A diversity of N2-fixing (diazotrophic) bacteria was isolated from two traditional rice cultivars, Sataria and Kartiki, from the rice growing area of Mithila region of North Bihar, India, where low levels of nitrogen fertilizers are applied. Nitrogen-free semisolid media NFb, JMV and LGI with different carbon sources and pH-values were used for enrichment and isolation of root-associated diazotrophs. The colonization density of roots by diazotrophs, as estimated from positive pellicle formation at highest dilution in nitrogen-free enrichment media, was 106–108 diazotrophic bacteria per g fresh root weight. Roots of the cultivar Kartiki were found to be more densely colonized endophytically by diazotrophs as detected after chloramine T (1%) surface disinfection. To ascertain the phylogenetic affiliation of the isolates, phylogenetic oligonucleotide probes and the Fluorescent in situ Hybridization (FISH) technique were applied. Using group-specific rRNA directed oligonucleotide probes, the majority of the isolates could be identified as alpha-, beta-, or gamma-proteobacteria. Using 16S and 23S rRNA-directed genus- or species-specific probes, Herbaspirillum seropedicae, Azospirillum amazonense, Burkholderia cepacia/vietnamiensis, Rhizobia and Pseudomonas spp. were found to be the most prominent root associated culturable diazotrophs. Diazotrophic Gluconacetobacter spp. were also demonstrated as colonizers of rice roots. Burkholderia cenocepacia, Pseudomonas sp. and three diazotrophic PGPR reference strains were used for the inoculation of axenically grown rice seedlings to determine the plant growth promoting potential. Significant increases in the shoot length (up to 60%), shoot dry weight (up to 33%) and the grain yield (up to 26%) per plant were observed in non-axenic pot and field trials. Using semisolid enrichment media after surface sterilization of field grown inoculated rice roots and oligonucleotide probing of the diazotrophic enrichment cultures, a sustainable colonization with the inoculated bacteria could be demonstrated.  相似文献

Plants respond differentially to different arbuscular mycorrhizal fungi (AMF) as well as to the different soil moisture levels. Based on this background, the present study was carried out to investigate the effects of different levels of soil moisture and AMF inoculations on mycorrhization and growth of important agroforestry plants, viz., Phaseolus mungo, Triticum aestivum, Eucalyptus tereticornis, and Albizia procera. The experiments consisted of main treatment, i.e., three levels of soil moisture [field capacity (FC?=?16 %), half-field capacity (FC/2?=?8 %) and double-field capacity (2×FC?=?32 %)] and four subtreatments (mycorrhizal inoculations), viz., Acaulospora scrobiculata, Glomus cerebriforme, Glomus intraradices, and un-inoculated (control). AMF inoculations significantly (P?<?0.05) increased growth and P uptake, in all tested plant species. In P. mungo, maximum AMF efficiency was observed at FC while in other plants, AMF were equally effective at FC/2 and 2×FC. Different inoculants were effective at different moisture levels. Furthermore, mycorrhization was the highest at FC. AMF inoculations were more important than soil moisture (explaining 33–97 % variation in growth) in P. mungo, T. aestivum, and A. procera (forward selection method), whereas soil moisture was more important for growth of E. tereticornis. Thus, it may be stated that depending upon soil moisture, inoculation of plants with suitable AMF consortium can be beneficial.  相似文献

To compare the phenolic responses under oxidative stressors, plants of two Italian cultivars of durum wheat (Claudio and Mongibello) were (a) exposed to ozone (O3) (80 ppb, 5 hr/day for 70 consecutive days), with the aim to investigate the changes of phenolic compound contents in their leaves, or (b) flooded (seven consecutive days). Plants showed O3-induced visible injury, but their photosynthetic performance was not affected by the pollutant. Specifically, Claudio showed a higher O3 tolerance than Mongibello. The major value of the present study is undoubtedly the pioneering investigation of phenolic metabolism of durum wheat under O3. We identified 12 foliar phenolic compounds in all leaf samples (i.e. controls, exposed to O3 and flooded): ten phenolic acids, a flavanol (catechin hydrate) and a phenolic aldehyde (syringaldehyde). Overall, O3 exposure resulted in accumulations of phenolic compounds, especially in Claudio. These responses can be likely considered a fine-regulated repair process that equipped Claudio stressed plants with an antioxidant system capable of scavenging oxidative stress. Different phenolic variations were found in flooded plants, suggesting that phenolic response to environmental constraints is stress specific. Our study confirms that investigations and characterization of specific phenolic profiles of crop cultivars under oxidative stress may be helpful in breeding programmes.  相似文献

