High nitrogen fixing, phosphate solubilizing, phytohormones producing isolates of Azotobacter, Azospirillum, Acetobacter and Pseudomonas were used as inoculants for cotton. Important cultures were selected on the basis of their effect on root/shoot length and chemotactic behaviour. Selected bioinoculants were earlier tested for their beneficial properties like nitrogen fixation (ARA), ammonia excretion, IAA production etc. These bio-inoculants were further tested for phosphate solubilization property. Various chosen strains were tested with Desi (HD 123) and American (H 1098) cotton under field conditions (as for wheat). Plant height and boll weight were determined at the time of harvesting whereas survival rate of inoculated bacteria was determined after 30, 80 and 130 days respectively. In the year 2000–01, on the basis of boll number and boll weight plant?1 AVK 51 (36; 76.2?g plant?1), HT 57 (27; 56.9?g plant?1), AC18 (33; 61.5?g plant?1), Ala 27 (36; 61.4?g plant?1) and Pseudomonas (34; 71.3?g plant?1) were identified as significant both for American and desi cotton varieties. Highest survival rate was observed with Mac 68 (33.4 × 105) followed by HT54 (31.5 × 105) after 30 days of sowing, which decreased after 80 days and remained constant up to 130 days. Harmful algal blooms (HABs) of the toxic dinoflagellate, Karenia brevis, produce red tide toxins, or brevetoxins. Significant health effects associated with red tide toxin exposure have been reported in sea life and in humans, with brevetoxins documented within immune cells from many species. The objective of this research was to investigate potential immunotoxic effects of brevetoxins using a leukemic T cell line (Jurkat) as an in vitro model system. Viability, cell proliferation, and apoptosis assays were conducted using brevetoxin congeners PbTx-2, PbTx-3, and PbTx-6. The effects of in vitro brevetoxin exposure on cell viability and cellular metabolism or proliferation were determined using trypan blue and MTT (1-(4,5-dimethylthiazol-2-yl)-3,5-diphenylformazan), respectively. Using MTT, cellular metabolic activity was decreased in Jurkat cells exposed to 5 – 10 μg/ml PbTx-2 or PbTx-6. After 3 h, no significant effects on cell viability were observed with any toxin congener in concentrations up to 10 μg/ml. Viability decreased dramatically after 24 h in cells treated with PbTx-2 or -6. Apoptosis, as measured by caspase-3 activity, was significantly increased in cells exposed to PbTx-2 or PbTx-6. In summary, brevetoxin congeners varied in effects on Jurkat cells, with PbTx-2 and PbTx-6 eliciting greater cellular effects compared to PbTx-3. 相似文献

A previously described method for the non-aqueous fractionation of cereal flours by sedimentation in non-aqueous solvents was carried out using flours of three wheat cultivars differing in baking performance, as well as one rye and one barley flour. The method was based on differences in the densities of starch (higher) and protein (lower). Thus, suspending finely milled flour in an inert solvent mixture with a density in between the densities of starch and protein yielded a sedimented starch fraction and a protein-rich fraction at the surface of the solvent. Further purification of this upper fraction provided a protein fraction, a middle fraction, and a lipid fraction. The protein fractions were examined by means of rheological methods such as micro-extension tests and creep-recovery tests. They also were reconstituted to standard flour with a protein content of 13.5%, which was used for micro-scale baking tests. Compared to aqueous isolated gluten, the hydrated protein fractions from wheat were much more extensible and had a lower resistance to extension. The baking performance of the wheat protein fractions was superior to gluten and comparable to the native wheat flours. The protein fraction from rye gave a wheat-like bread crumb, whereas the barley protein was not suited for bread making. 相似文献

Studies within and outside the U.S. indicate recurring oak (Quercus spp.) regeneration problems. In deciduous forests of the eastern U.S., a prevailing explanation for this trend is fire suppression leading to high competitor abundance and low understory light. In response, prescribed fire is increasingly used as a management tool to remedy these conditions and encourage future oak establishment and growth. Within eastern Kentucky, we implemented single and repeated (3×) prescribed fires over a 6-yr period (2002–2007). Pre- and post-burn, we quantified canopy cover and oak seedling survival and growth compared to other woody seedlings deemed potential competitors, primarily red maple (Acer rubrum L.) and sassafras (Sassafras albidum (Nutt.) Nees.). Burning temporarily decreased canopy cover 3–10%, but cover rebounded the subsequent growing season. Repeated burning ultimately produced canopy cover about 6% lower than sites unburned and burned once, suggesting a cumulative effect on understory light. Red maple exhibited low survival (∼40%) following single and repeated burns, but growth remained similar to unburned seedlings. Burning had little impact on sassafras survival and led to total height and basal diameters 2× greater than unburned seedlings. A single burn had no impact on red oak (Erythrobalanus spp.) survival and increased height and basal diameters 25–30%, but this positive growth response was driven by seedlings on several plots which experienced high burn temperatures and consequently high overstory mortality. White oaks (Leucobalanus spp.), however, exhibited twice as high mortality compared to those unburned, with no change in growth parameters. Repeated burning negatively impacted survival and growth of both oak groups compared to unburned seedlings. With both burn regimes, oaks with smaller pre-burn basal diameters exhibited the lowest post-burn survival. Thus, despite the ability of prescribed burns to temporarily increase understory light and reduce red maple survival, neither single or repeated burns placed oaks in an improved competitive position. These findings result from a combination of highly variable yet interdependent factors including the (1) life history traits of oaks compared to their co-occurring competitors, (2) pre-burn stature of pre-existing oak seedlings, and (3) variability in fire temperature and effects on understory light. 相似文献

M-mode and Doppler echocardiographic values were obtained from 30 normal adult ferrets (14 neutered females, 13 neutered males, 3 intact males) sedated with an intramuscular combination of ketamine hydrochloride and midazolam. Routine M-mode measurements of the left and right ventricle, left atrium (LA) and aorta (AO) and Doppler measurements of aortic and pulmonic outflow, and mitral inflow were recorded. The following values were calculated: LA:AO diameter, ratio of peak E: peak A wave velocity (E:A ratio) for mitral inflow, and stroke volume (SV), cardiac output (CO) and cardiac index (CI) for both pulmonary and aortic outflow tracts. Maximal aortic velocities (AOmax) and velocity-time integral values (AO VTI) were significantly less than corresponding pulmonary outflow tract values (PAmax, PA VTI) but there was no difference in calculated values for SV, CO or CI. Calculated CO values were in the range expected based on the size of the species. Difficulties in aligning the aortic outflow tract for Doppler imaging may make pulmonary outflow Doppler values more consistent for use in estimating volume flow in ferrets. 相似文献

Various magnetic resonance (MR) imaging techniques have been used to assess lumbar spinal abnormalities in people. Four, young adult, clinically normal dogs were used to compare images of the spinal cord acquired using conventional spin-echo and rapid acquisition relaxation-enhanced (RARE), commonly called fast spin-echo (FSE), magnetic resonance imaging techniques. Lateral myelograms were made as an anatomic control. The T2-weighted FSE technique was characterized by better image quality than the T2-weighted conventional spin-echo technique. The short acquisition time with the FSE technique allowed increases in the matrix size and number of excitations, thus improving resolution and signal-to-noise ratio. In canine lumbar spinal MR imaging, use of a FSE technique is recommended to reduce the overall time for imaging and to improve image quality. 相似文献
