Trilostane-induced inhibition of cortisol secretion results in reduced negative feedback at the hypothalamic–pituitary axis

Cushing's disease caused by pituitary corticotroph adenoma in dogs is usually treated by medical treatment, and the efficacy of this treatment has been reported. However, controversy remains as to whether reduced negative feedback through the inhibition of cortisol secretion, similar to Nelson's syndrome, may appear as an adverse effect. The purpose of this study was to investigate the effect of reduced negative feedback through the inhibition of cortisol secretion by daily trilostane administration on the pituitary–adrenal axis in clinically normal dogs. Dogs were administered 5 mg/kg trilostane twice a day every day for 8 weeks (n = 8) or 16 weeks (n = 3). After the initiation of trilostane administration, plasma adrenocorticotropic hormone (ACTH) concentrations were increased remarkably. As assessed by magnetic resonance imaging (MRI) during administration, the pituitary became enlarged. After trilostane administration, the cytoplasmic areas of the pituitary corticotrophs were increased and the ratio of pituitary corticotrophs to all cells in the anterior lobe was greater in the trilostane-treated dogs than that in untreated animals. In addition, histological examinations revealed bilateral adrenal cortical hyperplasia. Using real-time PCR quantification, the expression of proopiomelanocortin (POMC) mRNA in the pituitary and ACTH receptor (ACTH-R) mRNA in the adrenal gland was greater in the dogs treated with trilostane than in untreated dogs. These results indicate that reduced negative feedback induced hyperfunction of the pituitary corticotrophs and pituitary enlargement in healthy dogs. These changes suggest that the inhibition of cortisol secretion by trilostane may increase the risk for accelerating the growth of corticotroph adenomas in dogs with Cushing's disease.     

White root rot disease of the lacquer tree <Emphasis Type="Italic">Toxicodendron vernicifluum</Emphasis> caused by <Emphasis Type="Italic">Rosellinia necatrix</Emphasis>

In early August 2010, lacquer trees (Toxicodendron vernicifluum) severely damaged by a root rot disease were found on plantations in Iwate, Japan. The causal agent was a fungus identified as Rosellinia necatrix, based on morphology and the sequence of the ribosomal DNA internal transcribed spacer region. The fungus was clearly pathogenic on T. vernicifluum root plantings. This report is the first of white root rot on T. vernicifluum.     

Development of high-grade B-cell lymphoma concurrent with T-cell chronic lymphocytic leukemia in a dog

Second malignancies are frequent complications in human patients with chronic lymphocytic leukemia (CLL). However, the clinical details and outcome of this phenomenon were unclear in their canine counterparts. Here, we report a dog with high-grade lymphoma concurrent with T-cell CLL. A 10-year-old male golden retriever presented with lymphadenopathies. The lymph nodes contained large-sized lymphocytes, raising suspicion of high-grade lymphoma. Meanwhile, small lymphocytic lymphocytosis in the peripheral blood was consistent with CLL. Interestingly, molecular biological analyses revealed that CLL cells were of the T-cell type, whereas lymphoma cells were of the B-cell type. Chemotherapy using the L-VCA short protocol was effective for 155 days, but the dog died on day 194 after diagnosis, despite rescue therapies.     

Histopathology of incidental findings in beagles used in toxicity studies

The purpose of our publication is to widely communicate the pictures of spontaneous findings occurring in beagles. Spontaneous arteritis occurs commonly in beagles. Frequent sites of arteritis are the heart, spleen, pancreas, epididymis and spinal cord. Morphological similarities between spontaneous and drug-induced arterial lesions may cause confusion when evaluating vascular toxicity of chemicals such as vasodilating agents. Focal and minimal inflammatory lesions are occasionally seen in the lung and may be associated with aspiration of food particles or of unknown causes. A cystic change with copious mucin production occurs occasionally in the mucosal epithelium of the gall bladder. Nesidioblastosis is seen rarely in the pancreas of beagles. C-cell complex and lymphocytic thyroiditis are common thyroid lesions. Spontaneous focal hypospermatogenesis and lobular Sertoli-cell-only seminiferous tubules occurring frequently in beagles must be distinguished from drug-induced damage of the seminiferous tubules in toxicity studies. The morphological differences of the female genital system in each cycle need to be understood; therefore, we present the normal features of the cyclic changes of the female genital organs. Further, we provide more information on spontaneous findings in beagles for exact diagnoses in toxicity studies.     

Ehrlichia canis infection in two dogs that emigrated from endemic areas

Two dogs, emigrated from Zambia and China to Japan, were diagnosed with Ehrlichia canis infection. Both cases had thrombocytopenia, non-regenerative anemia, and hypergloblinemia with polyclonal gammopathy. Case 1 had ataxia of the hind limbs. Severe meningitis was revealed by magnetic resonance imaging examination. Intracytoplasmic inclusions were observed in mononuclear cells of cerebrospinal fluid. Case 2 had a history of bilateral epistaxis, and severe pancytopenia was noticed in complete blood count. Diagnosis was finally achieved by nested polymerase chain reaction and sequence analysis. Thus, even in non-endemic areas, E. canis infection should be included in the differential diagnosis of clinically ill dogs that emigrated from endemic areas.     

Difference in age at first conception between early and late litters of feral raccoon (Procyon lotor) in Kanagawa prefecture

Due to the long parturition period of raccoons, we assumed that age at first conception of late born females was later than that of early born females. From March 2005 to September 2008, 201 females estimated to be younger than 24 months were separated into early- and late-born groups on the basis of their estimated birth month (to the nearest 2 months), and parous status and body mass index (BMI) were examined. Age at first conception of late-born females (over 18 months old) was estimated to be later than that of early-born females (over 12 months old). The average BMI in early-born individuals at 12 months old might have affected their ability to conceive because of body fat deposition.     

Projections of nutrient supply to a wakame (Undaria pinnatifida) seaweed farm on the Sanriku Coast of Japan

Wakame (Undaria pinnatifida) aquaculture is one of the major fishery industries on the Sanriku Coast of Japan. Low concentrations of nutrients in the autumn, when the germinated wakame sporophytes are installed at aquaculture facilities, induce defective growth, resulting in withering. To determine the characteristics of the nutrient variations in this region, we conducted hydrographic measurements off the coast of Iwate Prefecture. The measured results showed that in the autumn, vertical mixing pumped nutrients from the deeper layers to the mixed layer, where nutrients had been depleted during the summer. Based on these results, we developed a one‐dimensional model incorporating vertical mixing by atmospheric cooling to project the vertical distribution of the nutrient concentrations. By conducting ensemble calculations, we created a probability projection calendar for the nutrient supply up to 50 days ahead. This information was provided to wakame farmers before they installed the germinated sporophytes. The projected probability calculated from the initial conditions on 11 October 2016 was 0% until 25 October and gradually increased to 85% on 30 November. The nitrate concentration detected by an ultraviolet nitrate sensor moored at a wakame farm revealed that the nutrient concentrations reached a critical threshold on 18 November. The projected probability on that date was 75%; therefore, the projection was successful. The observed temporal evolution of the temperature, salinity, and nutrient concentrations was well reproduced by our model. The information obtained using this model is expected to reduce the risk of wakame withering and improve the efficiency of wakame production.
