In sap from plants of lisianthus (Eustoma grandiflorum) with necrotic stunt symptoms in Yamaguchi Prefecture in Japan, spherical particles with a diameter of about 30 nm were observed with transmission electron microscopy. We sequenced the complete genome of an isolate using the tombusvirus-specific primers and found that it shared about 96 % nucleotide identities with Moroccan pepper virus (MPV) in the genus Tombusvirus. The isolate reproduced necrotic stunt symptoms on lisianthus plants after mechanical inoculation. This is the first report of MPV on lisianthus in Japan. 相似文献

Alfalfa mosaic virus (AMV), Cucumber mosaic virus (CMV), Potato virus Y (PVY), Tomato bushy stunt virus nipplefruit strain (TBSV-Nf), and an unknown spherical virus were isolated from nipplefruit (Solanum mammosum) cultivated in Chiba Prefecture, Japan. The spherical virus was identified as Grapevine Algerian latent virus nipplefruit strain (GALV-Nf) from the genus Tombusvirus, based on its physical properties, serological relationships, and analysis of genomic RNA. The genomic RNA of GALV-Nf is 4731
nucleotides long and encodes five open reading frames as well as those of other tombusviruses. Nipplefruit infected with GALV-Nf
had severe stunting, leaf deformation, and clear mosaic symptoms. This is the first report of an isolation of GALV in Japan.

The Shimokita Peninsula in Aomori Prefecture, Japan, which is inhabited by Japanese monkeys (Macaca fuscata), is the northernmost habitat for wild primates in the world. This study was the first to determine the conception dates of specific individuals and estimate the pregnancy rate of wild populations in this region. The pregnancy rate of animals aged 5 years or more at delivery was estimated to be 40.9% (27/66). Conception dates of each fetus were also estimated using a regression line of Pig-tail monkeys (Macaca nemestrina), which are taxonomically related to Japanese monkeys and have a similar physique. The conception dates were distributed across 90 days between September 24th and December 23rd, with a mean conception date of November 4th (SD=22.3 days, n=53). Using these findings, the mean birth date was estimated as April 25th, more than two weeks earlier than the mean birth date in previous research determined using direct observations carried out over the past 20 years ago. Global warming due to climate change is thought to be one of the main causes of this difference. 相似文献

High-velocity weakening of faults may drive fault motion during large earthquakes. Experiments on simulated faults in Carrara marble at slip rates up to 1.3 meters per second demonstrate that thermal decomposition of calcite due to frictional heating induces pronounced fault weakening with steady-state friction coefficients as low as 0.06. Decomposition produces particles of tens of nanometers in size, and the ultralow friction appears to be associated with the flash heating on an ultrafine decomposition product. Thus, thermal decomposition may be an important process for the dynamic weakening of faults. 相似文献

Sex-determining region Y-box 2 (SOX2)-positive cells are stem/progenitor cells in the adenohypophysis, comprising the anterior and intermediate lobes (AL and IL, respectively). The cells are located in the marginal cell layer (MCL) facing Rathke’s cleft (primary niche) and the parenchyma of the AL (secondary niche). We previously demonstrated in vitro that the tetraspanin superfamily CD9 and SOX2 double-positive (CD9/SOX2-positive) cells in the IL-side MCL migrate to the AL side and differentiate into hormone-producing and endothelial cells in the AL parenchyma. Here, we performed in vivo studies to evaluate the role of IL-side CD9/SOX2-positive cells in pregnancy, lactation, and treatment with diethylstilbestrol (DES; an estrogen analog) when an increased population of prolactin (PRL) cells was observed in the AL of the rat pituitary. The proportions of CD9/SOX2-, CD9/Ki67-, and PRL/TUNEL-positive cells decreased in the primary and secondary niches during pregnancy and DES treatment. In contrast, the number of CD9/PRL-positive cells increased in the AL-side MCL and AL parenchyma during pregnancy and during DES treatment. The proportion of PRL/Ki67-positive cells increased in the AL-side MCL and AL parenchyma in response to DES treatment. Next, we isolated CD9-positive cells from the IL-side MCL using an anti-CD9 antibody. During cell culture, the cells formed free-floating three-dimensional clusters (pituispheres). Furthermore, CD9-positive cells in the pituisphere differentiated into PRL cells, and their differentiation potential was promoted by DES. These findings suggest that CD9/SOX2-positive cells in the IL-side MCL may act as adult stem cells in the AL parenchyma that supply PRL cells under the influence of estrogen. 相似文献

