Informal economy of gold mining has contaminated some important river basins in Amazon. Follow-up studies on critical compartments showed some areas with high Hg levels in fish as well as in human hair samples. Average Hg in piscivorous fish in the Madeira river itself was 846 ppb (N=284) with a maximum of 3921 ppb. Mercury in fish from non polluted areas in this basin shows high variability, even for single species. A seasonal variation in Hg content was observed, with higher values at the end of the dry season. In the upper Tapajós basin comparable values were found for fish but with a definite decreasing trend downstream. Average value for piscivorous fish in the whole Tapajós basin is 482 ppb (N=122) with a maximum value of 3770 ppb. Hair Hg was higher in fishing villages in the Tapajós (average: 17 ppm; with N=432 and maximum value of 176 ppm) than in the Madeira (average: 9 ppm; N=169; maximum 71 ppm), and data from some areas of the Tapajós suggest a decrease with time. Mercury was much higher in urine of goldshop workers in Santarém (low Tapajós) than in Alta Floresta (high Tapajbs) and show a decreasing trend in both cases, probably related to the significant decline in gold mining activities during the study period (1986–1994).  相似文献

Formulas are presented for the calculation of extraction volume for pesticide residue procedures that use a single extraction with acetone, acetonitrile, or methanol, with or without prior dilution of these solvents with water. These formulas account for the volume change on mixing and for the volume contribution from the soluble sugars sucrose, glucose, and fructose. Results using these formulas are in agreement with the observed volumes of such mixtures within 0.3% throughout the following ranges, expressed as percent water in the mixtures: 15-38% for acetone, 4.8-59% for acetonitrile, and 9.3-33% for methanol.  相似文献

Cells of Dunaliella spp., unicellular marine green algae, differ from most other plant cells in lacking a cell wall. The responses of D. tertiolecta Butcher (DUN clone) (WHO I) when grown in a synthetic sea-water medium with Pb added were similar to those of other algae previously tested, thus disproving the idea that the presence of a cell wall might ‘protect’ cells. The Marine Algal Assay Procedure published by the National Environmental Research Center at Corvallis, Oregon, was used as an assay for the effects of Pb on this organism, with the intent of supporting attempts to standardize studies of this sort.  相似文献

A technique was developed to estimate the amount of sulfur held by the microbial population in soil. This method involves lysing the microbial cells with chloroform and measuring the S released to 10 mm CaCl2 or 0.1 m NaHCO3 extracts. Calculations of biomass-S were made from the experimentally determined proportion of cell S released. To determine this proportion, two species each of bacteria and fungi, which were grown at three concentrations of S, were added to the soil and treated with CHCl3. The average values of 34.6% with CaCl2 and 41.2% with NaHCO3 were calculated for the proportion of microbial S extracted following CHCl3 treatment. Biomass-S was found to represent approximately 2.3% of the total S in the soil.  相似文献

This non-linear model is expressed as a differential equation which cannot be integrated analytically. This paper describes the identification and utilisation of a sequence-characterised amplified region (SCAR) marker specific for the Trichoderma virens biocontrol isolate GV4. The marker was developed from a RAPD-PCR amplification product unique to isolate GV4. When used as a hybridisation probe in Southern blot analysis, it hybridised to the DNA of the species T. virens alone and not to that of other Trichoderma species or closely related genera Gliocladium and Verticillium. The marker also produced a GV4-specific RFLP, distinguishing it from other T. virens isolates when probed to blots with HindII, BamHI or PstI genomic DNA digests. Primers designed from the sequence of the RAPD marker produced a diagnostic amplification product of 346 bp for GV4 alone, distinguishing it from all other test isolates. With the exception of one, test isolates did not produce an amplification product with the SCAR primers. The exception was a single Verticillium psalliotae isolate (ICMP5509) that produced a product of approx. 400 bp that was easily distinguished from the 346 bp product of GV4. The reliability of the SCAR-based diagnostic test was further improved with the introduction of a positive PCR reaction control to each test, achieved by converting the test to a duplex PCR system. Two universal primers flanking the two ITS and the 5.8S region of the ribosomal gene complex were introduced to each reaction to provide a test for PCR reaction inhibitors to eliminate false negatives in the diagnosis. Amplification of this multi-copy genomic region did not reduce diagnostic sensitivity of the single copy SCAR marker. To further increase the sensitivity of detecting GV4 propagules while maintaining a fast sample assessment assay, soil was amended with cornmeal, as a nutrient source, and a mix of antibiotics to favour Trichoderma growth. The soil mix was subsequently incubated for 5 d before total DNA was extracted. Under these conditions, the duplex soil PCR assay detected GV4 down to a concentration of 10 spores g−1 soil in non-sterile agricultural field soil. This study is the first to report the use of a duplex-PCR diagnostic bioassay for a species within the Hypocrea/Trichoderma genus.  相似文献

