We evaluated the effects of feeding high volumes of milk replacer on growth and reproductive performances in Japanese black heifers. Fifty-one heifers were fed milk replacer at 9 L/day for 60 days (9 L × 60 days; n = 18) or 41 days (9 L × 41 days; n = 15), or at 7 L/day for 40 days (7 L × 40 days; n = 18). Artificial insemination (AI) was performed on heifers with ≥270 kg body weight and ≥116 cm body height at 300 days of age. The age at the first AI was 0.35 month later for 7 L × 40 days than the other groups (p < .01). However, age at calving did not differ among treatments (22.1 months). The interval from the first AI to pregnancy tended to be ~2 months longer for the 9 L × 60 days than the other groups (p = .07). Our results showed that feeding high volumes of milk replacer may reduce the age at calving via an improved rate of growth. In addition, we propose that feeding a maximum of 7 L milk replacer for 40 days may be the most appropriate rearing regime because the success of pregnancy per AI may be reduced in calves fed a maximum of 9 L for 41 and 60 days.  相似文献

The Japanese cormorants used in traditional fishing in Japan are wild derived and their sex cannot be determined from their appearance. Applicability of molecular sex determination based on the size difference between CHD1Z and CHD1W introns was confirmed in male and female Japanese cormorants whose sexes had been ascertained by pathological autopsy. All of 21 birds of unknown sex reared by a cormorant fishing master were identified as males. The molecular sexing method will provide valuable information on sex differences of wild Japanese cormorants, including tameness, trainability, behavior and fishing capability, as well as for future trials involving artificial reproduction.  相似文献

ABSTRACT:   Genetic evidence of the occurrence of natural hybridization between female Pinctada fucata and male Pinctada maculata among wild pearl oysters ( n  = 20) collected for use as the mother shell for private pearl farming in the Oshima Strait at Amami-o-shima, Kagoshima Prefecture, Japan, were obtained. A polymerase chain reaction-based species identification method for Pinctada was developed using polymorphisms in the internal transcribed spacer (ITS) region of the nuclear ribosomal RNA (rRNA) gene. This method enabled the amplification of the ITS regions using a primer set specific for P. maculata and P. fucata . However, 10 of 20 individuals morphologically identified as P. fucata had sequences specific to both P. maculata and P. fucata in the ITS region. These putative hybrids showed sequences of a maternally inherited mitochondrial 16S rRNA gene, identical to that of P. fucata . Shells of the putative hybrids were difficult to discriminate from those of P. fucata exhibiting similar taxonomic traits. Moreover, the hybrids exhibited slower growth than P. fucata but faster growth than P. maculata .  相似文献

Summary To improve the efficiency of papaya anther culture, we investigated (1) hormonal medium conditions for inducing haploids or dihaploids; (2) identified the sex of established plantlets using a sex-specific DNA molecular marker and (3) estimated their ploidy by flow cytometry analysis of DNA content. Anthers with a mixture of uninucleate, mitotic, and binucleate microspores were collected from a male plant, and cultured on MS agar medium with different concentrations of CPPU and NAA. An embryo induction rate of 13.8% was attained on MS agar medium with 0.01 mg l−1 CPPU and 0.1 mg l−1 NAA. The induced embryos were subcultured on medium with 0.0025 mg l−1 CPPU. Rooting of the developed shoots was promoted by treating their basal parts with 1500 mg l−1 IBA in a 50% ethanol solution for about 10 seconds. All the embryo-derived plantlets (27 plants) were identified as female, implying that they were derived from microspores. In addition, 26 plants were determined to be triploids and one to be tetraploids. We also observed a wide range of morphological variation (e.g., in tree height and fruit size) among the established plants. Based on the results, we discussed a potential value of anther culture techniques for the breeding of papaya.  相似文献

The study was conducted to clarify how early high plane of nutrition related to metabolic imprinting affected growth, carcass characteristics, and meat quality of grass-fed Wagyu (Japanese Black cattle). Wagyu steers were allocated randomly into 2 dietary groups: (1) steers fed milk replacer (crude protein 26.0%, crude fat 25.5%; maximum intake 0.6 kg/d) until 3 mo of age and then fed roughage (orchard grass hay) ad libitum from 4 to 10 mo of age (roughage group, RG; n = 11); (2) steers fed milk replacer (maximum intake of 1.8 kg/d) until 3 mo of age and then fed a high-concentrate diet from 4 to 10 mo of age (early high nutrition, EHN; n = 12). After 11 mo of age, all steers were fed roughage ad libitum until 31 mo of age and then slaughtered. Growth performance, carcass traits, longissimus muscle (LM) meat quality and intramuscular fat (IMF) content, plasma insulin-like growth factor I (IGF-I) concentration, and bone mineral density were measured. Body weight was greater in EHN steers (571 kg) than RG steers (520 kg; P < 0.01). Plasma IGF-I levels were higher in EHN steers than in RG steers at 3, 10, and 14 mo of age (P < 0.01, P < 0.005, P < 0.001, respectively); however, plasma IGF-I levels were lower in EHN steers compared with RG steers at 30 mo of age (P < 0.01). The total weight of the muscles and bones of the left half of the carcass was not different between the 2 groups (P = 0.065). Five of the 19 muscles investigated (semimembranosus, P = 0.036; infraspinatus, P = 0.024; supraspinatus, P = 0.0019; serratus ventralis cervicis, P = 0.032; serratus ventralis thoracis, P = 0.027) were heavier in EHN steers. Total fat weight in the left half of the carcass was 30% greater (P = 0.025) in HNE carcasses. Subcutaneous and perirenal fat weights were 53% and 84% greater (P = 0.008, P = 0.002, respectively) in EHN carcasses. The LM IMF content was greater in EHN loins (13.2%) compared with RG loins (9.4%) at 31 mo of age (P = 0.038); however, no differences were found for shear force, tenderness, and cook loss. These results suggested early high-nutrition affected the growth and meat quality of livestock.  相似文献

