Unchecked exploitation of wildlife resources is one of the major factors influencing species persistence throughout the world today. A significant consequence of exploitation is the increasing rate at which genetic diversity is lost as populations decline. Recent studies suggest that life history traits affecting population growth, particularly in long-lived species, may act to moderate the impact of population decline on genetic variation and lead to remnant populations that appear genetically diverse despite having passed through substantial demographic bottlenecks. In this study we show that the retention of genetic variation in a partially recovered population of Nile crocodile is deceptive, as it masks the reality of a significant decline in the population’s effective size (Ne). Repeated episodes of unchecked hunting in the mid to late 20th century have today led to a five-fold decrease in the population’s Ne. Using current census data we estimate the contemporary Ne/N ratio as 0.05 and, in light of quotas that permit the ongoing removal of adults, simulated the likely effects of genetic drift on extant levels of variation. Results indicate that even if the current effective size is maintained, both allelic diversity and heterozygosity will decline. Our findings have complex implications for long-lived species; an emphasis on the retention of genetic variation alone, whilst disregarding the effects of population decline on effective size, may ultimately obscure the continued decline and extinction of exploited populations.  相似文献

This study was conducted to improve our understanding of how earthworms and microorganisms interact in the decomposition of litter of low quality (high C : N ratio) grown under elevated atmospheric [CO2]. A microcosm approach was used to investigate the influence of endogeic earthworm (Aporrectodea caliginosa Savigny) activity on the decomposition of senescent Charlock mustard (Sinapis arvensis L.) litter produced under ambient and elevated [CO2]. Earthworms and microorganisms were exposed to litter which had changed in quality (C : N ratio) while growing under elevated [CO2]. After 50 d of incubation in microcosms, C mineralization (CO2 production) in the treatment with elevated‐[CO2] litter was significantly lower in comparison to the ambient‐[CO2] litter treatment. The input of Charlock mustard litter into the soil generally induced N immobilization and reduced N2O‐emission rates from soil. Earthworm activity enhanced CO2 production, but there was no relationship to litter quality. Although earthworm biomass was not affected by the lower quality of the elevated‐[CO2] litter, soil microbial biomass (Cmic, Nmic) was significantly decreased. Earthworms reduced Cmic and fungal biomass, the latter only in treatments without litter. Our study clearly showed that A. caliginosa used the litter grown under different [CO2] independent of its quality and that their effect on the litter‐decomposition process was also independent of litter quality. Soil microorganisms were shown to negatively react to small changes in Charlock mustard litter quality; therefore we expect that microbially mediated C and N cycling may change under future atmospheric [CO2].  相似文献

The measurement of antioxidant capacity in fruits differs from that of other biological samples due to their low pH and very low lipophilic antioxidant capacity. In this report, we present a modified 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) method for fruits and compare its performance with the other commonly used antioxidant methods of 2,2'-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP). The antioxidant capacity and reaction kinetics of four phenolic compounds, two antioxidant standards, and five fruits were also investigated. The modified ABTS method prepared at a pH of 4.5 with sodium acetate buffer is highly stable and easily applied to fruit samples as compared to the standard (pH 7.4) version. The measured antioxidant capacity of samples varied with the assay method used, pH, and time of reaction. Traditional antioxidant standards (trolox, ascorbic acid) displayed stable, simple reaction kinetics, which allowed end point analysis with all of assays. Of the phenolic compounds examined, chlorogenic and caffeic acids exhibited the most complex reaction kinetics and reaction rates that precluded end point analysis while gallic acid and quercetin reached stable end points. All fruit extracts exhibited complex and varied kinetics and required long reaction times to approach an end point. Because the antioxidant capacity of fruit extracts is a function of the array of individual antioxidants present, accurate comparisons among fruit samples require that reaction times be standardized and of sufficient length to reach steady state conditions and that more than one assay be used to describe the total antioxidant activity of fruit samples.  相似文献

Most tree species in tropical mountain rain forests are naturally associated with arbuscular mycorrhizal fungi. Previous studies in southern Ecuador of 115 tree species revealed that only three species were not associated with arbuscular mycorrhizal fungi. Seedlings of tropical tree species raised in the nursery may need to be associated with arbuscular mycorrhizal fungi to survive transplantation shock in higher numbers. Methods for establishing plantations with native tree species are not yet established for Ecuador. Thus, we investigated the feasibility of using mycorrhizal roots of seedlings of Inga acreana, Tabebuia chrysantha, Cedrela montana and Heliocarpus americanus that had trapped mycorrhizal fungi from forest humus in the nursery to inoculate C. montana and H. americanus with native arbuscular mycorrhizal fungi. Inoculation with either a mixture of mycorrhizal roots from the four species or only with mycorrhizal roots from the same tree species were compared with effects of moderate fertilization. Assessment of plant growth and mycorrhizal status of 6-months-old Cedrela montana and Heliocarpus americanus revealed an improvement in growth and diverse associated fungi through mycorrhizal root inoculation in comparison with moderate fertilization. Moderate fertilization did not suppress mycorrhization.  相似文献

