Seven of 18 elk on a deer farm were found by the official Rose‐Bengal agglutination test (RBT) and tube agglutination test to be brucellosis reactors/suspects. Evaluation with the competitive ELISA (C‐ELISA) and the fluorescence polarization assay (FPA) tests revealed that six and five sera were positive respectively. The seven reactors/ suspects were slaughtered and their blood and tissues were collected. Brucella species could be isolated from three of the slaughtered animals, with nine isolates being obtained from the popliteal, supramammary and submandibular lymph nodes, vaginal discharge, mammary tissue and spleen. Brucella genus‐specific PCR based on 16S rRNA and AMOS‐PCR, which is specific for differential Brucella species, revealed that all nine isolates were Brucella abortus. These nine were further confirmed to be B. abortus biovar 1 by classical biotyping scheme assays. This is the first report of an outbreak of brucellosis in domestic elk in Korea. Our observations suggest that deer should be included in the routine Brucella surveillance programme for the effective control and prevention of brucellosis in Korea.  相似文献

Waxy corn starch was modified by dry heating (130 °C, 2 h) in the presence of octenyl succinic acid (OSA) at different pHs (4, 6, and 8), and the physicochemical properties of the modified starch were evaluated. The dry-heated octenyl succinylated (DH-OS) starch was also evaluated as a partial fat substitute in a muffin. Dry heating of starch under a more acidic condition resulted in lower degree of polymerization (DPn) but lower degree of substitution (DS). The pasting viscosity of the DH-OS starch prepared under basic conditions was higher than that of the unheated counterparts, whereas that under acidic conditions was lower. It was because the substitution favored basic conditions, and thermal degradation was accelerated by acids. The gelatinization characteristic was not significantly altered by OSA substitution and dry heating. The muffin containing the DH-OS starch had a higher specific volume and a softer texture than the muffin containing maltodextrin. Among the DH-OS starches tested, the starch prepared under acidic conditions produced muffins that had a greater volume, which was most likely due to its low molecular size. The muffin prepared with DH-OS starch at pH 6 was the softest in texture and had characteristics that were relatively close to the full-fat muffin.  相似文献

The objective of this study was to determine the frequency of different mastitis pathogens in 1255 milk samples collected from 368 lactating cows on 24 dairy farms in Korea during 2008. The proportion of cows and quarter milk samples having SCC ? 200,000 cells/ml, an indicator of udder infection, was 54.3% (200/368 cows) and 35.5% (446/1255 samples), respectively. Of the 446 milk samples subjected to bacteriological examination, 16.5% (74) showed no bacterial growth and 3.5% (16) were contaminated. In total, 356 of 1255 (28.3%) samples were bacteriologically positive, from which 415 bacteria were isolated. The most frequently isolated pathogen was coagulase-negative staphylococci (40.7%), followed by Gram-negative bacteria (19.5%) other than Escherichia coli, Staphylococcus aureus (12.2%), Streptococcus uberis (5.3%), Enterococcus spp. (4.8%), E. coli (4.5%), and environmental streptococci (3.1%) other than S. uberis. This study demonstrates that environmental pathogens were the vast majority of bacteria isolated from mastitic bovine milk samples in Korea.  相似文献

Many oomycete and fungal plant pathogens are obligate biotrophs, which extract nutrients only from living plant tissue and cannot grow apart from their hosts. Although these pathogens cause substantial crop losses, little is known about the molecular basis or evolution of obligate biotrophy. Here, we report the genome sequence of the oomycete Hyaloperonospora arabidopsidis (Hpa), an obligate biotroph and natural pathogen of Arabidopsis thaliana. In comparison with genomes of related, hemibiotrophic Phytophthora species, the Hpa genome exhibits dramatic reductions in genes encoding (i) RXLR effectors and other secreted pathogenicity proteins, (ii) enzymes for assimilation of inorganic nitrogen and sulfur, and (iii) proteins associated with zoospore formation and motility. These attributes comprise a genomic signature of evolution toward obligate biotrophy.  相似文献

