In 2007–2008, a canine distemper virus (CDV) epidemic occurred among wild animals in
Wakayama Prefecture, Japan, and many mammals, including the wild boar and deer, were
infected. In this study, CDV prevalence among wild animals was surveyed before and after
the epidemic. At first, an enzyme-linked immunosorbent assay (ELISA) with horseradish
peroxidase-conjugated protein A/G was established to detect CDV antibodies in many
mammalian species. This established ELISA was available for testing dogs, raccoons and
raccoon dogs as well as virus-neutralization test. Next, a serological survey of wild
mammalians was conducted, and it was indicated that many wild mammalians, particularly
raccoons, were infected with CDV during the epidemic, but few were infected before and
after the epidemic. On the other hand, many raccoon dogs died during the epidemic, but CDV
remained prevalent in the remaining population, and a small epidemic occurred in raccoon
dogs in 2012–2013. These results indicated that the epidemic of 2007–2008 may have been
intensified by transmission to raccoons. 相似文献

Myoblasts are myogenic precursors that develop into myotubes during muscle formation. Improving efficiency of myoblast differentiation is important for advancing meat production by domestic animals. We recently identified novel oligodeoxynucleotides (ODNs) termed myogenetic ODNs (myoDNs) that promote the differentiation of mammalian myoblasts. An isoquinoline alkaloid, berberine, forms a complex with one of the myoDNs, iSN04, and enhances its activities. This study investigated the effects of myoDNs on chicken myoblasts to elucidate their species-specific actions. Seven myoDNs (iSN01–iSN07) were found to facilitate the differentiation of chicken myoblasts into myosin heavy chain (MHC)-positive myotubes. The iSN04–berberine complex exhibited a higher myogenetic activity than iSN04 alone, which was shown to enhance the differentiation of myoblasts into myotubes and the upregulation of myogenic gene expression (MyoD, myogenin, MHC, and myomaker). These data indicate that myoDNs promoting chicken myoblast differentiation may be used as potential feed additives in broiler diets. 相似文献

The objective of this study was to clarify the current status of endangered Kiso horse,
population statistics and biological traits, in order to take a step for the conservation
by scientific approach. We surveyed 125 Kiso horses (86.2% of the whole breed), analyzed
the construction of the population, and calculated the coefficient of inbreeding and
effective population size. Moreover, we confirmed coat color variations and the
traditional traits of the Kiso horse, and measured their height at the withers and chest
circumference to clarify their physical characteristics. The population pyramid of the
horses was stationary or contractive, suggesting a reduction of the population in the near
future. The effective population size of the horse (47.9) suggested that the diversity was
much less than their census size, and the high coefficient of inbreeding, 0.11 ± 0.07 on
average, suggested that the horses were surely inbred. The horses had only 4 coat colors;
bay, dark bay, buckskin dun, and chestnut, and 116 horses (92.8%) were bayish color,
suggesting the fixation in their coat color. Moreover, the majority of them had dorsal
stripe (83 horses; 66.4%), and the average heights at withers(131.9 ± 4.4 cm) and chest
circumference (167.1 ± 10.1 cm) were not significantly different between males and

Calpains are intracellular Ca2+‐dependent proteases and enzymes that contribute to growth and meat quality. In the present study, we identified polymorphisms in four calpain genes (CAPN1, CAPN2, CAPN3, and CAPN1.5) expressed ubiquitously in chicken using polymerase chain reaction‐restriction fragment length polymorphism, and mapped them using two backcross families (East Lansing (EL) and Kobe University (KU)). CAPN2 and CAPN1.5 mapped to two locations on chromosome 3 about 30 cM apart, while CAPN3 mapped to chromosome 5. CAPN1 was linked to a previously unlinked microsatellite marker LEI0140 to form a new linkage group called E66. CAPN2 and CAPN3 extend the amount of conserved synteny between chicken chromosome 3 and human chromosome 1, and between chicken chromosome 5 and human chromosome 15, respectively. Although CAPN2, CAPN3, and CAPN1.5 were found in the University of California Santa Cruz chicken genome browser gateway, CAPN1 and LEI0140 were not in specific genomic positions. 相似文献

