In a comparison of four parenteral copper preparations for sheep administered at similar dose rates, copper as diethylamine oxyquinoline sulphonate (copper DOS) was found to alleviate hypocupraemia in ewes more consistently than copper calcium EDTA and copper-methionate, given in a cream (C) or aqueous base (A). In similar comparisons of preparations for cattle, copper calcium EDTA was 19 per cent more effective than copper DOS and 36 to 48 per cent more effective than copper methionate (A). Methionate preparations were characterised by marked reactions at and slow translocation from the site of injection whereas copper DOS was rapidly translocated and gave little or no tissue reaction in sheep or cattle. Attributes of an improved parenteral copper preparation are discussed in the light of disadvantages of existing preparations.  相似文献

Some effects of copper deficiency on leucocyte function in sheep and cattle   总被引:6,自引:0,他引:6  
Peripheral blood leucocytes from copper-deficient (less than 8 mumol per litre plasma) and copper-sufficient (more than 8 mumol per litre) ewes and lambs were tested for their ability to ingest and kill Candida albicans in vitro. Killing capacity was significantly reduced in both copper-deficient ewes and lambs, although phagocytic capacity was apparently unimpaired. Similar results were obtained with copper-deficient calves, in which it was further shown that copper repletion restored candidacidal activity. Leucocyte and erythrocyte superoxide dismutase (LSOD and ESOD respectively) activities were both significantly decreased in hypocupraemic ewes and there was a linear relationship between their activities in the two cell types: LSOD activities were reduced by approximately 35 per cent and ESOD activities by 67 per cent. Cyanide inhibition tests indicated that the manganese-dependent form of the enzyme contributed little to the total activity of LSOD or ESOD. There was a tendency for leucocytes from copper-deficient ewes to release greater amounts of superoxide anion after stimulation with opsonised zymosan. Their viability was similar to that of leucocytes from control sheep and was unaffected for up to three hours after stimulation.  相似文献

At harvest, potato (Solanum tuberosum L.) tubers are dormant and will not sprout. As the period of postharvest storage is extended, tuber dormancy is broken and sprout growth commences. The loss of tuber dormancy and onset of sprout growth is accompanied by numerous biochemical changes, many of which are detrimental to the nutritional and processing qualities of potatoes. Endogenous hormones have been proposed to play a significant role in tuber dormancy regulation. The involvement of all major classes of endogenous hormones in tuber dormancy is reviewed. Based on available evidence, it is concluded that both ABA and ethylene are required for dormancy induction, but only ABA is needed to maintain bud dormancy. An increase in cytokinin sensitivity and content appear to be the principal factors leading to the loss of dormancy. Changes in endogenous IAA and GA content appear to be more closely related to the regulation of subsequent sprout growth.  相似文献

The magnetometer and electron reflectometer investigation (MAG/ER) on the Mars Global Surveyor spacecraft has obtained magnetic field and plasma observations throughout the near-Mars environment, from beyond the influence of Mars to just above the surface (at an altitude of approximately 100 kilometers). The solar wind interaction with Mars is in many ways similar to that at Venus and at an active comet, that is, primarily an ionospheric-atmospheric interaction. No significant planetary magnetic field of global scale has been detected to date (<2 x 10(21) Gauss-cubic centimeter), but here the discovery of multiple magnetic anomalies of small spatial scale in the crust of Mars is reported.  相似文献

Five groups of four, housed Cheviot ewes (mean live-weight 50 kg) were given single doses of 0, 2.5, 5, 10 or 20 g cupric oxide particles in gelatin capsules while receiving a diet of marginal copper content based on pelleted oats. After 65 days liver copper concentrations had increased curvilinearly in relation to dose and all ewes given 10 or 20 g cupric oxide particles showed increases of at least 13.4 mmol kg-1 dry matter (850 ppm). Liver copper concentrations had generally declined after 85 days but biochemical and histological evidence of copper toxicity was recorded in one ewe which had received 20 g cupric oxide particles. Despite marked variations between individual sheep a dose of 0.1 g kg-1 liveweight (5 g) was considered to be safe and did not induce clinical copper toxicity in five sheep of the susceptible North Ronaldsay breed given the same basal diet.  相似文献

The distribution and prevalence of Thelleria buffeli in Queensland cattle were investigated using serum samples and blood films collected primarily for brucellosis surveillance and tick fever diagnosis. Serums from 8654 cattle from 357 farms throughout Queensland were examined by an indirect fluorescent antibody test for antibody to T buffeli. In addition, 347 peripheral blood films collected from 147 farms in south-eastern Queensland were examined for piroplasms of T buffeli. The overall herd and animal prevalences for T buffeli were 75% and 41%, respectively. There was significant variation among regions in both herd and animal prevalences (P less than 0.001). Herd and animal prevalences were highest in the north and east decreasing westward. The results indicate that T buffeli is more widespread in Queensland than previously thought.  相似文献

