Vitamin D and sterol composition of 10 types of mushrooms from retail suppliers in the United States

Vitamin D(2) (ergocalciferol) and sterols were analyzed in mushrooms sampled nationwide in the United States to update the USDA Nutrient Database for Standard Reference. Vitamin D(2) was assayed using HPLC with [(3)H]-vitamin D(3) internal standard and sterols by GC-FID mass spectrometric (MS) confirmation. Vitamin D(2) was low (0.1-0.3 μg/100 g) in Agaricus bisporus (white button, crimini, portabella) and enoki, moderate in shiitake and oyster (0.4-0.7 μg/100 g), and high in morel, chanterelle, maitake (5.2-28.1 μg/100 g) and UV-treated portabella (3.4-20.9 μg/100 g), with significant variability among composites for some types. Ergosterol (mg/100 g) was highest in maitake and shiitake (79.2, 84.9) and lowest in morel and enoki (26.3, 35.5); the range was <10 mg/100 g among white button composites but 12-50 mg/100 g among samples of other types. All mushrooms contained ergosta-5,7-dienol (22,23-dihydroergosterol) (3.53-18.0 mg/100 g) and (except morel) ergosta-7-enol. Only morel contained brassicasterol (28.6 mg/100 g) and campesterol (1.23-4.54 mg/100 g) and no ergosta-7,22-dienol. MS was critical in distinguishing campesterol from ergosta-7,22-dienol. 相似文献

This investigation was designed to characterize phenolic metabolism of the olive cultivar, Hardy's Mammoth, by examining its constitutive tissues. The phenolic profiles of pulp, seed, stone, and new and old season leaves were monitored over two fruiting seasons, to investigate possible relationships between tissues and phenol content and to determine the impact of alternate fruit bearing. No major qualitative differences in phenolic composition were found between the various tissues; however, distinct differences between the tissues with respect to quantifiable phenols were established. Relationships between 2-(3,4-dihydroxyphenyl)ethyl (3E,4E)-4-formyl-3-(2-oxoethyl)hex-4-enoate ester, oleuropein, and hydroxytyrosol in pulp and leaf were identified and found to be related to alternate bearing. Concentrations of 5-caffeoylquinic acid in old season leaves differed dramatically between seasons, confirming earlier studies. 相似文献

An attempt was made to evaluate gluten structural changes in refined and whole wheat pasta from hard white winter wheat to elucidate the impact of whole wheat components on the formation and structure of the gluten network in pasta. Attenuated total reflectance–FTIR spectroscopy was used to track gluten secondary structure through most of the major steps in pasta processing: raw material, mixing, drying, and cooking. Protein solubility, accessible thiols, and SDS‐PAGE data were also collected to provide additional information on the nature of protein interactions and network composition. Few secondary structural differences were observed between refined and whole wheat flours from hard white wheat. However, mixing induced a significant shift to β‐sheet structures in refined dough that was not equally matched by whole wheat dough. Drying under both high temperature, short time (HT) and low temperature, long time (LT) conditions resulted in a reversion to structural distributions similar to those for flour in both pastas. However, greater protein denaturation in HT samples was indicated by lower protein solubility also in the presence of denaturants and disulfide reducing agents. Cooking generated a substantial increase in β‐sheet structures for both pasta systems. This structure was greatest in refined and LT samples. Thiol accessibility data indicate the presence of a highly aggregated, compact gluten network in refined pasta, mostly driven by hydrophobic association. Conversely, the network in whole wheat pasta was more loosely associated and dependent on disulfide bonding, both of which fit well with the secondary structural data. 相似文献

