Cotton blue disease (CBD) is the most important disease present in cotton crops in South America and cotton leafroll dwarf virus (CLRDV) is the causal agent. The disease has been controlled by sowing cotton varieties resistant to CLRDV. However, in the 2009/10 growing season, an outbreak due to an atypical CLRDV isolate (CLRDV-at) occurred in northwest Argentina. Although CLRDV and CLRDV-at genomes are very closely related, the symptoms they produce in cotton plants are quite different. P0 is the most divergent protein between the isolates and in CLRDV is a silencing suppressor protein. This work characterized the silencing suppressor activity of the P0 protein encoded by CLRDV-at (P0CL-at) and evaluated its role in Cbd-resistance break in cotton plants. It was demonstrated that P0CL-at, despite having a mutation in the consensus of the F-box-like motif, was able to suppress local RNA silencing, but displayed lower activity than P0CL. P0CL and P0CL-at showed no differences in the interaction with Gossypium hirsutum SKP1 orthologue (GSK1) and Nicotiana benthamiana SKP1 and both P0 proteins triggered destabilization of ARGONAUTE1. However, when the ability to enhance PVX symptoms was evaluated, P0CL-at was shown to be a weaker pathogenicity factor than P0CL in N. benthamiana. Interestingly, trans-expressed P0CL-at enabled CLRDV to systemically infect CBD-resistant plants, and a chimeric CLRDV-P0CL-at infectious clone succeeded in establishing infection in CBD-resistant cotton varieties with symptoms resembling those produced by CLRDV-at. These results strongly suggest that P0CL-at is the avirulence (Avr) determinant involved in breaking cotton Cbd gene-based resistance. 相似文献

Fungal mycotoxins are important contaminants of agricultural commodities that pose serious concerns to producers, consumers and exporters. Aflatoxin is a carcinogenic metabolite produced by the fungi Aspergillus flavus and A. parasiticus. These fungi and other mould species living in the soil contaminate several crops including maize. This study was undertaken with the following objectives: (i) to assess the presence of A. flavus and other moulds in Ghanaian soil; (ii) to determine the distribution and density of A. flavus under three agro-ecologies noted for major maize production; and (iii) to assess the effect of percentage of carbon (C) and nitrogen (N) and soil pH on A. flavus population densities and diversity in the isolates found across the agro-ecologies. The data showed seven moulds that were common across the agro-ecologies. Significant differences (P < 0.05) were observed in the A. flavus density and distribution within and across these agro-ecologies. Fumesua soils recorded the highest levels of A. flavus (1.185 × 103 cfu g−1) while Akomadan recorded the least (9.76 × 102 cfu g−1). Percentage available C, N and soil pH did not significantly influence A. flavus density. The A. flavus isolates identified in this study varied in genetic sequence within the aflatoxin gene cluster, but these differences were not distinguishable by origin. 相似文献

Tomato is the most economically important fruit/vegetable crop grown worldwide. However, viral diseases remain an important factor limiting its productivity, with estimated quantitative and qualitative yield losses in tomato crops often reaching up to 100%. Many viruses infecting tomato have been reported, while new viral diseases have also emerged. The climatic changes the world is experiencing can be a contributing factor to the successful spread of newly emerging viruses, as well as the establishment of disease in areas that were previously either unfavourable or where the disease was absent. Because antiviral products are not available, strategies to mitigate viral diseases rely on genetic resistance/tolerance to infection, control of vectors, improvement in crop hygiene, roguing of infected plants and seed certification. Tomato brown rugose fruit virus (ToBRFV) is an emerging viral threat to tomato productivity and is currently spreading into new areas, which is of great concern to the growing global production in the absence of mitigation measures. This review presents the current knowledge about ToBRFV and future prospects for an improved understanding of the virus, which will be needed to support effective control and mitigation of the impact it is likely to cause. 相似文献

BackgroundBone foreign bodies are commonly encountered in small animal practice. Esophageal bone foreign bodies (E‐bFBs) warrant removal, whereas gastric bone foreign bodies might not.ObjectivesDescribe management and outcomes for dogs with esophageal or gastric bone foreign bodies.AnimalsOne hundred twenty‐nine dogs with esophageal (n = 45) or gastric (n = 84) bone foreign bodies.MethodsRetrospective review of medical records.ResultsDogs with E‐bFBs were younger than dogs with gastric bone foreign bodies (median age esophageal, 4 years [IQR 2‐8]; median age gastric, 6 years [IQR 3‐10]; P = .03), and had a higher bone cross‐sectional area relative to body weight (median esophageal, 98.21 mm2/kg [IQR 48.25‐142.6]; median gastric, 28.6 mm2/kg [IQR 17.25‐64.28]; P < .001). Forty‐two of 45 esophageal foreign bodies were resolved non‐surgically and 3 by esophagotomy. Esophageal erosions were more likely with distal entrapment (OR 12.88, [95% CI 31.95‐129.29], P = .01) and longer duration (OR 18.82 [95% CI 2.22‐273.97], P = .01). Sixty‐two of 84 bone gastric foreign bodies were left in situ. Endoscopic removal was successful in 20 of 22 (91%; 95% CI 70‐99) attempts.Conclusions and Clinical ImportanceWhile all E‐bFBs were dislodged either by advancement into the stomach, endoscopic removal, or esophagotomy, the majority of gastric bone foreign bodies were left in situ for dissolution, with no reported complications. Gastric advancement of E‐bFBs should be considered when oral removal is not feasible, and dissolution can be considered even with large bones. 相似文献

