Microglial cells represent the endogenous immune system of the central nervous system (CNS). Upon pathological insults they reveal their immunological potential aimed at regaining homeostasis. These reactions have long been believed to follow a uniform and unspecific pattern which is irrespective to the underlying disease entity. Evidence is growing that this view seriously underrates microglial competence as the defenders of the CNS. In the present study, microglial cells of 47 dogs were examined ex vivo by means of flow cytometry. Ex vivo examination included immunophenotypic characterization using eight different surface markers and functional studies such as phagocytosis assay and the reactive oxygen species (ROS) generation test. The dogs were classified according to their histopathological diagnoses in disease categories (controls, canine distemper virus (CDV) induced demyelination, other diseases of the CNS) and results of microglial reaction profiles were compared. Immunophenotypic characterization generally revealed relative high conformity in the microglial disease response among the different groups, however the functional response was shown to be more specific. Dogs with intracranial inflammation and dogs with demyelination showed an enhanced phagocytosis, whereas a significant up-regulation of ROS generation was found in dogs with demyelination due to CDV infection. This strongly suggests a specific response of microglia to infection with CDV in the settings of our study and underlines the pivotal role of microglial ROS generation in the pathogenesis of demyelinating diseases, such as canine distemper.  相似文献

The aim of the study was to elucidate the distribution and possible transmission routes of Toxocara spp. infection in Estonia. Out of 454 faecal and sand samples collected from park lawns and sandpits in the town of Tartu, 19 were Toxocara positive (4.2%). Out of the 45 sandpit samples 17.8% were Toxocara positive. Cat faeces was found in 21 sandpit samples. Parasitological necropsies were performed on 41 euthanised stray dogs and 27 cats in the Tallinn Dog Home. Additionally, 13 wild free-roaming brown rats (Rattus norvegicus) were captured from the Tallinn Dog Home territory, necropsied and studied for the presence of Toxocara larvae. Toxocara canis adults were found in 14.6% of the dogs and Toxocara cati (syn. mystax) adults in the small intestines of 48.2% of the cats examined. Larval infection was detected in the kidney and liver in 5 dogs (12.2%). Our study demonstrated only low-level larval Toxocara infections in adult dogs. Toxocara larvae were not found in cats and brown rats. According to the results of this study, cats more often carry Toxocara infection than dogs. Under our conditions, stray and free-roaming cats are the main contaminators of the environment with Toxocara eggs. Children playing in sandpits are the main risk group for larval toxocarosis.  相似文献

An observational study was conducted in order to assess the impact of a contract breeding program on the reproductive performance in a selected group of Ohio dairies using event-time analysis. The contract breeding program was offered by a breeding co-operative and featured tail chalking and daily evaluation of cows for insemination by co-operative technicians. Dairy employees no longer handled estrus detection activities. Between early 2002 and mid-2004, test-day records related to production and reproduction were obtained for 16,453 lactations representing 11,398 cows in a non-random sample of 31 dairies identified as well-managed client herds of the breeding co-operative. Of the 31 herds, 15 were using the contract breeding at the start of the data acquisition period, having started in the previous 2 years. The remaining 16 herds managed their own breeding program and used the co-operative for semen purchase.

Cox proportional hazards modeling techniques were used to estimate the association of the contract breeding, as well as the effect of other significant predictors, with the hazard of pregnancy. Two separate Cox models were developed and compared: one that only considered fixed covariates and a second that included both fixed and time-varying covariates. Estimates of effects were expressed as the hazard ratio (HR) for pregnancy.

Results of the fixed covariates model indicated that, controlling for breed, herd size, use of ovulation synchronization protocols in the herd, whether somatic cell score exceeded 4.5 prior to pregnancy or censoring, parity, calving season, and maximum test-day milk prior to pregnancy or censoring, the contract breeding program was associated with an increased hazard of pregnancy (HR = 1.315; 95% CI 1.261–1.371). The results of the time-varying covariates model, which controlled for breed, herd size, use of ovulation synchronization protocols, somatic cell score above 4.5, parity, calving season, and testing season also found that the program was associated with an increased hazard of pregnancy (HR = 1.387; 95% CI 1.327–1.451).