Shoot:root (S:R) response in four winter C3-grasses (cereals) viz. wheat (Triticum aestivum L.), rye (Secale cereale L.), barley (Hordeum vulgare L.) and oats (Avena sativa L.) was investigated under eight different NPK sources (S1 = 20-20-20, S2 = 20-27-5, S3 = 7-22-8, S4 = 10-10-10-20S, S5 = 11-15-11, S6 = 31-11-11, S7 = 24-8-16, and S8 = 19-6-12) in pot experiment at Dryland Agriculture Institute, West Texas A&M University, Canyon, Texas, USA during winter 2009–2010. The experiment was performed in completely randomized design (CRD) with three repeats. The objective of this experiment was to investigate whether the S:R of winter cereals differ or not when applied with different NPK sources. Considerable variations in the shoot: root was noticed in the four grasses at different growth stages, i.e., 30, 60 and 90 days after emergence (DAE) when applied with different NPK fertilizers sources. Wheat had the highest S:R of 3.4 at 30 days after emergence (DAE), barley at 60 DAE (6.5), while oats at 90 DAE (3.9). Among the NPK sources, crops had the highest S:R of 3.6, 6.2 and 3.7 when applied with S3, S2, and S6 at 30, 60 and 90 DAE, respectively. Shoot to root ratio showed positive relationship with increase in shoot weight and negative relationship with increase in root weight. The NPK fertilizer S6 (31: 11: 11), an acid loving fertilizer had negative effects on the shoot and root development of cool season cereals, but among these cereals under study, barley and oats roots were affected more than wheat and rye. It was concluded from this present study, that the four cool season cereals responded differently in terms of shoot: root to different NPK fertilizers at different growth stages. 相似文献

Under semi-arid Mediterranean conditions, limited moisture is the main constraint to rainfed cropping with wheat (Triticum aestivum), barley (Hordeum vulgare), and food and forage legumes. With increasing land-use pressure, moisture-conserving fallowing is being replaced by continuous cropping, which is considered an unsustainable practice. Thus, a long-term trial with durum wheat (T. turgidum var. durum) was established in 1983 at Tel Hadya, Aleppo, Syria (mean annual rainfall 330 mm) to assess alternative rotation options to fallow and continuous cropping. Nitrogen (N) and grazing/residue management were secondary factors. Soil aggregation, infiltration, hydraulic conductivity, and total soil organic matter and component fractions (fulvic and humic acids and polysaccharides) were determined at the end of 12 years. Some rotations, e.g., medic (Medicago sativa) and vetch (Vicia faba), significantly increased soil organic matter (12.5–13.8 g kg−1 versus 10.9–11 g kg−1 for continuous wheat and wheat/fallow). All measurements, or indices, indicated parallel trends with increasing organic matter, e.g., coefficients of macro-structure, micro-aggregation, and water-stable aggregates, and decreasing dispersion. Similarly, legume rotations had higher infiltration rates (16.2–21.8 cm h−1 versus 13.9–14.4 cm h−1 with continuous wheat and wheat/fallow) and hydraulic conductivity rates (8.7–12.4 cm h−1 versus 6.2–7.4 cm h−1 with continuous wheat and wheat/fallow). We conclude that cereal/legume rotations, in addition to being biologically and economically attractive, also enhance soil quality and thus promote soil use sustainability in fragile semi-arid areas as in the Mediterranean zone. 相似文献

