Tumorous crown gall tissue induced on the stems of sunflower (Helianthus annus L. cv. Mammoth Russian) plants by the soil bacterium Agrobacterium tumefaciens strongly reduced Fe3+ to Fe2+ as evidenced by the formation of Fe2+ BPDS complex compared to surrounding “normal” stem tissue. Tumorous tissue contained significantly more Fe, Cu, K, and P, and less Zn and Mg than the nontumorous stem tissue. The reducing environment of the tumorous tissue appears to affect the general nutrition and growth of this tissue.  相似文献

Iron (Fe) uptake and use in plants is genetically controlled and physiological mechanisms such as Fe reduction are induced during Fe‐deficiency stress to make it available. Transfer of DNA into the cell genome by Agrobacterium tumefaciens alters physiological processes and causes undifferentiated growth. Tumor cells in sunflower (Helianthus annus L. cv. Mammoth Russian) show enhanced Fe reduction compared to normal stem tissue in a manner similar to root cells in plants that are genetically switched on or off to manage Fe acquisition. This study addresses whether alterations caused by the DNA transfer from A. tumefaciens result in Fe reduction or whether A. tumefaciens inoculum alone reduces Fe. Reduction of Fe was quantified from A. tumefaciens inoculum and from uninoculated or inoculated sunflower stem tissues daily over a 14‐day period. Neither A. tumefaciens inoculum nor uninoculated stem tissue alone activated massive Fe reduction. High rates of Fe reduction were associated with the proliferation of cells modified by A. tumefaciens. The mechanisms that transformed normal tissue to uncontrolled tumor growth appeared to be linked to active Fe reduction. These modified cells may provide a key to locating and understanding the genetic control of the Fe reduction process in plant cells. Our results suggest a critical role for Fe in development of tumorous tissues and raises the question of whether other tumor cells induce similar mechanisms for Fe acquisition.  相似文献

The effects of nighttime air temperature (NTAT) on the color of milled rice were investigated. Elevated NTATs occurring during the critical grain‐filling stages of kernel development impacted the color of head rice. Six cultivars, grown at multiple field locations from northern to southern Arkansas during 2007–2010, were evaluated for head rice color, using whiteness (L*) and yellowness (b*) indices, and for chalk. Nighttime temperatures were recorded throughout production at each of the selected locations, and the 95th percentiles of NTAT frequencies (NT95) were calculated for each cultivar's reproductive (R) stages. Head rice color values were analyzed in relation to NT95 occurring during the grain‐filling (R6–R8) stages and in relation to percent chalkiness. Whiteness generally increased with increasing NTAT and with increasing chalkiness. Yellowness decreased as chalkiness increased. Moreover, kernel whiteness increased even when measured in the absence of chalky kernels, suggesting that starch granule organization throughout the kernel, even in nonchalky portions, was altered, which could result in compromised physical integrity and processing functionality. Cultivars varied in their susceptibility to the effect of NTAT on color, as has been previously demonstrated with milling quality and functional properties.  相似文献

Fourteen United States (U.S.) seed potato certification agencies surveyed all U.S. seed potato growing areas for presence of the potato spindle tuber viroid (PSTVd). The survey included general surveillance, which involved searching for the occurrence of PSTVd in state seed potato certification records from 1990 through 2000, and a field survey, which involved testing selected crops for PSTVd infection by nucleic acid dot blot hybridization during 1999 through 2001. No PSTVd incident was documented in any of the state certification records, nor was PSTVd detected in the field surveys. All U.S. seed-growing areas were determined to be free of PSTVd. It is concluded that PSTVd has been eradicated and freedom from potato spindle tuber viroid has been successfully maintained in all of the seed potato growing areas in the United States.  相似文献

‘Beacon Chipper’ is a high-yielding, round white potato for the chip-processing market. This variety has an attractive round appearance with shallow eyes and high percentage of oversize tubers. Specific gravity in Michigan ranges from 1.077 to 1.083. Beacon Chipper may fit a window in raw product requirements between out-of-the-field ‘Atlantic’ crop and a late storage crop. This variety has some tolerance to common scab (Steptomyces scabies Thaxter) in comparison to Atlantic and ‘Snowden’. Beacon Chipper also has good chip-processing storage characteristics (light color and low amount of defects) and better tolerance to pressure bruise than Snowden. Based upon nitrogen and spacing management studies, we recommend that Beacon Chipper be planted at 20-cm within-row spacing with 180 lbs N/acre to maximize U.S. No. 1 yield and quality. The origin and pedigree of Beacon Chipper is currently unknown, despite extensive fingerprint analyses. The name Beacon Chipper was chosen to reflect the common nautical theme of Michigan and Maine.  相似文献

In vegetated filter strips (VFS) the presence of perennial vegetation, rhizodeposition of labile organic substrates and the accumulation of an organic residue thatch layer may enhance microbial numbers and activity, thereby increasing the potential for mineralisation of herbicides and herbicide metabolites retained during run-off events. The objective of this laboratory experiment was to compare the mineralisation of atrazine and metolachlor with that of their respective metabolites in VFS and cultivated soil. With the exception of total bacteria, propagule density of the microbial groups, endogenous soil enzymes and microbial diversity were higher in the VFS soil. This correlated with increased mineralisation of metolachlor and its metabolites in the VFS soil and indicates potential for VFS to curtail the subsequent transport of these compounds. In contrast, the mineralisation of atrazine and the majority of its metabolites was substantially reduced in VFS soil relative to cultivated soil. Consequently, the potential for subsequent transport of atrazine and many of its metabolites may be greater in VFS soil than in cultivated soil if reduced mineralisation is not offset by increased sorption in the VFS.  相似文献

Apoptosis is necessary for both initiation and control of spermatogenesis; however, an increase in apoptosis can lead to subfertility/infertility in stallions, causing substantial financial loss in the equine industry. The ability of stem cell factor (SCF), leukemia-inhibiting factor (LIF), granulocyte–macrophage colony-stimulating factor (GM-CSF), and estradiol (E2), alone or in combination, to prevent apoptosis of germ cells in short-term equine testicular cultures was examined. Testicular tissue was sectioned into approximately 2-mm cubes and placed in media-filled culture chambers. Concentrations of SCF (100 ng/mL), LIF (10 ng/mL), GM-CSF (5 ng/mL), and E2 (10−9 mol/l) were added alone or in combination to each well. After 6 hours in culture, the tissue was fixed and immunohistochemically (terminal deoxynucleotidyl transferase-mediated nick-end labeling; TUNEL) stained for apoptosis detection. Apoptotic cells per 100 Sertoli cell nuclei within seminiferous tubules were counted until the 500th Sertoli cell nuclei was reached. This counting procedure was used for each slide. An analysis of variance (ANOVA) with a Tukey's test was used to compare apoptotic rates. In comparison with the control, GM-CSF alone lowered apoptosis by 34.77%. GM-CSF–treated tissue combined with SCF and LIF as well as GM-CSF combined with SCF, LIF, and E2 reduced apoptosis when compared with the control (37.45% and 44.40%, respectively) or other treatment combinations. GM-CSF alone reduced apoptosis; results suggest possible synergy for the combinations of SCF and LIF with GM-CSF and for E2 with SCF, LIF, and GM-CSF.  相似文献

Eleven products were compared for their ability to eliminate cat urine odor in carpet. Their performance on new urine spots and on old spots was compared as was their ability to prevent the return of urine odors after the carpet was cleaned. Newer products marketed to eliminate urine odor did the best in getting rid of and in preventing the return of urine smells. Other products also scored well. A few products were considered to be of little value for the removal of urine odor in carpet.  相似文献
