Recent research has proven soil nitrite to be a key element in understanding N-gas production (NO, N2O, N2) in soils. NO is widely accepted to be an obligatory intermediate of N2O formation in the denitrification pathway. However, studies with native soils could not confirm NO as a N2O precursor, and field experiments mainly revealed ammonium nitrification as the source of NO. The hypothesis was constructed, that the limited diffusion of NO in soil is the reason for this contradiction. To test this diffusion limitation hypothesis and to verify nitrite and NO as free intermediates in native soils we conducted through-flow (He/O2 atmosphere) 15N tracer experiments using black earth soil in an experimental set up free of diffusion limitation. All of the three relevant inorganic N soil pools (ammonium, nitrite, nitrate) were 15N labelled in separate incubation experiments lasting 81 h based on the kinetic isotope method. During the experiments the partial pressure of O2 was decreased in four steps from 20% to about 0%. The net NO emission increased up to 3.7 μg N kg−1 h−1 with decreasing O2 partial pressure. Due to the special experimental set up with little to no obstructions of gas diffusion, only very low N2O emission could be observed. As expected the content of the substrates ammonium, nitrate and nitrite remained almost constant over the incubation time. The 15N abundance of nitrite revealed high turnover rates. The contribution of nitrification of ammonium to the total nitrite production was approx. 88% under strong aerobic soil conditions but quickly decreased to zero with declining O2 partial pressure. It is remarkable that already under the high partial pressure of 20% O2 12 % of nitrite is generated by nitrate denitrification, and under strict anaerobic conditions it increases to 100%. Nitrite is present in two separate endogenous pools at least, each one fed by the nitrification of ammonium or the denitrification of nitrate. The experiments clearly revealed that nitrite is almost 100% the direct precursor of NO formation under anaerobic as well as aerobic conditions. Emitted N2O only originated to about 100% from NO under strict anaerobic conditions (0-0.2% O2), providing evidence that NO is a free intermediate of N2O formation by denitrification. To the best of our knowledge this is the first time that NO has been detected in a native soil as a free intermediate product of N2O formation at denitrification. These results clearly verify the “diffusion limitation” hypothesis.  相似文献

Willows (Salix spp.) are an integral component in the restoration of wetland plant communities that have been impacted by the fluvial deposition of mine tailings. A greenhouse study was conducted to compare growth and metal uptake of Geyer (S. geyeriana) and mountain (S. monticola) willow grown in topsoil versus lime and biosolids amended mine tailings. Biomass, leader length, and tissue metal contents were measured after four months growth. Above and belowground biomass and leader length of Geyer willow were greater for plants grown in topsoil compared to amended mine tailings. However, soil type did not affect mountain willow growth. Analysis for five metals yielded complex results for the two willow species and soil types. As compared to mountain willow, Geyer had greater concentrations of Mn and Pb in aboveground tissues, and Cu in senesced leaves and bark-less leaders when grown in tailings; mountain willow leaves contained greater levels of Cd than Geyer when grown in tailings. Both willow species had foliar Cd levels which were above livestock toxicity tolerance values. Based on growth characteristics, mountain willow appeared better suited for restoration of mine tailings compared to Geyer willow. However, because of the high Cd uptake by both willow species, care should be taken in restoration efforts where wildlife and domestic livestock are likely to browse on the willows.  相似文献

Contemporary soil organic matter (SOM) models have been successful at simulating decomposition across a range of spatial and temporal scales using first-order kinetics to represent the decomposition process; however, recent work suggests the simplicity of the first-order representation of decomposition is not adequate to capture the microbially-driven dynamics of SOM decomposition over short timescales. For example, the response of soils to drying-rewetting events may best be explained by microbial and/or exoenzyme controls on decomposition. To test if adding these microbial mechanisms improves the ability of SOM models to simulate the response of soils to short-term environmental changes, we developed four different SOM decomposition models with varying mechanistic complexity and compared their ability to simulate soil respiration from a pulsed drying-rewetting laboratory-based experiment. Specifically, we tested the ability of the models to capture the timing and magnitude of soil CO2 efflux in response to rewetting or constant moisture conditions. The results of the comparison suggest that the inclusion of exoenzyme and microbial controls on decomposition can improve the ability to simulate pulsed rewetting dynamics; however, less mechanistic first-order models prevail under steady-state moisture conditions. These modeling results may have implications for understanding the long-term response of soil carbon stocks in response to local and regional climate change.  相似文献