为了探究低温等离子体(Non-Thermal Plasma,NTP)耦合催化剂对粮仓中磷化氢(PH3)的去除效果,该文采用介质阻挡放电低温等离子体与催化剂耦合的反应装置,研究了NTP与Fe2O3催化剂耦合对PH3去除率的影响,并对耦合反应后的催化剂进行表征,分析产物及反应机理。结果表明,载体和催化剂与NTP耦合改变了NTP的放电状态,提高了PH3去除率;不同载体负载Fe2O3后,在输入功率超过53 W时,NTP与Fe2O3催化剂耦合对PH3去除率均高于无填充,Fe2O3/Al2O3与NTP耦合效果最好,去除效率达到了100%。耦合前后催化剂的表征结果表明,耦合反应之后,Fe2O3/Al2O3催化剂的比表面积降低了11.23%,Fe3+/(Fe2++Fe3+)含量提高了21.06%,Osur/(Olatt+Osur+Oads)含量减少了2.24%,Fe含量降低了4.69%,P含量增加了4.34倍。产物分析表明耦合后生成的主要产物为磷酸。研究结果表明NTP耦合Fe2O3/Al2O3催化剂具有很好的PH3去除效果,为粮食仓储行业去除磷化氢提供理论依据。 相似文献

Abstract. In 1969, a new kind of epizootic occurred among eels in Japan and virological investigation was initiated. A new virus designated eel virus european (EVE) was isolated and biological, cytological, serological tests and infectivity trials were carried out. In its CPE on RTG-2 cells and additionally in its biological properties and particle size, EVE was found to be similar to infectious pancreatic necrosis virus (IPNV). Serologically, EVE was similar to the ď Honnincthun, France, strain of IPNV. However, infectivity trials showed that EVE and IPNV differed; EVE killed Japanese eels Anguilla japonica but not rainbow trout fry Salmo gairdneri , while IPN virus killed rainbow trout fry but not eels. We consider EVE to be the primary agent causing the new epizootic and propose the name viral kidney disease for the resulting clinical condition. 相似文献

Histochemical localization of proteolytic activities in the dorsal epidermis of Japanese eel was demonstrated by fluorescent microscopy utilizing 4-methoxy-2-naphthylamide (4M$\beta$NA) derivatives as substrates and 5-nitrosalicylaldehyde as a trapping agent. Carbobenzoxy-L-phenylalanyl-L-arginyl-4M$\beta$NA (Cbz-Phe-Arg-4M$\beta$NA) and Cbz-Arg-Arg-4M$\beta$NA were used for direct detection of cathepsins L and B activities, respectively, in fresh frozen sections and unfixed cells of the eel epidermis. The fluorescing areas, where Cbz-Phe-Arg-4M$\beta$NA was hydrolyzed by cathepsin L, were shown in mucus secretory cells and club cells and broadly around skin surface. The fluorescing areas due to Cbz-Arg-Arg-4M$\beta$NA hydrolysis by cathepsin B were localized similarly in these tissues. The fluorescing intensity for both catheptic activities in mucus secretory cells was higher than that in club cells, where small fluorescing granules were distributed. These results indicate that eel cathepsins L and B are stored in epidermal secretory cells at different levels and probably serve as defense factors before or after secretion by these cells. The effect of temperature on the food consumption rate and the digestive enzyme activities of Clarias batrachus (80.60 ± 5.34 g) were evaluated. Fish were exposed to six different temperatures of 10, 15, 20, 25, 30 and 35 °C following an acclimation temperature of 25 °C. The rate of temperature change was 2 °C day?1. Highest food consumption was recorded at 25 °C. It gradually reduced with decreasing water temperature. Food consumption rate was significantly (P < 0.05) lower at 10 °C compared with other treatments. Hence, 46.67, 8.20–23.58 and 1.02–6.15% reduced food consumptions were recorded in groups exposed at 10, 15 and 20 °C temperatures, respectively, compared with the 25 °C. The consumption rate was not affected in fish exposed at 30 and 35 °C. Total protease, trypsin and chymotrypsin activities were significantly (P < 0.05) higher in fish exposed at 25 °C compared with others. Lipase activity was significantly (P < 0.05) higher in fish exposed at 30 °C compared with others. Lowest enzyme activities were recorded at 10 °C. Water temperature below 25 °C affected the food consumption and digestive enzyme activities in fish that served as indicators of stress in fish. 相似文献