The mithun (Bos frontalis), synonymous with mithan and gayal, is considered to be a domesticated form of gaur (B. gaurus). However, there has been a controversy concerning its origin. In an effort to address this issue, the mitochondrial cytochrome b (cytb) genes of 20 mithun from Myanmar and 13 from Bhutan were sequenced to trace its maternal origin. Seven cytb haplotypes were found in the 33 mithun, and the phylogenetic tree for these haplotypes clearly showed three embranchments involving five gaur types, a B. indicus type, and a B. taurus type. Sixteen Myanmar and 12 Bhutan mithun had gaur haplotypes, while a B. indicus haplotype was found in three Myanmar and one Bhutan mithun. The B. taurus haplotype was detected in a single Myanmar animal. These results demonstrated that the principal maternal origin of mithun was gaur and suggested that it was directly domesticated from gaur. However, some introgression of domestic cattle existed in current mithun populations. The presence of cattle mtDNA raised the question of how many cattle nuclear genes might have been integrated into the gene pool of mithun. 相似文献

Accumulating evidence suggests that the arcuate nucleus (ARC) kisspeptin/neurokinin B
(NKB)/dynorphin (KNDy) neurons play a role in estrogen negative feedback action on
pulsatile gonadotropin-releasing hormone (GnRH)/luteinizing hormone (LH) release. The
present study aimed to determine if dynorphin (Dyn) is involved in estrogen negative
feedback on pulsatile GnRH/LH release. The effect of the injection of nor-binaltorphimine
(nor-BNI), a kappa-opioid receptor (KOR) antagonist, into the third cerebroventricle (3V)
on LH pulses was determined in ovariectomized (OVX) adult female rats with/without
replacement of negative feedback levels of estradiol (low E2). The mean LH
concentrations and baseline levels of LH secretion in nor-BNI-injected, low
E2-treated rats were significantly higher compared with vehicle-treated
controls. On the other hand, the nor-BNI treatment failed to affect any LH pulse
parameters in OVX rats without low E2 treatment. These results suggest that Dyn
is involved in the estrogen negative feedback regulation of pulsatile GnRH/LH release. The
low E2 treatment had no significant effect on the numbers of ARC
Pdyn (Dyn gene)-,Kiss1- and
Tac2 (NKB gene)-expressing cells. The treatment also did not affect
mRNA levels of Pdyn and Oprk1 (KOR gene) in the
ARC-median eminence region, but significantly increased the ARC kisspeptin
immunoreactivity. These findings suggest that the negative feedback level of estrogen
suppresses kisspeptin release from the ARC KNDy neurons through an unknown mechanism
without affecting the Dyn and KOR expressions in the ARC. Taken together, the present
result suggests that Dyn-KOR signaling is a part of estrogen negative feedback action on
GnRH/LH pulses by reducing the kisspeptin release in female rats. 相似文献

AIM:To study the effect of domain 5 of the active high molecular weight kininogen (HKa) on the vitronectin-mediated adhesion and migration and angiogenesis of human umbilical vein endothelial cells (HUVECs) in vitro. METHODS:Recombinant HK domain 5 (GST-D5H) was prepared. WST-1 was used to study the effects of adhesiveness by GST-D5H on HUVECs. The Boyden Chamber membrane invasion culture system was used to study the migration, and tube formation was used to study angiogenesis. RESULTS:The adhesion rate was down regulated in GST-D5H group (P<0.05). In the membrane invasion culture system, the number of migrating HUVECs was significantly lower in GST-D5H group than that in control group (P<0.01). The number and length of tubes were significantly lower in GST-D5H group (P<0.05) than that in control. CONCLUSION:GST-D5H inhibits vitronectin-mediated adhesion, migration and angiogenesis of HUVECs in vitro. 相似文献

Recently, developmental exposure to clothianidin (CLO) has been shown to cause reproductive toxicity in male mice, but the effects in female mice remain to be clarified. Pregnant C57BL/6N mice were given a no-observed-adverse-effect-level (NOAEL) dose of CLO until weaning. We then examined ovaries of 3- or 10-week-old female offspring. In the CLO-administered group, morphological changes, a decrease in the immunoreactivity of the antioxidant enzyme glutathione peroxidase 4 (GPx4), and activation of genes in the steroid hormone biosynthesis pathway were observed in 3-week-old mice, and decreases of GPx4 immunoreactivity, 17OH-progesterone and corticosterone levels were observed in 10-week-old mice, along with high rates of infanticide and severe neglect, providing new evidence that developmental exposure to CLO affects juvenile and adult mice differently. 相似文献
