Primary degenerative joint disease of the shoulder in a colony of beagles   总被引:1,自引:0,他引:1  
Shoulder joints of 149 Beagles over 8 years old at the time of death (mean age, 13.8 years +/- 3.21), were examined radiographically throughout their life-times for the frequency of degenerative joint disease (DJD). Clinical histories revealed no underlying cause for DJD. The shoulder joints of a subgroup of 18 dogs were examined at necropsy, and thin sections of the joints were evaluated radiographically and histologically. Serial clinical radiographic studies indicated that normal shoulder joint development during the first year of life was followed by the appearance of subchondral bone sclerosis and bony remodeling of normal joint contour, and by the formation of periarticular osteophytes and enthesiophytes. All changes were progressive with age and typical for DJD in dogs. Bilateral involvement was common. Evaluation of specimens obtained at necropsy revealed: articular cartilage change with roughening of the surface layer, degeneration and death of superficial chondrocytes, exposure of deeper layers of chondrocytes that had proliferated with fissuring of the damaged cartilage, total cartilage loss with polishing of the exposed subchondral bone, mixed patterns of subchondral bone sclerosis and osteoporosis, change in contour of the articular surfaces, and formation of periarticular osteophytes and enthesiophytes. Joint capsule thickening, synovitis, pannus formation, and synovial chondroma formation were observed. Because of the available clinical information, in addition to the typical changes of DJD, it was thought that the changes were primary. Instability appeared to play a role in the pathogenesis of the joint disease described; however, it was not clear whether the instability caused abnormal forces on healthy cartilage or whether the primary cartilage wear caused the instability.  相似文献

Chondroitin sulfate (CS) isomers, 6-sulfate (CS6) and 4-sulfate (CS4), change their ratio to each other in cartilaginous tissues with aging. In this study, a quantitative measurement method of CS6 and CS4 was developed, using capillary electrophoresis (CE). Various buffer solutions, pH, and digestion times were studied, and the use of 0.1 M Tris-HCl at pH of 8.0 allowed the isolation of CS6 and CS4 from CS most efficiently when combined with chondrotinase ABC at a concentration of 1 mU/microg of the substrate during a 3 hr digestion period. Amounts of newly synthesized CS6 and CS4 in the intervertebral disk chondrocyte three-dimensional culture were quantified by this method after the proteoglycans were extracted by equilibrium density centrifugation.  相似文献

To predict the performance of coppice forests with Japanese oak (Quercus mongolica var. crispula) in future changing environment, we studied the growth, photosynthesis, and powdery mildew (Erysiphe alphitoides) infection of sprouts of Japanese oak under free-air CO2 enrichment. Elevated CO2 reduced powdery mildew infection in both leaves of the shoot emerged in spring (1st flush) and the lammas and proleptic shoots (2nd flush) of sprouts. We observed significant increase in the net photosynthetic rate at growth CO2 concentration (i.e., 370 and 500 μmol mol?1 for ambient and elevated CO2 treatments, respectively) in both 1st and 2nd flush leaves of sprouts grown under elevated CO2. On the other hand, no significant increase in net photosynthetic rate under elevated CO2 was found before cutting. The photosynthetic activity of 2nd flush leaves in the sprouts under ambient condition was greatly reduced by severe infection to powdery mildew. Growth of sprouts was enhanced in the elevated CO2 condition. We conclude the growth enhancement in Japanese oak sprouts under elevated CO2 in the present study was achieved not only by physiological response (i.e., photosynthetic stimulation) but also by disease interaction.  相似文献

The effectiveness of air injection for preventing the blowout of particleboards manufactured using a radio-frequency hot press was investigated by evaluating the board properties under artificially created conditions that were conducive to blowout. For evaluation, 10-mm-thick boards with densities of 0.7 and 0.8 g/cm3 and 20-mm-thick boards with a density of 0.7 g/cm3 were manufactured. Pressing times for the 10-mm-thick boards were 2, 4, 6, and 8 min, and those for the 20-mm-thick boards were 4, 6, 8, and 10 min. Without air injection, blowout occurred in all manufactured boards. With air injection, however, blowout did not occur in the 10-mm-thick boards with a density of 0.7 g/cm3. Moreover, air injection prevented blowout even when the board density and board thickness were increased to 0.8 g/cm3 (for 10-mm-thick boards) and 20 mm (the density was kept at 0.7 g/cm3), respectively. Air-injection radio-frequency pressing reduced the pressing time from 4 to 2 min for 10-mm-thick boards, and from 6 to 4 min for 20-mm-thick boards. Moreover, this reduction in the pressing time was achieved without a large reduction in the internal bond strength of the boards.  相似文献

