of typical chemical soil properties were identified. Dependencies of clusters’ chemical properties (total iron, organic and inorganic carbon) on annual rainfall amounts were found to be significant. Two spectral Landsat TM band ratios introduced to assess soil conditions showed significant correlations with these chemical properties. These ratios were found to be higher correlated than individual chemical properties with average rainfall suggesting that spectral data may represent a broader range of chemical, physical, and biological site properties. Examining the three-way relationships between soil chemical properties, their spectral reflectance characteristics and rainfall, facilitates a better understanding of the generalized trends along the climatic gradient and more specifically of modes of transition from semi-arid to arid zones. 相似文献
