Growth‐related changes in the histochemical properties and collagen architecture of the Musculus pectoralis were compared among Silky, layer‐type and meat‐type cockerels. Histochemical and immunohistochemical methods were used and collagen architecture was studied using scanning electron microscopy. The total amount of collagen present was also measured. The diameter of type IIB myofibers was similar or rather larger in the layer‐type birds compared with the meat‐type. The collagen content was generally low for 5–10 weeks across the breeds and then increased in the other breeds except for Silky. In the perimysium, the collagen bundles gradually increased in size and the density of the fibrils also increased during growth. At 30 weeks of age, the layer‐type birds showed compact collagen bundles while the meat‐type had loose bundles. The endomysial collagen network appeared relatively denser in the meat‐type chicks compared to the others at week 1. At 30 weeks of age, compact and felt‐like structure of endomysium was shown by Silky and layer‐type chickens, while the meat‐type showed a relatively loose arrangement of tissue in the endomysial collagen. From these results, it appears that the meat‐type chicken can produce a large M. pectoralis with many, relatively thinner myofibers and a relatively undeveloped form of intramuscular collagen structure.  相似文献

The common marmoset has been increasingly used for research in the biomedical field; however, there is littleinformation available regarding effective methods of anesthesia in this species. This study retrospectivelyanalyzed 2 regimens of anesthesia induction: intramuscular injection of ketamine followed by inhalation of 5%sevoflurane, and intramuscular injection of midazolam, butorphanol and ketamine followed by inhalation of 5%sevoflurane. Anesthetic depth did not reach the surgical anesthesia stage in 7 out of 99 animals receiving theformer regimen, whereas there were only 2 such animals out of 273 receiving the latter regimen. The latterregimen, when followed by maintenance anesthesia with 3% sevoflurane inhalation, was successfully used invarious nociceptive procedures. These results indicate that the injection of a combination of midazolam,butorphanol and ketamine followed by inhalation of a high concentration of sevoflurane is effective foranesthesia induction in marmosets.  相似文献

Recently, hyperbaric oxygen therapy with a lower pressure and oxygen concentration (L-HBOT) than previous methods has been used for dogs in Japan; however, the influences of L-HBOT on dogs have not been clarified. To verify the influences of L-HBOT on physiological mechanism in dogs, we investigated blood gas parameters, glutathione peroxidase (GPx) activity, heart rate variability, stress-related hormones and skin conductance (SC) in 4 clinically normal beagle dogs with catheters in their carotid arteries and jugular veins when they were quiet, after running, after receiving L-HBOT (30% oxygen concentration, 1.3 atmospheres absolute, 30 min) or after not receiving L-HBOT. The results showed there were no changes in blood gas parameters, heart rate variability and catecholamine levels after L-HBOT. GPx activity was significantly higher, and the SC and cortisol level were lower in dogs that received L-HBOT than those when they were quiet. These results suggested that L-HBOT may have a small influence on oxygenation dynamics, activate antioxidant enzymes such as GPx, restrain autonomic nervous activity and control the balance between oxidation and antioxidation inside the body.  相似文献

Growth‐related changes in the histochemical property and collagen architecture of the iliotibialis lateralis muscle were compared among Silky, layer and meat cockerels. Histochemical and immunohistochemical methods were employed to observe the collagen architecture. The total intramuscular collagen was also determined. The muscle consisted of type IIA, IIB and IIC myofibers, of which type IIB occurred at the highest frequency. The diameter of type IIB myofibers in each week was largest in the layer, followed by the meat, and was smallest in the Silky. The total amount of collagen reached 3.38 mg/g in the meat bird, 3.03 mg/g in the layer and 2.71 mg/g in the Silky by 30 weeks of age, respectively. In the perimysium, the collagen bundles increased in size and density of fibrils with growth. At 30 weeks of age the layer had compact collagen platelets while the Silky had loose collagen bundles. In the meat bird, the collagen bundles were moderately compact. The endomysial collagen network had a large mesh size at 1 week and thereafter accumulated many collagen fibrils to form a felt‐like fabric of fibrils at 30 weeks of age. From these results it appears that growth‐related changes in the iliotibialis lateralis muscle are not necessarily causally affected by the different growth rates of chicken breeds.  相似文献