A hot press was used to manufacture particleboards (H boards). A radio-frequency hot press (for RH boards) and an air-injection radio-frequency hot press (for ARH board) were also used, and the effects of air injection on preventing blowout and board properties were analyzed. The thicknesses and densities of manufactured boards were 10 and 30 mm, and 0.6, 0.7, and 0.8 g/cm3, respectively. The investigation ascertained the effects of air injection in preventing blowout when a radio-frequency hot press is used. The increasing order of temperature was ARH board > RH board > H board during the final pressing stage. For the 30-mm-thick boards, the temperature of H board increased to 100 °C and remained constant at 100 °C even when the pressing time was extended. The temperature of the RH board increased to 100 °C more quickly than in the case of the H board and remained constant at 110–118 °C. The temperature of the ARH board increased linearly to 130–142 °C. For both the 10- and 30-mm-thick boards, the internal bond strength of the RH board was almost the same as that of the ARH board at densities of 0.6 and 0.7 g/cm3. In contrast, the internal bond strength of the RH board was lower than that of the ARH board at a density of 0.8 g/cm3. For the 10-mm-thick boards, the thickness swelling in the RH board was almost the same as that in the ARH board irrespective of the density. However, for the 30-mm-thick boards, the thickness swelling in the RH board was higher than that in the ARH board. The low plasticization of particles due to air injection presumably results in a high degree of thickness swelling.  相似文献

Ovalbumin (OVA) was phosphorylated by dry-heating in the presence of pyrophosphate at pH 4.0 and 85 degrees C for 1 and 5 days, and the physicochemical and structural properties of phosphorylated OVA were investigated. The phosphorus content of OVA increased to 1.01% by phosphorylation, and the electrophoretic mobility of PP-OVA also increased. Although the solubility of dry-heated OVA decreased, the decrease was slightly depressed by phosphorylation. The circular dichroism spectra showed that the change of the secondary structure in the OVA molecule, as measured by alpha-helix content, was mild by phosphorylation. The exchange reaction between the sulfhydryl and disulfide groups was enhanced and the surface hydrophobicity of OVA increased by phosphorylation. The tryptophan fluorescence intensity of OVA decreased by phosphorylation, suggesting that the conformational change occurred in the OVA molecule by phosphorylation. Although the differential scanning calorimetry thermograms of OVA showed a lowering of the denaturation temperature from 78.3 to 70.1 degrees C by phosphorylation, the stability of OVA against heat-induced insolubility at pH 7.0 was improved. The results indicated molten (partially unfolded) conformations of OVA formed by dry-heating in the presence of pyrophosphate.  相似文献

The hybrid larch F(1) (Larix gmelinii var. japonica × Larix kaempferi) is considered one of the most important tree species not only for timber production but also as an afforestation material for severe conditions such as infertile soil. To predict the ability of hybrid larch F(1) as an afforestation material under potential climates in the future, it is important to understand the response of hybrid larch F(1) to elevated CO(2) concentration ([CO(2)]) under low nutrient availability. Three-year-old seedlings of hybrid larch F(1) were grown under two different levels of [CO(2)], 360 (ambient) and 720 μmol mol(-1) (elevated), in combination with two different levels of nitrogen (N) supply (0 and 30 kg ha(-1)) for one growing season. Elevated [CO(2)] reduced the maximum rates of carboxylation and electron transport in the needles. Net photosynthetic rates at growth [CO(2)] (i.e., 360 and 720 μmol mol(-1) for ambient and elevated treatment, respectively) did not differ between the two CO(2) treatments. Reductions in N content and N use efficiency to perform photosynthetic functions owing to the deficiency of nutrients other than N, such as P and K, and/or increase in cell wall mass were considered factors of photosynthetic down-regulation under elevated [CO(2)], whereas stomatal closure little affected the photosynthetic down-regulation. Although we observed strong down-regulation of photosynthesis, the dry matter increase of hybrid larch F(1) seedlings was enhanced under elevated [CO(2)]. This is mainly attributable to the increase in the amount of needles on increasing the number of sylleptic branches. These results suggest that elevated CO(2) may increase the growth of hybrid larch F(1) even under low nutrient availability, and that this increase may be regulated by changes in both crown architecture and needle photosynthesis, which is mainly affected not by stomatal limitation but by biochemical limitation.  相似文献

