四倍体小麦矮秆基因的赤霉素敏感性及对农艺性状的影响   总被引:1,自引:0,他引:1  
为明确四倍体小麦矮秆基因Rht14、Rht16和Rht18的赤霉素敏感性及其对小麦农艺性状的影响,促进这些矮秆基因的合理利用。选用分别含有Rht14、Rht16和Rht18的四倍体小麦近等基因系ANW16D(Rht14)、ANW16F(Rht16)、ANW16G(Rht18)及其高秆轮回亲本LD222以及六倍体小麦中国春(Chinese spring),测量其不同浓度GA3处理下小麦的株高,计算赤霉素敏感系数(GRI)并推断3种矮秆小麦的赤霉素反应类型。在成熟期对LD222近等基因系小麦的农艺性状如株高、穗长、主穗穗下第一茎节(P-1)节间长、节间表皮细胞、种子表皮细胞及种子体积等进行测量,分析Rht14、Rht16、Rht18这3个矮秆基因对小麦这些农艺性状的效应。结果表明,ANW16D、ANW16F和ANW16G这3个矮秆小麦株高恢复到正常LD222株高的最适GA3浓度为10-4mol/L;3个矮秆品种均为赤霉素敏感型且敏感性大小为中国春ANW16GANW16FANW16DLD222;Rht14、Rht16、Rht18均是通过降低主穗穗下第一茎节(P-1)节间长度来使小麦株高降低,降低效应为Rht18Rht16Rht14;它们降低小麦株高的根本原因均是缩短了小麦节间表皮细胞长度且缩短效应与降低株高效应一致;3个矮秆基因均在降低小麦株高的同时不影响种子的体积。  相似文献

To investigate the pathological role of staphylococcal enterotoxins (SEs) and toxic shock syndrome toxin-1 (TSST-1) in bovine mastitis, the production of SEs and TSST-1 was investigated in staphylococci isolated from 120 mammary gland secretions (MGS, 51 from no clinical sign-mammary glands and 69 from staphylococcal mastitic-mammary glands) collected from dairy farms where staphylococcal mastitis frequently occurred in Miyagi and Yamagata prefectures from 1997 to 1998. Concentrations of these toxins and specific antibody titers in each MGS were also measured. Furthermore, SEC and TSST-1 were inoculated into lactating mammary glands and inflammatory responses were analyzed. A high percentage of staphylococci including Staphylococcus aureus and coagulase-negative staphylococci isolated from both no clinical sign- and mastitic-MGS produced both SEC and/or TSST-1. The concentration of SEC increased with the severity of the mastitis, and was significantly higher (P<0.05) in acute mastitic-than in no clinical signs-MGS. Titers of specific antibodies to TSST-1 in MGS were significantly higher (P<0.05) than those to SEC, regardless of whether or not the cows were lactating or mastitic. Specific antibodies purified from MGS neutralized each toxin in vitro. A significant increase (P < 0.05) in somatic cell counts was induced by the intramammary inoculation of SEC but not TSST-1. These findings indicated that SEC rather than TSST-1 plays an important role in the pathology of staphylococcal bovine mastitis. The inflammatory activity of TSST-1 was probably neutralized by specific antibodies in MGS.  相似文献

选取1953~2006年准葛尔盆地南缘石河子垦区3个地面观测站的资料,对比分析了沙尘暴、大风的时空分布特征,探讨沙尘暴和大风之间的关系及沙尘暴发生时的最多风向和最小风速指标。结果表明:准葛尔盆地南缘石河子垦区沙尘暴与大风的地理分布刚好相反,沙尘暴多的地方大风较少,北部莫索湾沙漠边缘一带为沙尘暴高发区;但有着基本一致的气候变化趋势。沙尘暴发生时的最多风向为西北风,引起沙尘暴的最小风速指标大于10m/s。  相似文献