The Afrotropical mosquito Anopheles gambiae sensu stricto, a major vector of malaria, is currently undergoing speciation into the M and S molecular forms. These forms have diverged in larval ecology and reproductive behavior through unknown genetic mechanisms, despite considerable levels of hybridization. Previous genome-wide scans using gene-based microarrays uncovered divergence between M and S that was largely confined to gene-poor pericentromeric regions, prompting a speciation-with-ongoing-gene-flow model that implicated only about 3% of the genome near centromeres in the speciation process. Here, based on the complete M and S genome sequences, we report widespread and heterogeneous genomic divergence inconsistent with appreciable levels of interform gene flow, suggesting a more advanced speciation process and greater challenges to identify genes critical to initiating that process.  相似文献

We investigated the effects of hydrogen peroxide on mucous cells and lysozymes in gill tissue of olive flounder. Morphological analysis revealed significant changes in the number of mucous cells after exposure to 300 and 500 mg L ? 1 hydrogen peroxide; however, there was no detectable difference in the 100 mg L ? 1 group. Lysozyme activities declined gradually in the gill mucus, but in contrast activities in serum increased significantly following the completion of treatment. The hepatic c‐ and g‐type lysozyme levels showed significant responses, indicating that the liver is a major lysozyme producer in response to stress condition; however, in the head kidney, there were no significant changes in the g‐type lysozyme level in all the groups, while the only detectable difference with the c‐type gene in the 300 mg L ? 1. While the treatments showed no significant changes compared with the control for either branchial lysozyme, there were significant differences in c‐type lysozyme between the 3rd treatment and 5 day. Taken together, our data indicate that hydrogen peroxide treatment may lead to modulation of the innate immune response of olive flounder; therefore, the application of hydrogen peroxide in a field system will require more attention to dose and timing effects.  相似文献

A nationwide investigation was performed to detect the presence of 1014 mutation(s) in voltage gated sodium channel (kdr) gene of Culex quinquefasciatus from 14 residential areas across 13 states and a federal territory in Malaysia. Molecular genotyping of kdr mutation was performed via a modified three tubes allele-specific-polymerase chain reaction (AS-PCR) and direct sequencing of kdr gene. Based on the results of AS-PCR, homozygous susceptible (SS) genotype was found in nine out of 14 populations with 38 individuals from a total sample size of 140. Heterozygous (RS) genotype was most predominant (99 individuals) and distributed across all study sites. Homozygous resistance (RR) genotype was detected in Perak (one individual) and Selangor (two individuals). The resistance kdr allele frequencies ranged from 0.1 to 0.55, with the highest being detected in Cx. quinquefasciatus population from Selangor. This study has documented the first field-evolved instance of 1014F mutation in Malaysian mosquitoes and the findings of this study could be utilized in the implementation of strategic measures in vector control programs in Malaysia.  相似文献

In this study, Brazilian coffee beans processed to different stages of roast at 210, 220, 230, and 240 °C were analyzed for pH value, titratable acidity, moisture content, and color lightness. Fourier transform infrared (FTIR) spectroscopy, in conjunction with principal component analysis, was conducted to study the effects of process time and temperature on the IR-active components of the acetyl acetate extract of the roasted coffee. The results showed that high-temperature-short-time resulted in higher moisture content, higher pH value, and higher titratable acidity when the beans were roasted beyond the start-of-second-crack stage, as compare to low-temperature-long-time process (LTLT). The LTLT process also resulted in greater IR absorbance for aldehydes, ketones, aliphatic acids, aromatic acids, and caffeine carbonyl bands on the FTIR spectra. Clusters for principal component score plots were well separated, indicating that the changes IR-active components in the coffee extracts, due to the different roasting treatments, can be discriminated by the FTIR technique. On the basis of the loading plots of principal components, changes of IR-active compounds in the coffee extract at various stages of roasting were discussed.  相似文献
