Anthracnose, caused by Colletotrichum gloeosporioides, is a major post-harvest disease in papaya fruit. The major objectives of the present work were to isolate, select and test the in vitro and in vivo ability of epiphytic microorganisms, isolated from papaya fruit and leaf surfaces, in controlling anthracnose onset after harvest. A total of 75 bacteria, 67 yeasts and 22 mycelial fungi were isolated. Thirty yeast isolates were able to inhibit the mycelial growth of C. gloeosporioide in vitro and seven of those were used in in vivo assays, resulting in the identification of two very effective isolates. Isolate CEN63, identified molecularly as Cryptococcus magnus, was the most effective in controlling the disease and therefore was studied in more detail. The results of the assays with C. magnus provided evidence that when fruit were treated with the antagonists at concentrations of 107 to 108 cells/ml, as early as 24 h, preferentially 48 h, before inoculation with the pathogen, the development of disease was significantly reduced. C. magnus is a potential antagonist for the development of a commercial product. Additional studies on the modes of action of this yeast isolate, as on its ability to interact with fungicides are being conducted to generate solid basis for the development of an environmentally friendly control agent. 相似文献

An attempt was made to understand the molecular systematics and genetic differences between 10 original chrysanthemum cultivars and 11 mutants. The similarity among the cultivars and mutants varied from 0.17 to 0.90 using RAPD analyses, a simple but efficient method to distinguish cultivars and to assess parentage. Two distinct groups were found. Two cultivars were present as a separate group showing differences from all other cultivars. Mutants with different flower colour could be identified at the molecular level using RAPD technique holding promise to identify unique genes as SCAR markers. A high genetic distance among the different chrysanthemums showed that there exists a possibility of introgressing new and novel genes from the chrysanthemum gene pool. 相似文献

The present investigation was conducted to study organogenesis of a conventionally easy-to-regenerate (‘Royal Gala’) and a difficult-to-regenerate (‘Freedom’) apple cultivar using leaf transverse thin cell layers (tTCLs). The position of the leaf source, explant type and light regime applied to tTCLs were investigated as was their influence on the organogenic outcome. Light during the onset of regeneration delayed morphogenesis or, in ‘Freedom’, completely inhibited organogenesis on leaf tTCLs. Genotype and initial position of the leaf also influenced percentage regeneration and number of regenerated shoots per explant. This study demonstrates the successful application of leaf tTCLs for adventitious shoot regeneration in apple, even though further studies are needed to increase the number of shoots more than control explants. Shoot regeneration from apple leaf tTCLs serves as an alternative protocol for studying fundamental and applied aspects of regeneration. 相似文献

Sea buckthorn (Hippophae L.) is a woody, outcrossing dioecious pioneer plant, being widely planted as a new berry crop with rich nutritional and medicinal compounds, as well as a means of prevention of soil erosion. Inter simple sequence repeat (ISSR) markers were generated in 219 plants of 40 cultivars from China and other countries, representing two species of Hippophae salicifolia and Hippophae rhamnoides that includes two subspecies (ssp. mongolica and ssp. rhamnoides) and intraspecific hybrids between ssp. mongolica and ssp. sinensis. Fifteen selected ISSR primers generated 385 bands with an average of 25.6 bands per primer. Thirty hundred and eighty-five bands were used to generate a Dice's coefficient matrix of pairwise comparisons between individual ISSR profiles. Cluster analysis based on this matrix showed clustering of plants into groups which was in good agreement with their taxonomic classification. The results also revealed the presence of different cultivars with the same name, identical cultivars but with a different name and the genetic relationship of cultivars with unknown parentage to those with known parentage. Our data will be helpful in collection and conservation of sea buckthorn germplasm and potential hybrid strategy for breeding. 相似文献

The intensification of olive production methods in southern Spain, has involved a widespread use of chemicals and the progressive loss of many Mediterranean forest patches, natural landscapes and semi-natural vegetation. Species level inventories require an enormous amount of time and financial resources, mainly due to the necessity of employing taxonomists in the early stages of a study. The use of a higher taxonomic level is particularly useful when rapid biodiversity surveys are required. This survey investigates the reliability of a rapid method to detect which high taxonomic level of epigeal insects could best distinguish different farming systems in olive agroecosystems, taking into account three higher taxonomic categories as surrogate measures of insect diversity: orders for all the insects, families of Coleoptera and carabid morphospecies. Insects were collected in pitfall traps in olive orchards under organic, integrated and conventional farming systems in two provinces in Andalusia (Córdoba and Granada) with different surrounding landscapes over three different years. In 1999 and 2000 a study of trends over time was undertaken between March and October of both years in Granada, and in 2003 a geographically extended study was conducted in 18 different orchards during pre-blooming and post-blooming periods in Granada and Córdoba. To rapidly assess biodiversity in olive agroecosystems by examining soil communities, in order to classify farming systems on a regional scale, the taxonomic level of order seems to be a more reliable approach than a lower taxonomic level in insects. This methodology could be used as a possible useful short-cut to assess biodiversity in olive orchards at a local scale, the order surrogacy being useful when results are required rapidly and in a context of limited financial resources. 相似文献

Species of the Santalum genus are well known for their fragrant hardwood, which has great value in medicinal, pharmaceutical and cosmetic industries. Sandalwood oil is derived from the heartwood of Santalum sp. and contains α-, β- and epi-β-santalols, which are responsible for its pleasant fragrance. Oil content can vary from species to species. Pressure on natural populations due to habitat loss, legal and illegal harvesting and disease is increasing. This paper highlights the development of molecular markers for the refinement of phylogenetic studies, identification of various Santalum and adulterant species, assessment of genetic diversity, genetic differentiation, clonality and management units within species, and for marker-assisted breeding. The identification of quantitative trait loci for sandal spike disease and for other traits such as specific rare secondary metabolites in the essential oil and related to its fragrance, would also benefit from molecular advances. RNA sequence analyses have already identified changes in gene expression and metabolic pathways in developing Santalum album L. haustoria. 相似文献

ABSTRACTInformation regarding carbon concentration and wood density are lacking in Chilimo dry Afromontane forest.The aim of this study was to estimate carbon concentration and wood density for Allophyllus abyssinicus, Olea europaea, Olinia rochetiana, Rhus glutinosa, and Scolopia theifolia. A total of 105, 30–50 mm thick wood discs were collected and oven dried at 102°C and 67°C to constant weight, chopped and finally grinded into 0.2 mm with a grinding mill. Carbon concentration was analyzed using the ash method, while wood density was estimated using the water displacement method. The highest carbon concentration (57.12%) was found for O. rochetiana, however, the lowest carbon concentration (56.43%) was found for A. abyssinicus. Stem parts had higher carbon concentration (56.98%) than branch (56.74%) and leave (54.53%) parts. O. europaea exhibited the highest wood density (0.67 g cm?3) value than other species. However, the lowest wood density (0.42 g cm?3) was exhibited for A. abyssinicus. Wood density was also showed a decreasing trend along with increases in stem height and maximum wood density (0.62 g cm?3) was found under stump position, while, the minimum wood density (0.4 g cm?3) was found under tree commercial height. 相似文献