采用培养皿法和常规生化方法,研究了新型除草剂唑嘧氯草胺(暂定名,代号:ZJ-2725)的作用机制。结果显示,同时添加20mg/L浓度的缬氨酸、亮氨酸和异亮氨酸能完全恢复唑嘧氯草胺对苘麻芽的生长抑制作用,而添加相同浓度的单一支链氨基酸只能部分消除其抑制作用。离体条件下,随着唑嘧氯草胺浓度的增大,对乙酰乳酸合成酶(ALS)活性的抑制率增加,ALS活性随反应时间的延长而增加,且两者之间符合Michaelis-Meten方程;活体条件下,唑嘧氯草胺对苘麻的ALS也表现出明显的抑制作用,ALS活性明显下降。表明唑嘧氯草胺在植物体内抑制了3种支链氨基酸的生物合成,进而导致植物蛋白质合成受阻而使植物生长受到抑制,ALS即为唑嘧氯草胺的作用靶标。  相似文献

Capsaicinoids are pungent compounds used for industrial and medical purposes including food, medicine and cosmetics. The Indian local variety ‘Bhut Jolokia’ (Capsicum chinense Jacq.) is one of the world's hottest chilli peppers. It produces more than one million Scoville heat units (SHUs) in total capsaicinoids. In this study, our goal was to identify quantitative trait loci (QTLs) responsible for the high content of capsaicin and dihydrocapsaicin in ‘Bhut Jolokia’. Capsicum annuum ‘NB1’, a Korean pepper inbred line containing 14 000 SHUs, was used as a maternal line. An F2 population derived by crossing between ‘NB1’ and ‘Bhut Jolokia’ was generated to map QTLs for capsaicinoids content. A total of 234 markers, including 201 HRM, 21 SSR, 2 CAPS and 10 gene‐based markers of the capsaicinoid synthesis pathway, were mapped. The final map covered a total distance of 1175.2 cM and contained 12 linkage groups corresponding to the basic chromosome number of chilli pepper. Capsaicin and dihydrocapsaicin content were analysed in 175 F2 pepper fruits using the HPLC method. The maximum total capsaicinoids content was 1389 mg per 100g DW (dry weight), and the minimum content was 11 mg per 100g DW. Two QTLs (qcap3.1 and qcap6.1) for capsaicin content were identified on LG3 and LG6, and two QTLs (qhdc2.1 and qdhc2.2) for dihydrocapsaicin content were located on LG2. We did not detect QTLs for total capsaicinoids content. The QTL positions for capsaicin content were different from those for dihydrocapsaicin content. These results indicate that the complexity of selecting for more pungent chilli peppers must be considered in a chilli pepper breeding programme. The QTL‐linked markers identified here will be helpful to develop more pungent pepper varieties from ‘Bhut Jolokia’, a very hot pepper.  相似文献

Salmonella Typhimurium ghost cells expressing K88ab, K88ac, K99, and FasA fimbriae of enterotoxigenic Escherichia coli (ETEC) in their envelopes were constructed. The genes encoding the fimbriae were individually cloned into an expression plasmid, pMMP81, carrying the asd gene, which was subsequently electroporated into the Δasd S. Typhimurium mutant. Plasmid pJHLP99, carrying the phiX174 lysis gene E, was also subsequently electroporated into the Salmonella mutant. The presence of the individual fimbriae on the ghost cells was examined by Western blot analysis. Forty BALB/c mice were equally divided into 2 groups of 20 mice each. Group A mice were intramuscularly vaccinated with a mixture of the 4 ghost cells expressing the individual fimbriae. The group B mice were inoculated with sterile phosphate-buffered saline as a control. The antigen-specific serum IgG concentrations were significantly higher in group A than in group B from week 2 until week 6 after inoculation. In addition, the antigen-specific IgA concentrations in fecal samples were significantly higher in group A than in group B at week 2 after inoculation. A large difference between the groups in the number of antigen-specific IgA-secreting cells in the small intestine was observed by immunohistochemical study. Also, the splenic lymphocyte proliferative responses were significantly greater in group A than in the control mice. These results suggest that vaccination with our Salmonella ghost cells can induce both humoral and cell-mediated immune responses and that the increased number of antigen-specific IgA-secreting cells in the small intestine may be correlated with the elevated fecal IgA immune response.  相似文献