Background, aim, and scope  Bed sediments are the major sink for many contaminants in aquatic environments. With increasing knowledge of and research on the environmental occurrence of antibiotics, there has been growing interest in their behaviour and fate in aquatic environments. However, there is little information about the behaviour of antibiotics in a dynamic water/sediment environment, such as river and coastal marine water. Therefore, the aims of the present study were: (1) to study the transport and distribution of four common antibiotics between water and sediment in both dynamic and quiescent water/sediment systems and (2) to understand the persistence and possible degradation of the four antibiotics in the two different systems. Materials and methods  A lid-driven elongated annular flume, designed to reduce the centrifugal effect, was used to simulate a dynamic water environment. In addition, a quiescent water/sediment experiment was conducted for comparison with the dynamic water system. The seawater and sediment, used in both experiments of flowing and quiescent water/sediment systems, were collected from Victoria Harbour, a dynamic coastal environment in an urban setting. The four antibiotics selected in this study were ofloxacin (OFL), roxithromycin (RTM), erythromycin (ETM), and sulfamethoxazole (SMZ), the most commonly used antibiotics in South China. Results and discussion  Antibiotics in an overlying solution decreased very quickly in the flume system due to the sorption to suspended particles and surface sediment. There were significant differences in the adsorption of the four antibiotics in sediment. OFL showed a high tendency to be adsorbed by sediment with a high K d value (2980 L/Kg), while the low K d values of SMZ indicated that there was a large quantity in water. The four antibiotics reached a depth of 20–30 mm in the sediment over a period of 60 days in the flume system. However, the compounds were only found in surface sediment (above 10 mm) in the quiescent system, indicating the influence of the dynamic flume system on the distribution of antibiotics in sediment. OFL showed a moderate persistence in the dynamic flume system, while other three antibiotics had less persistence in sediment. However, all of the four compounds showed moderate persistence in the quiescent system. Recommendations and perspectives  The study showed the rapid diffusive transfer of antibiotics from water to sediment in the dynamic flume system. The four antibiotics exhibited larger differences in their adsorption to sediment in both dynamic and quiescent systems due to their different K d values. The high sorption of antibiotics to marine sediment may reduce their availability to benthic invertebrates.  相似文献

Low-molecular-weight organic acids play an important role in the mobilization of soil phosphorus (P). The molybdate blue colorimetric method, based on the formation of P molybdenum blue compound, is commonly used for analysis of phosphate in soil and environmental samples. However, some organic acids can act as a ligand to bond with molybdenum acid, which subsequently interfere with the colorimetric reaction. The recoveries of P were inhibited by the addition of oxalic (>2 mM) and citric acids (>3 mM) both in standard P solutions and soil extracts, but formic and maleic acids did not interfere with the P determination. The inhibition of oxalic and citric acids on P recovery remained even at higher level of P (up to 100 μg P 100 ml−1) though such interferences decreased by increasing P concentration. Comparison between oxalic and citric acids revealed the more pronounced interference by the addition of oxalic acid. The results suggested that the interference of organic acids with P determination is related to the types of organic acids and the ratio of organic acid ligands to P anions in the solutions. Thus, analysis of P using the molybdate blue colorimetric method should be undertaken cautiously in the presence of relative strong ligands like oxalic and citric acids.  相似文献