The egg production industry is facing various problems that need to be solved. For amino acid nutrition to achieve scientific and economical feeding of laying hens, it is necessary to elucidate the content, digestibility, or availability of nutrients of feedstuffs and feeds and the requirement of amino acids for laying hens. In addition, improvement to quality of eggs and meat of spent hens, methods of management and development of new feedstuffs are essential. For sustainable animal production, decrease in excreta and animal welfare should be studied. The real‐time determination of content and digestibility of amino acids in feedstuffs are essential for formation of feeds. Recent advances in the near infrared reflectance analysis will be able to determine the content, digestibility and availability of nutrients in feedstuffs and feeds, if we have a supplemental amount of conventional analysis to define the calibration population. The amino acid requirements are affected by various factors. Therefore the method to quickly and exactly determine amino acid requirements in response to various factors is necessary. By using plasma free amino acid concentration as a criterion, it is possible to determine amino acid requirements in various conditions of laying hens within a short experimental period, repeatedly using the same animals. Because the amino acid requirements differ among individual animals, it should be expressed as grams per hen per day. Practically, it is impossible to formulate various feeds for individual hens. The various expressions have been developed and these expressions have advantages and disadvantages. The nitrogen excretion of laying hens is easily reduced by reducing dietary nitrogen levels and restricting the feed intake. The availability of amino acid may be improved by feeding management, and supplementing enzyme, but the quality of eggs and meat of spent hens and welfare of laying hens are not affected by amino acid nutrition.  相似文献

Histochemical and immunohistochemical methods were employed and collagen architecture was studied using scanning electron microscopy. Total intramuscular collagen was also determined. The myofibers were categorized as type I, type IIA and a transitional form, type I‐tr. The proportion of type I‐tr myofibers diminished and these myofibers were transformed entirely into type I myofibers in meat‐type but incompletely in the others. The largest diameter of type I myofiber was found in layer‐type at 30 weeks of age. At 30 weeks of age, layer‐type birds had attained well‐developed perimysial collagen bundles while meat‐type birds had less developed bundles. The endomysial collagen network had a large mesh size at 1 week and then accumulated many collagen fibrils to form a felt‐like fabric of fibrils by 30 weeks of age. Silky birds developed the thickest endomysial collagen plates of all the breeds. From these results it appears that growth‐related changes in the histological structure of M. puboischiofemoralis pars medialis are not necessarily causally affected by the different growth rates of chicken breeds.  相似文献

Four experiments were conducted to estimate dietary lysine (Lys) requirements using plasma amino acid concentrations as a criterion in mature thoroughbreds. In each experiment four adult thoroughbreds were used. The changes in plasma Lys concentration after feeding were observed in Experiment 1. Blood samples were taken from the cervix vein 0.5 h before, and 1, 3, 5.5, and 10 h after feeding. The plasma Lys concentration increased and remained constant until 3 h after feeding, decreased until 5.5 h and remained constant after then. Therefore, the bleed was done at 3 h after feeding in all later experiments. To make sure of the response speed of the plasma Lys to changes of dietary Lys levels, dietary Lys levels were changed from high to low, and low to high levels in Experiment 2. Blood samples were taken just on the changing day and 1, 2 and 3 days after changing diets. The plasma Lys concentration decreased until 2 days after changing the diet, and then remained constant with advancing days after changing the diet from high Lys to low Lys. On the other hand, the plasma Lys increased until one day after changing the diet, and then remained constant with advancing days after changing dietary Lys levels. Thus, blood samples were taken 3 days after feeding in the next experiments. The possibility of estimating the Lys requirement for maintenance using plasma Lys concentration was elucidated by two methods in Experiments 3 and 4. In Experiment 3, the horses were fed a diet containing 0.33 percent Lys for 3 days. After this the diet was changed to diets containing higher levels of Lys to 0.40, 0.47, 0.54, and 0.61 percent every third day. The Lys requirement was estimated to be 0.46 percent of diet from the response of plasma Lys concentration of five Lys levels. In Experiment 4, a 4 × 4 Latin square design was used for four dietary Lys levels. The Lys requirement was estimated to be 0.47 percent of the diet with a plasma Lys concentration of four Lys levels.  相似文献