【目的】探明不同用量生物质炭对草坪土壤微生物多样性特征的影响,为生物质在草坪土壤中的应用提供理论依据。【方法】以草坪土壤为研究对象,以不施用生物质碳为对照,设8、20、40和200 kg/m2共4个生物质炭用量处理,对照和4个处理分别标记为BC0、BC8、BC20、BC40和BC200。播种5个月后采集0~30 cm土壤,测定土壤有机碳、全氮、水溶性碳和微生物量碳含量及土壤微生物碳代谢群结构,分析不同用量生物质炭对草坪土壤微生物碳代谢群落的影响。【结果】与对照BC0相比,施用生物质炭处理的土壤全氮含量显著增加(P<0.05,下同),BC40和BC200处理的土壤有机碳显著增加,BC200处理的水溶性碳显著增加,而土壤微生物量碳无显著变化(P>0.05)。BC20处理的土壤平均光密度(AWCD)值较对照BC0提高12.4%,BC200处理的AWCD值则较对照降低51.3%。BC20处理增加了生物群落结构AWCD值,利用胺类物质微生物活性显著提高;BC200处理降低了土壤微生物数量,对利用多聚物类物质微生物的负面影响最小。主成分分析结果表明,第一主成分可解释变异的86.06%,主要为代谢吐温40、吐温80、L-丝氨酸、D-半乳糖酸内脂、D,L-α-磷酸甘油和4-羟基苯甲酸等六种碳源物质引起的,能有效区分适量(BC8、BC20、BC40)和过量(BC200)施用生物质炭含量处理的草坪土壤。相关分析结果表明,土壤微生物量碳与i-赤藓糖醇的光密度值呈显著正相关;土壤有机碳和水溶性碳含量与L-精氨酸、L-丝氨酸等多种碳源的光密度值呈显著或极显著(P<0.01)负相关。【结论】适量施用生物质炭可提高草坪土壤的微生物数量和活性。施用生物质炭的草坪土壤利用L-精氨酸、L-天门冬酰胺的能力较强,但对利用L-苏氨酸、i-赤藓糖醇、D-木糖和2-羟基苯甲酸等碳源基质的利用能力较弱。建议在种植草坪草时,生物质炭的施用量控制在20 kg/m2以内。  相似文献

针对苹果采摘机器人中果实姿态信息缺失造成果实损伤及采摘失败等问题,根据苹果尾部花萼遗迹区域是否可见分析了果实姿态信息机器视觉测量方法,研究了果实姿态信息的粒子滤波估计。将单目摄像机固定在机器人末端手爪上,在手爪趋近果实的采摘过程中,随机采集多帧果实图像。对每帧图像运用惯性主轴或重心点偏移两种方法计算果实在机器人坐标系下的姿态向量值,并通过粒子滤波融合来得到果实姿态信息的最优估计。实验结果表明,粒子滤波方法可以有效地融合多帧图像的苹果姿态信息,能够减小因依靠单帧图像直接测量而产生的误差。  相似文献

对脂肪酶Novozym 435催化高酸值米糠油酯化脱酸进行了研究.利用响应面分析法(RSM)优化脱酸条件.根据Box-Benhnken中心组合试验设计原理对试验条件进行优化,在分析各因素显著性及其交互作用的基础上,得出米糠油酯化脱酸最佳工艺参数为:酶质量分数1.1%、甘油添加量0.42 mg、温度56.4℃、反应时间23.2 h.实际测得脱酸后米糠油酸值(KOH)由56 mg/g降为5.04 mg/g,游离脂肪酸酯化率达到91%,与模型预测值基本相符.  相似文献