OBJECTIVES: Intake and status of carotenoids have been associated with chronic disease. The objectives of this study were to examine the association between carotenoid intakes as measured by two regional food-frequency questionnaires (FFQs) and their corresponding measures in serum, and to report on dietary food sources of carotenoids in Jackson Heart Study (JHS) participants. DESIGN: Cross-sectional analysis of data for 402 African American men and women participating in the Diet and Physical Activity Sub-Study (DPASS) of the JHS. RESULTS: Mean serum carotenoid concentrations and intakes in this population were comparable to those reported for the general US population. After adjustment for covariates, correlations between serum and dietary measures of each carotenoid, for the average of the recalls (deattenuated), the short FFQ and the long FFQ, respectively, were: 035 and 0-carotene; 026 and 0-carotene; 017 and 0-carotene; 034 and 0-cryptoxanthin; 015 and 037, 014 for lycopene. Major dietary sources of -carotene and lutein plus zeaxanthin, mustard, turnip and collard greens; of beta-cryptoxanthin, orange juice; and of lycopene, tomato juice. CONCLUSIONS: On average, carotenoid intakes and serum concentrations are not lower in this southern African American population than the general US population. The two regional FFQs developed for a southern US population and used as dietary assessment tools in the JHS appear to provide reasonably valid information for most of these carotenoids.  相似文献

Fish protein hydrolysate (FPH) produced by incubation of Pacific hake fillet with 3.00% Protamex at pH 6.5 and 40 degrees C for 125 min demonstrated in vitro ACE-inhibitory activity (IC50 = 165 microg/mL), which was enhanced by ultrafiltration through a 10 kDa molecular weight cutoff membrane (IC50 = 44 microg/mL). However, after simulated gastrointestinal digestion, FPH and ultrafiltrate had similar ACE-inhibitory activity (IC 50 = 90 microg/mL), indicating that FPH peptides act as "pro-drug type" inhibitors and that enrichment by ultrafiltration may be unnecessary. Matrix-assisted laser desorption/ionization-time of flight mass spectrometry confirmed that the molecular weights of major peaks were <1 kDa regardless of ultrafiltration. ACE-inhibitory activities of digested hydrolysates were not significantly affected by preincubation with ACE ( P > 0.05) and exhibited a competitive inhibitory mode. A permeability assay using fully differentiated colorectal adenocarcinoma (Caco-2) cells showed an apical to basolateral transport of peptides that ranged from approximately 2 to 20% after 2 h at 37 degrees C. Pacific hake fillet hydrolysates are a potentially bioavailable source of ACE-inhibitory peptides awaiting further in vivo study.  相似文献

Fresh citrus honey was stored at 10, 20, and 40 degrees C for 12 months. The effect of storage on the quality of honey was evaluated using physicochemical parameters, volatile compounds, mono-, di-, and trisaccharides, and sensory analysis. Diastase activity and HMF were out of the legal limit in honey stored 12 months at 40 degrees C. Volatile compounds (especially terpenes and terpene derivatives), monosaccharides, and disaccharides presented important losses during honey storage at any temperature. Honey storage at 10 or 20 degrees C maintained their floral, fresh, citric, and fresh fruit aroma, while the intensities of these attributes were diminished. Storage at 40 degrees C during 12 months resulted in the appearance of attributes such as "medicinal, smoked, toasted, cooked vegetable, and ripened fruit", associated with compounds formed during the Maillard reaction or through degradation of sugars such as volatile pyrroles, furanones, pyranones, and pyrazines, which appeared or increased in concentration during honey storage mainly at high temperature.  相似文献

Silvopastoral systems have great potential for storing carbon because of carbon assimilation in tree woody biomass, carbon input through litterfall and below-ground carbon turnover. In this study, we quantified and compared the carbon stocks at livestock ranches in Tabasco, Mexico, containing either scattered trees in grazing pastures (STP) or grass monocultures. Sampling plots were randomly established at each ranch where the above- and below-ground carbon stocks, carbon input from litterfall, grass production and arboreal biomass growth were measured. We found that silvopastoral systems stored an average of 257.45 Mg ha−1 of soil organic carbon (SOC) compared to 119.17 Mg SOC ha−1 at grass monoculture ranches (to 30 cm depth); silvopastoral systems also stored 44.64 Mg C ha−1 in wood biomass; and, grass monocultures had greater cumulative grass biomass production. Overall, it is concluded that livestock ranches in Tabasco, Mexico, with scattered trees in grazing pastures stored 58.8% more carbon than those grass monocultures, with carbon stocks of 327.01 Mg C ha−1and 134.47 Mg C ha−1, respectively. The results are useful for land management decision making for sustainable livestock systems framed in the Sustainable Development Goals (SDGs).  相似文献
