A novel A-seco limonoid was isolated from methanolic extract of Croton jatrophoides and designated as zumsin. This compound showed potent antifeedant activity against two lepidopteran larvae, pink bollworm, Pectinophora gossypiella (PC(50) = 1 microg/cm(2), PC(95) = 8 microg/cm(2)), and fall armyworm, Spodoptera frugiperda (PC(50) = 2 microg/cm(2), PC(95) = 16 microg/cm(2)). The structure of zumsin was determined as 1 using a variety of spectroscopic methods including nuclear magnetic resonance, mass spectrometry, and circular dichroism. The structure consists of an A'-B trans-fused ring while dumsin (2), a constituent of the same source, maintains an A'-B cis-fused ring, and suggests two unique biosynthetic processes after A ring oxidative expansion.  相似文献

The effects of ozone (O3) and excess soil nitrogen (N), singly and in combination, on growth, needle gas exchange rates and ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) contents of Pinus densiflora seedlings were investigated. One-year-old seedlings were grown in 1.5-L pots filled with brown forest soil with 3 levels of N supply (0, 100 or 300 mg N·L?1 fresh soil volume). The seedlings were exposed to charcoal-filtered air or 60±5 nL·L?1 O3 (8 hours a day) in naturally-lit phytotrons for 173 days from 22 May to 11 November. The exposure to O3 or high N supply to the soil caused a significant reduction in the dry weights of the seedlings. Although no significant interactive effects of O3 and excess soil N were detected on the dry weight growth of the seedlings, the whole-plant dry weight of the O3-exposed seedlings grown in the soil treated with 300 mg N·L?1 was greatly reduced compared with the control value. Ozone reduced net photosynthetic rate at 350 µmol·mol?1 CO2 (A 350 ), carboxylation efficiency (CE) of photosynthesis and Rubisco content without a significant change in the gaseous phase diffusive conductance to CO2 (gs) of the needles. The excess soil N reduced the A 350 , CE, gs and Rubisco content of the needles. These results suggest that the reduction in the dry weight growth of Pinus densiflora seedlings induced by the exposure to O3 and/or excess soil N was caused by reduction in the net photosynthetic rate mainly due to the decrease of Rubisco quantity in the chloroplasts.  相似文献

A new method has been proposed for detection of lac color in food. Lac color is a natural color additive derived from a secretion of the insect Coccus Laccae (Laccifer lacca Kerr). It is extracted from food with methanolic oxalic acid and eluted from a column of Amberlite XAD-2 with the same solvent. The fraction containing the lac color is treated with diazomethane to produce 2 reddish-orange markers. The marker species in the reaction mixture are detected by both thin-layer chromatography and reverse-phase liquid chromatography.  相似文献

Twenty-four plant lignans were analyzed by high-performance liquid chromatography-tandem mass spectrometry in bran extracts of 16 cereal species, in four nut species, and in two oilseed species (sesame seeds and linseeds). Eighteen of these were lignans previously unidentified in these species, and of these, 16 were identified in the analyzed samples. Four different extraction methods were applied as follows: alkaline extraction, mild acid extraction, a combination of alkaline and mild acid extraction, or accelerated solvent extraction. The extraction method was of great importance for the lignan yield. 7-Hydroxymatairesinol, which has not previously been detected in cereals because of destructive extraction methods, was the dominant lignan in wheat, triticale, oat, barley, millet, corn bran, and amaranth whole grain. Syringaresinol was the other dominant cereal lignan. Wheat and rye bran had the highest lignan content of all cereals; however, linseeds and sesame seeds were by far the most lignan-rich of the studied species.  相似文献

Measles virus (MeV) vaccine strain, AIK-C, is temperature sensitive (ts), which is thought to be associated with attenuation of virus pathogenicity. In this study, replication and antibody response were examined in cotton rats using viruses carrying different forms of the P gene, which is responsible for the ts phenotype of strain AIK-C and its parental Edmonston strain. When cotton rats were inoculated intranasally, ts viruses neither replicated in lungs, nor reproducibly generated an antibody response. When inoculated intramusculary (i.m.), however, ts strains raised an antibody titer in all animals. This response was not observed when ultraviolet-inactivated virus was used. ts virus, inoculated i.m., was recovered from cotton rat drainage lymph nodes. These results suggest that ts virus, inoculated i.m., could replicate in the cotton rat, presumably at the superficial lymph node, and induce an antibody response. Therefore, cotton rats can serve as a small-animal model for investigating immune responses to safer ts vaccine, as well as recombinant vaccine using AIK-C as a vector for protection against other infectious agents.  相似文献

