Here we describe the diagnostic utility of the indirect immunofluorescence assay (IFA) during a recent outbreak of highly pathogenic avian influenza (HPAI) subtype H5N1 virus in southern Thailand and demonstrate the usefulness of the cardiac tissue from infected chickens, quail, and ducks for diagnosis. The most reliable sample for IFA diagnosis of influenza A virus was cardiac tissue (83.0%; 44/53) which when divided by species (chicken, quail and duck cardiac tissues) gave respective positivity rates of 88% (22/25), 88.9% (16/18) and 60.0% (6/10). Cardiac tissue also gave the highest IFA intensity for the three species. We believe that the IFA method has wide applicability in developing countries or remote settings where clinically similar avian diseases with high morbidity and mortality such as Newcastle disease and fowl cholera are common and could be rapidly excluded thereby conserving valuable reference laboratory capacity for true HPAI outbreaks. 相似文献

By determining total and natural clay by the pipette method, it is possible to obtain a more reproducible measure of aggregate stability in soils. This index has been called the “index of structure” and is reported on a scale from 0 to 100. High values represent water stable materials. Where total clay contents are below 25%, multiple determinations are needed to produce reliable results. The index of structure can be used to differentiate between parent materials since it does not vary appreciably in the drainage catenas examined so far. It gives a good measure of the relative water stability of aggregates, especially when the time on the mixer is altered during the natural clay determination. The index is significantly related to porosity but there are appreciable differences between major soil groups. Humid region soils appear to give a more consistent relationship than those of seasonally dry areas. 相似文献

Many protected area (PA) systems have developed in response to socio-economic and aesthetic criteria and need to be modified to increase their conservation value. National gap analyses are an important step in describing and addressing this problem, so we sought to determine the representativeness of English PAs devoted to biodiversity conservation by using Natural Areas (NAs), elevation and PA boundary data. We found that National Nature Reserves (NNRs) and Sites of Special Scientific Interests (SSSIs) cover only 6.3% of England and are generally small, with respective median areas of 1.1 and 0.2 km2. The English PA system under-represents lowland areas and provides a median level of 2.5% protection for the NA types, with seventy nine per cent of NA types having less than 10% protection. Therefore, we suggest that England's PA system needs to be expanded, although this would probably entail modification of existing legislation to increase involvement by landowners. We also compare our results with previous appraisals that used species distribution record data and suggest that landscape-level analyses may give a more accurate and less positive assessment. 相似文献

Mammalian herbivores regularly browse plantation seedlings. In many areas, seedlings require some form of protection if they
are to survive and grow into a productive plantation. Two general approaches for reducing browsing damage to seedlings are
to apply chemical repellents and tree guards. Both methods have existed for a long time, and new variations are constantly
being developed. Seedling stocking guards, a type of tree guard, are being used operationally in Tasmania, Australia, but
there is limited data quantifying their effectiveness and concerns with negative effects on tree performance. Conversely,
although proven effective, repellents are not being used, but are potentially cheaper and less problematic. We therefore aimed
to determine which is more effective under operational conditions, whether this effectiveness can be improved or extended,
and if either treatment has any effects on seedling form or survival. We planted Eucalyptus nitens seedlings with combinations of repellent and stocking guards in six operational plantations to examine and compare their
effectiveness. Seedlings were monitored for 12 months to examine treatment longevity. We found that both stocking guards and
repellent significantly reduced and delayed browsing severity, with their effects being additive. No negative effects on growth
were evident after 12 months, but adverse effects on seedling form warrant further investigation. Both of these methods can
be easily and relatively cheaply applied in the nursery before planting, making them appealing options to reduce browsing.
However, the ideal method for a given site will depend upon local browsing intensity. 相似文献

Browsing by mammalian herbivores is a major problem in plantation forestry. Seedlings are most vulnerable to browsing during establishment, making protection crucial during this period. Aside from reducing herbivore numbers, browsing can be controlled through the application of tree guards or chemical repellents. These methods are generally short-term options. A promising, potentially longer-term method of reducing browsing damage on plantations is to plant individuals with enhanced natural browse resistance. This requires the development of a rapid, cost-effective means of identifying germplasm with enhanced resistance. Here we present such a screening methodology.We planted Eucalyptus globulus seedlings from 22 different seedlots in randomised blocks along edge rows of six operational plantations. Seedlings were monitored for mammal browsing damage and growth for 2 years. Natural variation between E. globulus seedlots resulted in significant variation in the extent to which they were browsed. Differential browsing was evident just 10 weeks after planting, and caused a significant effect on tree growth after 2 years. Differential browsing was thought to be due to variation in levels of plant secondary metabolites (PSMs). The identification and selective planting of high PSM seedlots in high risk areas could be an effective management tool to reduce browsing in plantation forestry. 相似文献

The release of alkanolamines and glycols into the subsurface soils poses a potential hazard to the environment through impacted soil and groundwater. This study investigated aerobic and anaerobic biodegradability of monoethanolamine (MEA), ethylene glycol (MEG) and triethylene glycol (TEG). Significant levels of MEA (31 000 mg/kg), MEG (500 mg/kg) and TEG (2100 mg/kg) were successfully aerobically biodegraded in bioreactors. The aerobic slurry experiments suggested initial phosphate (P) limitation, as biodegradation rates increased by one order of magnitude after phosphate addition. Anaerobic decay of MEA, MEG and TEG was unaffected by P-addition. MEA, MEG and TEG degradation products such as acetate, ethanol and ammonium at about 75 000 mg/kg, 8100 mg/kg and 8800 mg/kg degraded completely and did not prevent aerobic biodegradation. This study confirms proposed biodegradation pathways of MEA, MEG, TEG and their breakdown products in natural soil and groundwater using indigenous microbes. The commercial production of herbicide tolerant (HT) canola began in Western Canada in 1997. With more than a decade of use, the actual farm-level environmental impact of HT canola can be evaluated. This article reports on a spring 2007 survey of nearly 600 canola farmers in the three prairie provinces of Western Canada. Producers were asked about their crop production experiences for 2005 and 2006 and expected crop planting for 2007. A reduction in the total number of chemical applications over the 3-year period was reported, resulting in a decrease of herbicide active ingredient being applied to farmland in Western Canada of nearly 1.3 million kg annually. Fewer tillage passes over the survey period were reported, improving moisture conservation, decreasing soil erosion and contributing to carbon sequestration in annual cropland. An estimated 1 million tonnes of carbon is either sequestered or no longer released under land management facilitated by HT canola production, as compared to 1995. The value of this carbon off-set is estimated to be C$5 million. Comparisons with similar studies and against non-adoption of HT canola can guide future decisions about HT canola adoption. 相似文献