Identifying corn ( Zea mays L.) genotypes with faster rates of seedling emergence and leaf appearance is important in developing a corn crop with earlier canopy closure and better seasonal light interception. A greenhouse experiment was conducted twice to investigate whether corn genotypes known to vary in canopy architecture and yield potential differed in rates of seedling emergence and leaf appearance. The experiment was arranged in a randomized complete block design utilizing seven genotypes: five of the newly developed Leafy reduced-stature (LRS) types (LRS1, LRS2, LRS3, LRS4 and LRS5), one conventional type [Pioneer 3979 (P3979)], and one late-maturing big-leaf (LMBL) type. Five seeds were planted in each pot and seedling emergence was recorded every day until all seeds emerged. Leaf appearance was recorded from seedling emergence until the plants reached anthesis. There was variability among the genotypes for both seedling emergence and leaf appearance rate. Mean seedling emergence values of greater than 90 % were achieved by four of the five LRS genotypes, and the LRS types generally emerged more rapidly than the others. Leaf appearance rate showed linear increases over time; however, LRS genotypes (in particular LRS3, LRS4 and LRS5) had more rapid leaf appearance rates than the others. The LMBL hybrid ranked last for both seedling emergence (<80 %) and leaf appearance rate. Rapid seedling emergence and leaf appearance by early-maturing genotypes (LRS and P3979, especially LRS) may help these types of genotypes achieve earlier canopy closure and better use of the light available during the growing season, which is critical in a short growing season environment.  相似文献

Abstract –  We compared timing of smolt migration for two populations of naturally spawned Atlantic salmon, Salmo salar , originating from an upper catchment (River Tummel) and a lower catchment (River Almond) tributary of the River Tay, Scotland. Smolts from the upper tributary began migration earlier than those from the lower tributary. On both occasions when fry derived from River Isla (lower tributary) stock were transferred to a location in the upper catchment, smolt migration was later than for native fish. Similarly, when fry from Tummel and Isla stocks were stocked in a common, upper catchment location, Isla origin fish migrated at a later date, in both of the two following smolt years. These differences are indicative of a genetic basis for the timing of smolt migration and suggestive of local adaptation. Mounting evidence points towards local genetic adaptation for the timing of expression of behaviours associated with migration. These aspects of variation should be accommodated in management theory and practices.  相似文献

We compared the frequency and severity of osteochondrosis lesions in young Thoroughbred horses with cervical stenotic myelopathy (CSM) vs that in clinically normal Thoroughbreds of the same age. All lesions of the cervical vertebrae and appendicular skeleton were classified histologically as osteochondrosis or nonosteochondrosis and were measured for severity. Minimal sagittal diameter was significantly smaller in horses with CSM from C2 through C6; no difference was detected at C7. Severity of cervical vertebral osteochondrosis was greater in the horses with CSM, however frequency was not different. Frequency and severity of nonosteochondrosis lesions were not different in cervical vertebrae or appendicular skeleton. Frequency and severity of appendicular skeleton osteochondrosis lesions were both greater in horses with CSM. Osteochondrosis and nonosteochondrosis lesions were more severe on facets at sites of compression than on facets at noncompressed sites in horses with CSM. However, compression was also observed at sites with no articular facet lesions. The association of widespread osteochondrosis and spinal canal narrowing with CSM suggests CSM may represent a systemic failure in the development or maturation of cartilage and bone.  相似文献