The Andes of Ecuador are known for their outstanding biodiversity but also as the region with the highest deforestation rate in South America. This process is accompanied by accelerating degradation and loss of environmental services. Despite an extraordinary richness in native tree diversity, more than 90% of all forest plantations established in Ecuador consist of exotic species, primarily Eucalyptus spp. and Pinus spp. This is mainly due to the lack of information about the autecological and synecological requirements of the native species.  相似文献

Conifer woodlands expanding into sage-steppe (Artemisia spp.) are a threat to sagebrush obligate species including the imperiled greater sage-grouse (Centrocercus urophasianus). Conifer removal is accelerating rapidly despite a lack of empirical evidence to assess outcomes to grouse. Using a before-after-control-impact design, we evaluated short-term effects of conifer removal on nesting habitat use by monitoring 262 sage-grouse nests in the northern Great Basin during 2010–2014. Tree removal made available for nesting an additional 28% of the treatment landscape by expanding habitat an estimated 9603 ha (3201 ha [± 480 SE] annually). Relative probability of nesting in newly restored sites increased by 22% annually, and females were 43% more likely to nest within 1000 m of treatments. From 2011 (pretreatment) to 2014 (3 yr after treatments began), 29% of the marked population (9.5% [± 1.2 SE] annually) had shifted its nesting activities into mountain big sagebrush habitats that were cleared of encroaching conifer. Grouping treatments likely contributed to beneficial outcomes for grouse as individual removal projects averaged just 87 ha in size but cumulatively covered a fifth of the study area. Collaboratively identifying future priority watersheds and implementing treatments across public and private ownerships is vital to effectively restore the sage-steppe ecosystem for nesting sage-grouse.  相似文献

Background: Testing for canine blood types other than dog erythrocyte antigen 1.1 (DEA 1.1) is controversial and complicated by reagent availability and methodology. Objectives: The objectives of this study were to use available gel column technology to develop an extended blood‐typing method using polyclonal reagents for DEA 1.1, 1.2, 3, 4, 7, and Dal and to assess the use of gel columns for cross‐matching. Methods: Dogs (43–75) were typed for DEA 1.1, 1.2, 3, 4, 7, and Dal. Methods included tube agglutination (Tube) using polyclonal reagents, a commercially available DEA 1.1 gel column test kit (Standard‐Gel) using monoclonal reagent, and multiple gel columns (Extended‐Gel) using polyclonal reagents. Blood from 10 recipient and 15 donor dogs was typed as described above and cross‐matched using the gel column technique. Results: Of 43 dogs typed for DEA 1.1, 23, 25, and 20 dogs were positive using Standard‐Gel, Extended‐Gel, and Tube, respectively. Typing for DEA 1.2 was not achievable with Extended‐Gel. For 75 dogs typed for DEA 3, 4, and 7, concordance of Extended‐Gel with Tube was 94.7%, 100%, and 84%, respectively. Dal, determined only by Extended‐Gel, was positive for all dogs. Post‐transfusion major cross‐matches were incompatible in 10 of 14 pairings, but none were associated with demonstrable blood type incompatibilities. Conclusions: Gel column methodology can be adapted for use with polyclonal reagents for detecting DEA 1.1, 3, 4, 7, and Dal. Agglutination reactions are similar between Extended‐Gel and Tube, but are more easily interpreted with Extended‐Gel. When using gel columns for cross‐matching, incompatible blood cross‐matches can be detected following sensitization by transfusion, although in this study incompatibilities associated with any tested DEA or Dal antigens were not found.  相似文献

Wetland ecosystems provide an optimum natural environment for the sequestration and long-term storage of carbon dioxide (CO2) from the atmosphere, yet are natural sources of greenhouse gases emissions, especially methane. We illustrate that most wetlands, when carbon sequestration is compared to methane emissions, do not have 25 times more CO2 sequestration than methane emissions; therefore, to many landscape managers and non specialists, most wetlands would be considered by some to be sources of climate warming or net radiative forcing. We show by dynamic modeling of carbon flux results from seven detailed studies by us of temperate and tropical wetlands and from 14 other wetland studies by others that methane emissions become unimportant within 300 years compared to carbon sequestration in wetlands. Within that time frame or less, most wetlands become both net carbon and radiative sinks. Furthermore, we estimate that the world’s wetlands, despite being only about 5–8 % of the terrestrial landscape, may currently be net carbon sinks of about 830 Tg/year of carbon with an average of 118 g-C m?2 year?1 of net carbon retention. Most of that carbon retention occurs in tropical/subtropical wetlands. We demonstrate that almost all wetlands are net radiative sinks when balancing carbon sequestration and methane emissions and conclude that wetlands can be created and restored to provide C sequestration and other ecosystem services without great concern of creating net radiative sources on the climate due to methane emissions.  相似文献