The kinetic profiles of norfloxacin were evaluated in afebrile, febrile and probenecid pre-treated (70 mg/kg orally) febrile goats after a single intravenous (i.v) dose (5 mg/kg). Fever was induced and maintained for 12 h by injecting Escherichia coli endotoxin (0.2 g/kg, i.v.) and repeating it in half the dose (0.1 g/kg) 5 h later. The plasma pharmacokinetic values for norfloxacin were best represented using a two-compartment open model. The peak norfloxacin plasma level of 90.52±3.18 g/ml attained in the probenecid pre-treated febrile goats was higher than that in the febrile (75.46±0.72 g/ml) or afebrile goats (62.25±1.23 g/ml). ClB and K el values were significantly (p<0.01) decreased in febrile compared with afebrile goats. These values were further reduced in febrile goats after probenecid pre-treatment. However, t 1/2 was not affected by the fever-probenecid interaction. Residue retention and reduced tillage are both conservation agricultural practices that may enhance soil organic carbon (SOC) stabilization in soil. We evaluated the long‐term effects of no‐till (NT) and stover retention from maize on SOC dynamics in a Rayne silt loam Typic Hapludults in Ohio. The six treatments consisted of retaining 0, 25, 50, 75, 100 and 200% of maize residues on each 3 × 3 m plot from the crop of previous year. Soil samples were obtained after 9 yrs of establishing the experiment. The whole soil (0–10 and 10–20 cm of soil depths) samples under different treatments were analysed for total C, total N, recalcitrant C (NaOCl treated sample) and 13C isotopic abundance (0–10 cm soil depth). Complete removal of stover for a period of 9 yrs significantly (P < 0.01) decreased soil C content (15.5 g/kg), whereas 200% of stover retention had the maximum soil C concentration (23.1 g/kg). Relative distribution of C for all the treatments in different fractions comprised of 55–58% as labile and 42–45% as recalcitrant. Retention of residue did not significantly affect total C and N concentration in 10–20 cm depth. 13C isotopic signature data indicated that C4‐C (maize‐derived C) was the dominant fraction of C in the top 0–10 cm of soil layer under NT with maize‐derived C accounting for as high as 80% of the total SOC concentration. Contribution of C4‐C or maize‐derived C was 71–84% in recalcitrant fraction in different residue retained plots. Residue management is imperative to increase SOC concentrations and long‐term agro‐ecosystem necessitates residue retention for stabilizing C in light‐textured soils.  相似文献

We studied variations in water relations and drought response in five Himalayan tree species (Schima wallichii (DC.) Korth. (chilaune) and Castanopsis indica (Roxb.) Miq. (dhale katus) at an elevation of 1400 m, Quercus lanata Smith (banjh) and Rhododendron arboreum Smith (lali gurans) at 2020 m, and Quercus semecarpifolia Smith (khasru) at 2130 m) at Phulchowki Hill, Kathmandu, Nepal. Soil water potential at 15 (Psi(s15)) and 30 cm (Psi(s30)) depths, tree water potential at predawn (Psi(pd)) and midday (Psi(md)), and leaf conductance during the morning (g(wAM)) and afternoon (g(wPM)) were observed from December 1998 to April 2001, except during the monsoon months. There was significant variation among sites, species and months in Psi(pd), Psi(md), g(wAM) and g(wPM), and among months for all species for Psi(s15). Mean Psi(pd) and Psi(md) were lowest in Q. semecarpifolia (-0.40 and -1.18 MPa, respectively) and highest in S. wallichii (-0.20 and -0.63 MPa, respectively). The minimum Psi value for all species (-0.70 to -1.79 MPa) was observed in March 1999, after 4 months of unusually low rainfall. Some patterns of Psi(pd) were related to phenology and leaf damage. During leafing, Psi(pd) often increased. Mean g(wAM) and g(wPM) were highest in Q. semecarpifolia (172 and 190 mmol m(-2) s(-1), respectively) and lowest in C. indica (78 and 74 mmol m(-2) s(-1), respectively). Soil water potential (Psi) at 15 cm depth correlated with plant Psi in all species, but rarely with g(wAM) and not with g(wPM). Plant Psi declined with increasing elevation, whereas g(w) increased. As Psi(pd) declined, so did maximal g(w), but overall, g(w) was correlated with Psi(pd) only for R. arboreum. Schima wallichii maintained high Psi, with low stomatal conductance, as did Castanopsis indica, except that C. indica had low Psi during dry months. Rhododendron arboreum maintained high Psi(pd) and g(w), despite low soil Psi. Quercus lanata had low g(w) and low Psi(pd) in some months, but showed no correlation between tree Psi and g(w). Quercus semecarpifolia, which grows at the highest elevation, had low soil and plant Psi and high g(w).  相似文献

This review aims to highlight the effects of ochratoxin A (OTA) in the feed of meat-producing animals. The accumulation of OTA in feed and its distribution in various farm animals were compared and evaluated. Primarily, the oral administration of OTA-contaminated feed and the predisposition in an animal''s vital organ were critically examined in this work. The collated reports show that OTA directly associated with endemic nephropathy and its high concentration leads to degeneration of liver cells, and necrosis of intestinal and lymphoid tissues. At present, limited reports are available in the recent literature on the problems and consequences of OTA in feed. Therefore, this review focused on the OTA carryover from feed to farm animals and the interaction of its secondary metabolites on their biochemical parameters. Hence, this report provides greater insights into animal health related to OTA residues in meat and meat products. This article also explores mitigation strategies that can be used to prevent the carryover effects of OTA in livestock feeds and the effects in the food chain.  相似文献
