The slope‐ratio assay for rat was used to determine whether tryptophan (Trp) availability in soybean meal (SBM) is affected by the presence of other amino acids (AAs). In a preliminary study, rats were fed graded levels of Trp‐supplemented diets to establish the Trp concentration range over which the weight gain response was linear. This range was found to be from 0.04% to 0.12% Trp. Subsequently, rats were fed basal (0.045% Trp) or Trp‐supplemented diets from three different sources: l ‐Trp alone, SBM, or l ‐Trp mixed with other AAs to reflect AA levels in the test SBM (AA‐mix). Weight gain in rats increased linearly with supplemental Trp intake (p < .05) for all Trp sources. Compared to the slope achieved with l ‐Trp alone, the estimated availability of Trp in SBM was 84.4%, while for the AA‐mix it was 93.4%. It is evident that the 6.6% reduction in l ‐Trp availability in AA‐mix is due to metabolic costs derived from excess levels of other AAs beside Trp, given that the absorption of crystalline l ‐Trp in the small intestine is 100%. In conclusion, the Trp availability of SBM was estimated to be around 90.4% (i.e., 84.4/93.4 × 100) after correcting for the effects of the other AAs in SBM.  相似文献

Porcine edema disease (ED) is a toxemia that is caused by enteric infection with Shiga toxin 2e (Stx2e)‐producing Escherichia coli (STEC) and is associated with high mortality. Since ED occurs most frequently during the weaning period, preweaning vaccination of newborn piglets is required. We developed stx2eB‐transgenic lettuce as an oral vaccine candidate against ED and examined its protective efficacy using a piglet STEC infection model. Two serially developed Stx2eB‐lettuce strains, 2BN containing ingredient Stx2eB constituting a concentration level of 0.53 mg Stx2eB/g of powdered lettuce dry weight (DW) and 2BH containing ingredient Stx2eB constituting a concentration level of 2.3 mg of Stx2eB/g of powdered lettuce DW, were evaluated in three sequential experiments. Taken the results together, oral administration of Stx2eB‐lettuce vaccine was suggested to relieve the pathogenic symptoms of ED in piglets challenged with virulent STEC strain. Our data suggested that Stx2eB‐lettuce is a promising first oral vaccine candidate against ED.  相似文献

The long juvenile period of citrus seedlings before flowering is one of the major obstacles in citrus breeding. Under the citrus breeding program of National Institute of Fruit Tree Science in Japan, 299 citrus hybrid seedlings were grafted onto shiikuwasha (Citrus depressa HAYATA) and cultivated. Approximately one-third of the plants flowered two and a half years after grafting. The average length of the main stems of flowering plants was 308 cm, while that of non-flowering plants was 264 cm. Half of the plants having main stem length ≥300 cm formed flowers, while most of the plants having main stem length <210 cm formed no flower. These results indicate that plant vigor influences the flowering and that grafting effectively accelerated flowering. The proportion of flowering plants varied among cross combinations, ranging from 71.4% to 8.0% among the 10 cross combinations used.  相似文献

Fusariosis, one of the phytosanitary problems found in rubber producing areas in the northwest of the state of São Paulo, is a disease that affects the bark of the adult plants, affecting the exploitation of latex. The typical symptoms appear as cracks in the bark that expand from the rootstock towards the tapping panel, causing a drying of the latex flux in the injured region, impeding latex tapping. Due to the recent incidence of this disease in rubber plantations, the goal of this study was to characterize the Fusarium associated with symptomatic rubber tissue in three different locations in the state of São Paulo. In order to identify Fusarium species, pathogenicity, morphological, cultural and molecular studies were carried out. A total of 51 isolates were obtained and separated into three groups based on macroconidium morphology, presence or absence of sporodochia, types of chlamydospores, formation of phialides and conidiogenesis of microconidia and mesoconidia, mycelial growth rate and coloring of the colonies. These groups were corroborated using DNA sequence information for five different genetic loci, and were subsequently recognized as Fusarium oxysporum, F. incarnatum and F. decemcellulare. Our results further showed that all 51 of the Fusarium isolates recovered were pathogenic to rubber tree seedlings of RRIM 600 standard clone. This study also reports for the presence of F. oxysporum and F. incarnatum in rubber plantations in the state of São Paulo and in Brazil.