Forest fires affect both carbon (C) and nitrogen (N) cycling in forest ecosystems, and thereby influence the soil–atmosphere exchange of major greenhouse gases (GHGs): carbon dioxide (CO2), methane (CH4), and nitrous oxide (N2O). To determine changes in the soil GHG fluxes following a forest fire, we arranged a low-intensity surface fire in a white birch forest in northern Japan. We established three treatments, having four replications each: a control plot (CON), a burned plot (BURN), and a plot burned with removal of the resulting charcoal (BURN-CHA). Soil GHG fluxes and various properties of the soil were determined on four or five occasions during a period that spanned two growing seasons. We observed increased concentrations of ammonium-N (NH4-N) in BURN and BURN-CHA after the fire, while nitrate–N (NO3-N) concentration was only increased in BURN-CHA after the fire. The soil CO2 flux was significantly higher in CON than in BURN or BURN-CHA, but there was no difference in soil CH4 uptake between the three treatments. Moreover, the N2O flux from BURN-CHA soil was slightly greater than in CON or BURN. In BURN-CHA, the soil N2O flux peaked in August, but there was no peak in BURN. We found temporal correlations between soil GHG fluxes and soil variables, e.g. soil temperature or NO3-N. Our results suggest that environmental changes following fire, including the increased availability of N and the disappearance of the litter layer, have the potential to change soil GHG fluxes. Fire-produced charcoal could be significant in reducing soil N2O flux in temperate forests.  相似文献

We investigated susceptibility to photoinhibition in leaves acclimated to different light regimes in intermediately shade-tolerant Japanese oak (Quercus mongolica Fisch. ex Turcz. var. crispula (Blume) Ohashi) and shade-tolerant Japanese maple (Acer mono Maxim. var. glabrum (Lév. et Van't.) Hara), to elucidate adaptability to gap formation in leaves differing in shade acclimation. We hypothesized that there is a tradeoff between shade adaptation and capacity to mitigate photoinhibition associated with leaf morphology. We simultaneously measured chlorophyll fluorescence and gas exchange in seedlings that had been grown in full sunlight (open), 10% of full sun (moderate shade) and 5% of full sun (deep shade). Shade-tolerant A. mono adapted to deep shade through changes in leaf morphology, lowering its leaf mass per area (LMA), but Q. mongolica showed little change in LMA between moderate and deep shade. Photochemical quenching (qP) did not differ between species in full sunlight and moderate shade; however, in deep shade, qP of Q. mongolica was higher than that of A. mono, suggesting that Q. mongolica grown in deep shade is less susceptible to photoinhibition at gap formation. This is consistent with the finding that chronic photoinhibition 3 days after the transfer to full sunlight, indicated by the decrease in maximum photochemical efficiency, Fv/Fm, at predawn, was less in deep-shade-grown Q. mongolica than in deep shade-grown A. mono. In deep shade, the electron transport rate (ETR) of Q. mongolica was higher than that of A. mono, whereas thermal energy dissipation through photosystem II antennae, indicated by non-photochemical quenching, was lower in Q. mongolica than in A. mono. In deep shade, the greater ETR capacity in Q. mongolica in association with higher LMA and higher leaf N content could contribute to maintaining high qP and mitigating photoinhibition. These results indicate that, by maintaining a high electron transport capacity even in deep shade, the gap-dependent and intermediate-shade-tolerant Q. mongolica trades improved shade adaptation for higher growth potential when a gap event occurs.  相似文献
