Structure and ultrastructure of the celiac-mesenteric ganglion complex in the domestic dog (Canis familiaris)

A number of neurons of the autonomic nervous system are situated in the ganglia and can be systematically divided into pre-vertebrals, paravertebrals, intramural and para-viscerals. The celiac-mesenteric ganglion, an important pre-vertebral ganglion, is located together with the abdominal aorta and links the central nervous system to the peripheral system, participating in the coordination of peripheral reflexes and principally innervating the stomach, intestines, accessory glands (liver and pancreas). In addition, the celiac-mesenteric ganglion also contributes to the innervation of the spleen and has a role in gastrointestinal motility control. This study examined the structural and ultrastructural aspects of 40 celiac-mesenteric ganglia from domestic dogs. For light microscopy ganglia were included in paraplast and stained with haematoxylin-eosin, picrosirius, toluidine blue, Calleja's and Masson's trichrome. For examination by electron microscopy, the ganglia were submitted to cryofracture, enzyme digestion, hydrolysis and fixed in 5% glutaraldehyde in 0.1 M phosphate buffer (pH 7.4). The celiac-mesenteric ganglion was observed as a ganglionic complex composed of various ganglionic units separated by types I and III collagen fibres, predominantly unmyelinated nerve fibres and continuous capillaries. This complex is surrounded by a double-layer capsule (internal and external). The principal ganglion cells had eccentric nuclei with two nucleoli, the nucleolemma was double and presented nuclear pores. In the cytoplasm there were vesicles of the Golgi apparatus, electron-dense vacuoles, mitochondrias, smooth and granulated endoplasmic reticulum and free ribosomes. In conclusion, this ganglionic complex, in contrast to similar structures in the enteric nervous system, presents separate ganglionic units in a systematic arrangement related to the extrinsic and specific innervation of the target organs.     

Seasonal dynamics of Anocentor nitens on horses in Brazil

Anocentor nitens is a widespread ectoparasite of horses in Brazil. A study of the seasonal dynamics of the parasitic phase of this tick was made by regular census of engorging females (>/=4mm) on free-ranging horses, fortnightly from July 1995 to July 1997, at Pedro Leopoldo, Minas Gerais. Twenty horses were divided into four groups of five animals per group, each having a similar mix of resistant and less resistant animals, based on initial infestation. Every 2 weeks, one of the groups, in sequence, was treated with an acaricide to prevent excessive infestations, and female ticks were censused 38 days later. The acaricide dips did not interfere with the seasonal dynamics of the tick population, which produced three major peaks of activity over a 12-month period. The first two peaks were in the hot, wet season and the third in the cold dry season. Tick numbers were similar for the two 12-month periods, decreasing from the spring and summer to the autumn and winter. Infestation rates were not equal among different horses. Twenty percent of the horses were responsible for 48% of the total parasite load, while 30% accounted for only 2.5% of the total parasite load. Principal predilection sites were; the ear (61%), mane (21%), and muzzle and face (6%). Measures for the strategic control of A. nitens ticks are discussed.     

Development of an ELISA system for detection of anti-Anaplasma marginale antibodies in cattle in Brazil

An ELISA test was developed for detecting antibodies against Anaplasma marginale in bovine sera. Four antigenic preparations were produced from infected red blood cells. Some aliquots of this preparation were stored at -70 degrees C with 30% DMSO in phosphate-buffered saline (PBS) and others were lysed with 0.9% NH4Cl and stored at -20 degrees C. Typical anaplasmal structures were seen by electron microscopy in the antigenic preparations containing the erythrocytes that had been stored with DMSO. The performance of the ELISA test was evaluated by testing 298 positive serum samples collected from immunized cattle, 39 negative serum samples collected from cattle imported from areas free of A. marginale and 50 samples collected from cattle naturally infected in the field. The test gave a specificity of 94.87% and a sensitivity of 100%.     

Geographic variation in marine turtle fibropapillomatosis.

We document three examples of fibropapillomatosis by histology, quantitative polymerase chain reaction (qPCR), and sequence analysis from three different geographic areas. Tumors compatible in morphology with fibropapillomatosis were seen in green turtles from Puerto Rico and San Diego (California) and in a hybrid loggerhead/ hawksbill turtle from Florida Bay (Florida). Tumors were confirmed as fibropapillomas on histology, although severity of disease varied between cases. Polymerase chain reaction (PCR) analyses revealed infection with the fibropapilloma-associated turtle herpesvirus (FPTHV) in all cases, albeit at highly variable copy numbers per cell. Alignment of a portion of the polymerase gene from each fibropapilloma-associated turtle herpesvirus isolate demonstrated geographic variation in sequence. These cases illustrate geographic variation in both the pathology and the virology of fibropapillomatosis.     

Quantification of cysteine S-conjugate of 3-sulfanylhexan-1-ol in must and wine of petite arvine vine by stable isotope dilution analysis

Making use of a convenient synthetic approach to prepare the deuterated S-3-(hexan-1-ol)-cysteine by a Michael addition reaction, an analytical method was developed to measure the presence of the cysteine S-conjugate, precursor of 3-sulfanylhexan-1-ol (3-mercaptohexan-1-ol), in must and wine from Petite Arvine vine. The method uses a stable isotope dilution assay with a suitable one-step sample preparation and HPLC-MS detection. The method has limits of detection and quantification of 3 and 10 microg/L, respectively. A correlation between the increase of the precursor concentration and the increase of the degree of rot has been established.     

Analysis of the main components of the aguamiel produced by the maguey-pulquero (Agave mapisaga) throughout the harvest period

The main characteristics of the aguamiel (maguey-pulquero sap) during the harvest period of the Agave mapisaga plants were assessed to establish its stability through time and the industrial potential of its components. Only minor differences in aguamiel composition were detected among samples collected at different time points of the harvest period. The aguamiel analyzed contained 11.5 wt % of dry matter, which was composed mainly of sugars (75 wt %). Among these sugars, 10 wt % were fructo-oligosaccharides (FOS), which are known to be important in the food industry for their prebiotic properties. Other components include 0.3 wt % of free amino acids (with most essential amino acids and four neurotransmitters: GABA, GLY, GLX, and ASX), 3 wt % of proteins, and 3 wt % of ashes.