The immune system in juvenile calves is immature, so calves are susceptible to several diarrheal and respiratory diseases. Oral administration of lactic acid bacteria (LAB) is known to improve the growth performance and prevent diarrheal and respiratory diseases by stimulating the immune system in juvenile calves. Most of the immunostimulation by LAB is achieved by their cell wall components, and therefore we evaluated the immunostimulation of the cell preparation of Enterococcus faecalis strain EC-12 (EC-12) in juvenile calves in a clinical field. Twenty-nine 1-week old calves were used. Fourteen calves were administered 0.2% (w/w) of an EC-12 preparation that supplemented a milk replacer, and other calves were not supplemented. Feces and serum was collected at day 0, 7 and 49 after the administration to measure the IgA and IgG concentration. The fecal IgA concentration was increased by EC-12 administration at day 49, and the serum IgA concentration was also increased at day 7. These results suggested that oral administration of EC-12 in juvenile calves might have an immunostimulatory effect and provide earlier recovery of IgA levels in mucosal immunity.  相似文献

An experiment was conducted to examine the effect of CO2 enrichment on the nitrate uptake, nitrate reduction activity, and translocation of assimilated-N from leaves at varying levels of nitrogen nutrition in soybean using 15N tracer technique. CO2 enrichment significantly increased the plant biomass, apparent leaf photosynthesis, sugar and starch contents of leaves, and reduced-N contents of the plant organs only when the plants were grown at high levels of nitrogen. A high supply of nitrogen enhanced plant growth and increased the reduced-N content of the plant organs, but its effect on the carbohydrate contents and photosynthetic rate were not significant. However, the combination of high CO2 and high nitrogen levels led to an additive effect on all these parameters. The nitrate reductase activity increased temporarily for a short period of time by CO2 enrichment and high nitrogen levels. 15N tracer studies indicated that the increase in the amount of reduced-N by CO2 enrichment was derived from nitrate-N and not from fixed-N of the plant. To examine the translocation of reduced-N from the leaf in more detail, another experiment was conducted by feeding the plants with 15NO3-N through a terminal leaflet of an upper trifoliated leaf under depodding and/or CO2 enrichment conditions. The export rate of 15N from the terminal leaflet to other plant parts decreased by depodding, but it increased by CO2 enrichment. CO2 enrichment increased the percentage of plant 15N in the stem and / or pods. Depodding increased the percentage of plant 15N in the leaf and stem. The results suggested that the increase in the leaf nitrate reduction activity by CO2 enrichment was due to the increase of the translocation of reduced-N from leaves through the strengthening of the sink activity of pods and / or stem for reduced-N.  相似文献

Heavy application of ammonium nitrogen to plant roots results in the conversion of ammonium nitrogen to the nitrogen of amides, glutamine, and asparagine, which are stored in roots or translocated to shoot. Since the net synthesis of such amides requires the supply of corresponding carbon skeletons, the carbon metabolism in amide synthesis in response to ammonium supply was investigated in tomato and wheat roots. The content of major primary amino acids was determined in tomato and wheat roots during a 4-d period of ammonium nutrition after 1-d culture in nitrogen-free nutrient solution. Ammonium supply led to a continuous increase in the asparagine content in wheat roots, whereas in tomato roots, the glutamine content increased 1 d after ammonium supply and thereafter the glutamine content was higher than the asparagine content. The amounts of amino acids synthesized from glucose-14C increased while the amounts of organic acids decreased in tomato roots by the supply of ammonium nitrogen for 1 d, compared to the roots that did not receive nitrogen. In tomato roots, the proportion of labeled glutamine was higher than that of labeled asparagine and the C5 amino acids were more strongly labeled than the C4 amino acids. These findings were different from the previous ones in wheat roots where the proportion of asparagine was found to be extremely high (Koga and Ikeda 2000: J. Fac. Agr. Kyushu Univ45, 7–13). To examine the in vivo asparagine synthesis, aspartate-'4C was fed to the roots. The labeling of asparagine, which was the most strongly labeled amino acid among the free amino acids, was remarkably strong in wheat roots whereas the labeling of glutamine was also pronounced in tomato roots. These results indicate that the ability to replenish carbon skeletons for amide synthesis in ammonium nutrition is different between tomato and wheat roots.  相似文献