We evaluated the influence of texture information from remote sensed data on the accuracy of forest type classification at different spatial resolutions. We used 4-m spatial resolution imagery to create five different sets of imagery with lower spatial resolutions down to 30 m. We classified forest type using spectral information alone, texture information alone, and spectral and texture information combined at each spatial resolution, and compared the classification accuracy at each resolution. The classification and regression tree method was used for classification. The accuracy of all three tests decreased slightly with lower spatial resolution. The accuracy with the combined data was generally higher than with either the spectral or texture information alone. At most resolutions, the lowest accuracy was with texture information alone. However, there was no clear difference in accuracy between the combined data and spectral data alone at 25- and 30-m spatial resolution. These results indicate that adding texture information to spatial information improves the accuracy of forest type classification from very high resolution (4-m spatial resolution) to medium resolution imagery (20-m spatial resolution), but this accuracy improvement does not appear to hold for relatively coarse resolution imagery (25- to 30-m spatial resolution).  相似文献

Astaxanthin captured peroxynitrite to form nitroastaxanthins. 15-Nitroastaxanthin was a major reaction product of astaxanthin with peroxynitrite. Here, the anti-oxidative, anti-tumor-promoting, and anti-carcinogensis activities of 15-nitroastaxanthin were investigated. In addition to astaxanthin, 15-nitroastaxanthin showed excellent singlet oxygen quenching activity. Furthermore, 15-nitroastaxanthin showed inhibitory effects of in vitro Epstein-Barr virus early antigen activation and two-stage carcinogensis on mouse skin papillomas. These activities were slightly higher than those of astaxanthin. Similar results were obtained for the 15-nitrolutein, a major reaction product of lutein with peroxynitrite.  相似文献

We examined 24 microsatellite markers to select those most suitable for individual identification and parentage tests of brown bears (Ursus arctos) based on statistical parameters and experimental error using liver samples (38 issues) from nuisance bears killed in the Akan–Shiranuka Region, eastern Hokkaido, Japan from 1996 to 2006. We found seven microsatellite markers suitable for identifying individuals (G1A, G10B, G10L, UarMU5, UarMU23, UarMU50, and UarMU51; probability of identity P id = 3.17  × 10−7; probability of identity of sibs P id-sib  = 2.23 × 10−3), 12 microsatellite markers suitable for parentage testing when one parent is known [G1A, G1D, G10B, G10L, G10P, UarMU5, UarMU23, UarMU50, UarMU51, UarMU59, UarMU61, and UarMU64; probability of excluding one parent (PE I = 0.9991)], and 15 microsatellite markers for parentage testing when neither parents are known [three markers added to the above: G10M, G10X, and UarMU9; probability of excluding both parents (PE II = 0.9869)].  相似文献

Summary

We have developed a new garlic growing system from bulbils through transplanting. By this system, extremely small seed bulbils produced bulbs as large as those from cloves of bulbs. In the new system, the bulbils harvested in summer and stored at 18–20°C in winter are grown from the following spring to summer in paper pots, and then transplanted to the field. In Hokkaido, northern Japan, July transplanting produced larger bulbs than August or September transplanting, and the bulbs produced were almost the same size as those of traditional growing methods. One bulbil weighed 75.3±5.6 mg and 84.2±5.4 mg in cv. Furano and a clone, No. 205, respectively, while one clove weighed 5–7g. A clone, H500, produced bulbs of 79.9±3.0.g although its original bulbil weighed only 15.4±0.8.mg. Moreover, this clone produced a great number of bulbils, 250–300, per plant.  相似文献

Chemokines are important regulators of the selective recruitment of inflammatory cells into sites of allergic inflammation. Since canine atopic dermatitis (AD) shares many clinical features of human AD, patterns of chemokine production in dogs may also be similar with those in humans. The aim of this study was to examine mRNA expression of CCL27 and CCL28 in lesional skin of dogs with AD to demonstrate similarity of chemokine production with human counterparts. RNA was extracted from skin biopsy specimens of 12 dogs with AD. The mRNA expression of CC chemokines (CCL4, CCL19, CCL20, CCL21, CCL24, CCL27 and CCL28) was analyzed by quantitative real-time PCR and was compared between lesional and non-lesional skin. Seven types of chemokines examined were constitutively expressed in both lesional and non-lesional skin. It was found that mRNA expression levels of CCL27 and CCL28 among the chemokines were significantly different between lesional and non-lesional skin (P<0.05). Expression level of CCL27 mRNA in lesional skin was significantly lower than that in non-lesional skin. On the other hand, CCL28 mRNA expression in lesional skin was found to be higher than that in non-lesional skin. These results suggest that CCL28 but not CCL27 may play important roles in immunopathogenesis of canine AD, indicating that experimental canine study may provide additional information that can be extrapolated to human AD.  相似文献