An experiment was conducted to examine the effect of CO2 enrichment on the nitrate uptake, nitrate reduction activity, and translocation of assimilated-N from leaves at varying levels of nitrogen nutrition in soybean using 15N tracer technique. CO2 enrichment significantly increased the plant biomass, apparent leaf photosynthesis, sugar and starch contents of leaves, and reduced-N contents of the plant organs only when the plants were grown at high levels of nitrogen. A high supply of nitrogen enhanced plant growth and increased the reduced-N content of the plant organs, but its effect on the carbohydrate contents and photosynthetic rate were not significant. However, the combination of high CO2 and high nitrogen levels led to an additive effect on all these parameters. The nitrate reductase activity increased temporarily for a short period of time by CO2 enrichment and high nitrogen levels. 15N tracer studies indicated that the increase in the amount of reduced-N by CO2 enrichment was derived from nitrate-N and not from fixed-N of the plant. To examine the translocation of reduced-N from the leaf in more detail, another experiment was conducted by feeding the plants with 15NO3-N through a terminal leaflet of an upper trifoliated leaf under depodding and/or CO2 enrichment conditions. The export rate of 15N from the terminal leaflet to other plant parts decreased by depodding, but it increased by CO2 enrichment. CO2 enrichment increased the percentage of plant 15N in the stem and / or pods. Depodding increased the percentage of plant 15N in the leaf and stem. The results suggested that the increase in the leaf nitrate reduction activity by CO2 enrichment was due to the increase of the translocation of reduced-N from leaves through the strengthening of the sink activity of pods and / or stem for reduced-N.  相似文献

Heavy application of ammonium nitrogen to plant roots results in the conversion of ammonium nitrogen to the nitrogen of amides, glutamine, and asparagine, which are stored in roots or translocated to shoot. Since the net synthesis of such amides requires the supply of corresponding carbon skeletons, the carbon metabolism in amide synthesis in response to ammonium supply was investigated in tomato and wheat roots. The content of major primary amino acids was determined in tomato and wheat roots during a 4-d period of ammonium nutrition after 1-d culture in nitrogen-free nutrient solution. Ammonium supply led to a continuous increase in the asparagine content in wheat roots, whereas in tomato roots, the glutamine content increased 1 d after ammonium supply and thereafter the glutamine content was higher than the asparagine content. The amounts of amino acids synthesized from glucose-14C increased while the amounts of organic acids decreased in tomato roots by the supply of ammonium nitrogen for 1 d, compared to the roots that did not receive nitrogen. In tomato roots, the proportion of labeled glutamine was higher than that of labeled asparagine and the C5 amino acids were more strongly labeled than the C4 amino acids. These findings were different from the previous ones in wheat roots where the proportion of asparagine was found to be extremely high (Koga and Ikeda 2000: J. Fac. Agr. Kyushu Univ45, 7–13). To examine the in vivo asparagine synthesis, aspartate-'4C was fed to the roots. The labeling of asparagine, which was the most strongly labeled amino acid among the free amino acids, was remarkably strong in wheat roots whereas the labeling of glutamine was also pronounced in tomato roots. These results indicate that the ability to replenish carbon skeletons for amide synthesis in ammonium nutrition is different between tomato and wheat roots.  相似文献

ABSTRACT:   We have cloned a cDNA encoding the matrix metalloproteinase (MMP) from oyster Crassostrea gigas . The clone contains a 1797 bp open reading frame encoding a protein of 599 amino acids. Oyster MMP (Cg-MMP) has a transmembrane domain at its C-terminal and a furin/prohormone convertase cleavage site at the end of a propeptide domain, which are commonly observed in membrane-type MMPs (MT-MMPs). This suggests that Cg-MMP is an MT-MMP. The deduced amino acid sequence of oyster MMP shares approximately 30% identity with human MT4-MMP and MMPs from fruit fly and hydra. Cg-MMP mRNA was detected in the gill, mantle and adductor muscle, and more intense signals in the northern blot analysis were recognized in the gill and adductor muscle. Similar tissue distribution was observed for tissue inhibitor of metalloproteinase (Cg-TIMP) in oyster. In response to hypoxic stress, the abundance of Cg-MMP mRNA was elevated in the gill, while that of Cg-TIMP mRNA remained almost constant. These findings suggest that promotion of collagen metabolism may be implicated in the hypoxic adaptation in oyster.  相似文献