For soil water and solute transport research, time domain reflectometry (TDR) has been commonly used since its introduction to soil research in 1980 by Topp and his colleagues. Although TDR seems to be very versatile for laboratory and field experiments, it requires modest user skills for accurate measurements and the instrument itself is still expensive. A new soil moisture and electrical conductivity (EC) sensor was recently developed at a reduced cost. The new sensor independently measures soil water content, and soil bulk electrical conductivity using frequency phase-shift techniques, and soil temperatures using a thermistor. We evaluated the new sensor for measurements of water content, and EC using three different sensors in variably saturated soils. There was little deviation among the units for water content measurement except at near saturation. Separate exponential curves provided good calibrations for all the soils used in the full range of water content between air-dry and saturation. In saline water up to 54 mS m?1, the mV output in the EC measurement mode was linearly related to solution EC. Although slopes were quite similar among the three sensors, the intercepts differed from the manufacturer’s calibration. The calibrations for volumetric water content and electrical conductivity provided by the manufacturer were not good enough for any use. The temperature effect on volumetric water content could be negligible or easily compensated with a simple formula. Little EC dependency of the sensor on volumetric water content was observed. 相似文献

Serratia marcescens YS-1, a chitin-degrading microorganism, produced mainly N-acetylhexosaminidase. The purified enzyme had an optimal pH of approximately 8-9 and remained stable at 40 degrees C for 60 min at pH 6-8. The optimum temperature was around 50 degrees C, and enzyme activity was relatively stable below 50 degrees C. YS-1 N-acetylhexosaminidase hydrolyzed p-nitrophenyl beta-N-acetylgalactosamide by 28.1% relative to p-nitrophenyl beta-N-acetylglucosamide. The N-acetylchitooligosaccharides were hydrolyzed more rapidly, but the cellobiose and chitobiose of disaccharides that had the same beta-1,4 glycosidic bond as di-N-acetylchitobiose were not hydrolyzed. YS-1 N-acetylhexosaminidase efficiently transferred the N-acetylglucosamine residue from di-N-acetylchitobiose (substrate) to alcohols (acceptor). The ratio of transfer to methanol increased to 86% in a reaction with 32% methanol. N-Acetylglucosamine was transferred to the hydroxyl group at C1 of monoalcohols. A dialcohol was used as an acceptor when the carbon number was more than 4 and a hydroxyl group existed on each of the two outside carbons. Sugar alcohols with hydroxyl groups in all carbon positions were not proper acceptors. 相似文献

Here, we conducted a molecular phylogenetic analysis of 16 isolates of O. bornovanus taken from soil in which Cucurbitaceae plants had been grown in various locations throughout Japan. Changes in follicle-stimulating hormone (FSH), luteinizing hormone (LH), prolactin,
immunoreactive(ir)-inhibin, testosterone, estradiol-17β, and insulin-like growth factor
(IGF)-I in Thoroughbred stallions along with changes in prolactin secretion in geldings
were studied. The correlations of day-length with changes in the concentrations of these
hormones were also studied. Five stallions and thirteen geldings were employed to draw
blood samples in monthly basis and radioimmunoassay was performed to measure these
hormones. All hormones showed a seasonal pattern, the levels being highest during the
breeding season and lowest during the winter months. Most of the hormones were at their
highest concentration during the month of April, the mid of spring in northern hemisphere.
The concentration of circulating IGF-I also demonstrated seasonality, the peak lying on
the month of April. The plasma concentration of prolactin also increased during the
breeding season. This phenomenon was similar both in stallions and geldings although
geldings had lower concentration than that of stallions. The changes in concentration of
prolactin in stallions and geldings correlated more towards the day-length than towards
the temperature. These results clearly indicate the seasonality of pituitary and gonadal
hormones of Thoroughbred stallions, the activity being highest during the month of April