Soil structure heterogeneity in the form of macropores and preferential flow channels can complicate efforts to quantify the physical and biological characteristics of wetland systems. We collected soil cores from two riparian wetlands to determine whether soil associated with macropores had elevated denitrification potentials compared to bulk soil from the same core. Cores were inspected for obvious macropores, which were distinguished as visible holes in the core, sometimes with decaying root matter, or as highly unconsolidated layers that appeared to have a substantially lower bulk density than the surrounding soil. Denitrification potentials were significantly higher in pores (P<0.05) for six of the 16 cores that were obtained from the Cheraw State Park site. In cores obtained from a second site, denitrification potentials were significantly higher in pores for six of 20 cores and the trend of higher denitrification in pores was present in the majority of cores that had measurable activity. In cores with significant differences, denitrification was often 1-2 orders of magnitude greater in soil surrounding the macropore than in the bulk soil. Denitrification potentials of the bulk soils were similar in magnitude to the potentials measured in composited cores from previous studies. It is possible that the difference between macropore and bulk denitrification rates developed due to preferential flow of nitrate-rich water through the macropores. This paper describes the identification and utilisation of a sequence-characterised amplified region (SCAR) marker specific for the Trichoderma virens biocontrol isolate GV4. The marker was developed from a RAPD-PCR amplification product unique to isolate GV4. When used as a hybridisation probe in Southern blot analysis, it hybridised to the DNA of the species T. virens alone and not to that of other Trichoderma species or closely related genera Gliocladium and Verticillium. The marker also produced a GV4-specific RFLP, distinguishing it from other T. virens isolates when probed to blots with HindII, BamHI or PstI genomic DNA digests. Primers designed from the sequence of the RAPD marker produced a diagnostic amplification product of 346 bp for GV4 alone, distinguishing it from all other test isolates. With the exception of one, test isolates did not produce an amplification product with the SCAR primers. The exception was a single Verticillium psalliotae isolate (ICMP5509) that produced a product of approx. 400 bp that was easily distinguished from the 346 bp product of GV4. The reliability of the SCAR-based diagnostic test was further improved with the introduction of a positive PCR reaction control to each test, achieved by converting the test to a duplex PCR system. Two universal primers flanking the two ITS and the 5.8S region of the ribosomal gene complex were introduced to each reaction to provide a test for PCR reaction inhibitors to eliminate false negatives in the diagnosis. Amplification of this multi-copy genomic region did not reduce diagnostic sensitivity of the single copy SCAR marker. To further increase the sensitivity of detecting GV4 propagules while maintaining a fast sample assessment assay, soil was amended with cornmeal, as a nutrient source, and a mix of antibiotics to favour Trichoderma growth. The soil mix was subsequently incubated for 5 d before total DNA was extracted. Under these conditions, the duplex soil PCR assay detected GV4 down to a concentration of 10 spores g−1 soil in non-sterile agricultural field soil. A robust two-stage approach is used to reanalyze the repeated measurements from an experiment of airway responsiveness in
rats randomized to long-term exposure at four ozone doses. The concentration-response data generated for each rat may be represented
as a hierarchical nonlinear model encompasing the sources of variation within and between individual profile for each rat,
the conditional modeling approach can assess the adequacy of an assumed mean model, a fundamental advantage not intrinsic
to marginal techniques. The two-stage population inference is based on the estimated individual parameters, thus maintaining
an intuitive appeal to the toxicologists who traditionally have fitted a separate curve for each animal and then applied ANOVA
to the summary statistics. However, we formally adjust the standard errors for the extra variability due to the initial estimation
of the individual parameters and also allow for their within-rat correlation. The robust two-stage method appropriately down
weights the a berrant responses arising sporadically within individualsand, more importantly, the rats which may be outlying
relative to the usual population variation. The true effect of chronic ozone exposure, including a significant gender interaction,

Myoglobin (Mb) and its iron have been proposed to be major prooxidants in cooked meats. To understand the mechanisms and differentiate between the prooxidant and antioxidant potential of oxymyoglobin (OxyMb) and metmyoglobin (MetMb), their prooxidant activity, iron content, solubility, free radical scavenging activity, and iron binding capacity were determined as a function of thermal processing. The ability of native and heat denatured OxyMb and MetMb to promote the oxidation of muscle microsomes was different. MetMb promoted lipid oxidation in both its native and denatured states. Conversely, OxyMb became antioxidative when the protein was heated to temperatures >or=75 degrees C. The increased antioxidant activity of heat denatured OxyMb was likely due to a decrease in its prooxidative activity due to its loss of solubility. These data show that the impact on oxidative reactions of Mb is the result of the balance between its antioxidant and prooxidant activities. 相似文献