DNA transposons are mobile genetic elements that have shaped the genomes of eukaryotes for millions of years, yet their origins remain obscure. We discovered a virophage that, on the basis of genetic homology, likely represents an evolutionary link between double-stranded DNA viruses and Maverick/Polinton eukaryotic DNA transposons. The Mavirus virophage parasitizes the giant Cafeteria roenbergensis virus and encodes 20 predicted proteins, including a retroviral integrase and a protein-primed DNA polymerase B. On the basis of our data, we conclude that Maverick/Polinton transposons may have originated from ancient relatives of Mavirus, and thereby influenced the evolution of eukaryotic genomes, although we cannot rule out alternative evolutionary scenarios.  相似文献

Potato tuber pink eye (PE) is a disorder of unknown origin that results in significant postharvest quality deterioration and rot. Little is known about the physiology of PE, including the characteristic tissue autofluorescence that defines the PE syndrome. The objective of this research was to identify the source of PE-induced autofluorescence and PE-related susceptibility to infection. The suberized barrier of the native periderm and cellular characteristics of neighboring parenchyma tissues were investigated to determine their involvement in the PE disorder. The results create a new physiological model describing the disorder and addressing the enigma of PE. Characteristics of the PE model emerge from the following results: (1) the integrity of the suberized barrier of the native periderm was compromised or absent in some surface areas of PE tubers thereby implicating the breakdown of the native periderm and its associated suberin barrier with PE and the susceptibility of PE tubers to pathogen infection; (2) the PE complex was characterized by unusual suberin poly(phenolic) (SPP) accumulations in the cortical parenchyma followed by latent suberin poly(aliphatic) (SPA) accumulations that were generally insufficient to form a complete barrier that was competent to block infections by pathogenic bacteria and fungi; (3) the aberrant absence or compromised integrity of the suberin barrier, including associated waxes, resulted in erratic increased susceptibility to water vapor loss known to cause tuber shrinkage and flaccidity; (4) widespread accumulations of SPP on parenchyma cell walls were the durable source of autofluorescence commonly used to determine the presence of the disorder; (5) the erratic development of unusual internal phellogen and periderm layers that, if complete with SPA, blocked hyphal advancement; (6) combined, the data provide a plausible explanation for PE infection court and rot anomalies as they occur without ingress of a wound opening. Results also demonstrated that neutral red may be used as a sensitive fluorochrome to detect intact hydrophobic areas in hyphae. Collectively, the results provide compelling evidence that the PE disorder includes a physiological basis.  相似文献

AIM: To minimise the impact of initial variation in liver copper (Cu) on assessments of Cu supplements for cattle in depletion/repletion experiments.

METHODS: Efficacy of two Cu injections was assessed with 18 calves, weighing 200–250 kg, given a Cu-deficient barley diet, containing 4.1 mg Cu/kg dry matter (DM) and added molybdenum (3 mg/kg) and sulphur (3 g/kg). Initial liver biopsy Cu ranged from 3.15–14.17 mmol/kg DM and nine calves with the highest values were given three subcutaneous injections of 235 mg tetrathiomolybdate (TTM) after 42–46 days depletion to lower liver Cu. Untreated (L) and TTM-treated (H) calves were ranked separately for liver Cu after 50 days depletion and allocated to one of three treatments: 100 mg Cu given subcutaneously as CuCaEDTA in either a paraffin (CuP) or aqueous base (CuA) after 56 days depletion (Day 0) or no injection (O). Thereafter, plasma and liver biopsy Cu were measured every 2–4 weeks for 16 weeks. Responses in liver Cu to Cu injections were compared with and without loge transformation and by linear regression.

RESULTS: Prior to Cu injection, the fractional decline in liver Cu concentration (FDLCu) after 50 days depletion was 0.64 (SE 0.066) and 0.80 (SE 0.090) in H and L calves, respectively (p=0.09) and mean liver Cu did not differ on Day ?6 (6.65 (SE 0.516) and 4.91 (SE 0.681) mmol/kg DM, respectively). Mean plasma Cu was higher in H than L calves on Day 0 (16.6 (SE 0.52) and 13.3 (SE 0.49) μmol/L, respectively (p<0.001)). Rates of decline in loge liver Cu between Days 0–84 in treatments L and H were: 0.0138 and 0.0071 for Groups O; 0.0033 and 0.0016 for Groups CuP; 0.0073 and 0.0049 for Groups CuA (pooled SE 0.0014) mmol/kg DM/day, respectively. Between Days 84–114, FDLCu was uniformly high across experiments and groups (0.59 (SE 0.042)). Cu injections did not affect plasma Cu, which remained 3.1 (SE 0.41) umol/L higher in Experiment H than in L (p=0.017).

CONCLUSIONS: The use of rates of change in liver copper concentrations improved the assessment of efficacy for two parental copper supplements and that of pre-treatment with tetrathiomolybdate, which, contrary to expectation, slowed Cu turnover by mechanisms that remain unclear.  相似文献