The fixed and time-varying covariates models both found similar sets of predictors when analyzing the association of the contract breeding program with hazard of pregnancy. Both models identified a 30% or greater increase in hazard of pregnancy associated with use of the contract breeding program, suggesting that herds subscribing to the program achieved pregnancies in a more timely fashion.  相似文献

Discoid lupus erythematosus (DLE) is a well-known autoimmune disorder described in dogs and humans. In dogs, DLE is considered the second most common immune-mediated dermatitis and is usually localized to the nasal planum. DLE does not evolve to generalized disease, however lesions may spread to the bridge of the nose and less commonly may extend to periocular region, pinnae, distal limbs, and mucocutaneous junctions (lips, oral cavity, and genital region). A 4-year-old male Bavarian Mountain Scenthound developed a chronic, erosive, cutaneous lesion located exclusively in the perianal region without facial skin involvement. Clinical signs included erythema, depigmentation, severe alopecia, crusting, and ulceration. Histologically, the hallmarks of the changes were an interface dermatitis consisting of plasma cells, lymphocytes, neutrophils, and macrophages, hydropic degeneration of basal cells, few apoptotic cells in the basal layer, pigmentary incontinence, and a focal thickening of the basement membrane, which was characterized by linear deposition of IgG. Despite the unusual localization the lesion was diagnosed as DLE based on the characteristic histologic and immunohistologic features. Following diagnosis, corticosteroid therapy resulted in a complete resolution of perianal lesions.  相似文献

A field-based study was carried out to broaden our knowledge of fully mechanized cut-to-length harvesting productivity in naturally grown forests in the northern European part of Russia (NEPR). The recorded data comprised 38 midsized single-grip harvesters (JD 1270D) in clear-cutting operations in the Karelia, Komi, Vologda, Leningrad, Tver, and Kirov regions in NEPR, 4.3 million felled trees, and 1.4 million m3 u.b. (under bark) of processed timber. Harvesting operations were conducted in forest stands composed of spruce (48% on average), pine (19%), birch (22%), and aspen (11%), with an average stem volume 0.31 m3 u.b. The cut-to-length harvesters produced from 4.3 to 14.9 m3 u.b./productive machine hour (PMH) and 16.0–49.5 m3 u.b./stem processing machine hour (S proc MH). A machine evaluation analysis and a regression analysis were used to formulate models for predicting cutting productivity of modern single-grip harvester. The regression models were developed to estimate the productivity of the harvesters in the regions taking into account two significant factors influencing the productivity: the stem volume and tree species of the felled trees. Productivity/cubic meter u.b. of processed timber/PMH was calculated according to stem volume and tree species distributions in most forest-covered NEPR regions. Further research is suggested to improve the developed productivity models and to allow prediction of system performance over a broad range of stand and site conditions.  相似文献

Wind-induced motion of 29 neighbouring trees growing in a Scots pine plantation was measured over the period 14 March to 24 March 2008. The bi-orthogonal decomposition (BOD) of the complex tree motion field into a limited number of spatio-temporal modes provided the basis for the analysis of the response behaviour to wind excitation of the group of sample trees. It is shown that the first BOD-mode was the most energetic and differed from all other BOD-modes. The BOD-results as well as the results from wavelet analysis of the temporal eigenvectors of the BOD-modes demonstrate that two concomitant low-frequency components in the streamwise wind velocity component stimulated coherent response of the sample trees at the tree group level. However, in the range of the wind speed measured close to the top of the Scots pine forest canopy (hourly mean wind speed values lower than 6 m s−1), the wind loads associated with these low-frequency airflow structures were too low to harmonise the motion of the sample trees completely. It is shown that instantaneous single tree responses to wind excitation were highly irregular in magnitude and direction. Results from Fourier and wavelet analysis demonstrate that sway in the first mode dominated the wind-induced sway behaviour at the tree level.  相似文献

Forest liming is a common measure to counteract soil acidification. In forest practice, lime is applied to the forest floor where it changes the chemical properties. However, little is known about the depth impact of liming and the depth translocation of lime components. To investigate the long-term impact of forest liming, several study plots have been established in the 1980s in Germany in stands with different site conditions. We analysed soil chemical data obtained during the last 28?years from 45 of the study plots. We examined the depth impact of liming and predicted the main factors responsible for the increase in Calcium (Ca) and Magnesium (Mg) stocks after liming in the mineral soil using multiple linear regression analyses (MLR). Stocks of Ca and Mg as well as base saturation (BS) showed a strong depth gradient with significant differences between limed and control plots down to 40?cm of the mineral soil. About 65–70?% of applied Ca and Mg were recovered in the forest floor and the upper 40?cm of the mineral soil. BS in 0–40?cm increased by a mean of 11?%. MLR models could explain 48–74?% of the variation in mean changes of Ca and Mg in 0–10, 10–20 and 20–40?cm soil depth when soil and climate variables, amount of applied lime and years after liming are included in the model. After testing the model robustness with a cross-validating procedure, we concluded that these models might be applied to many regions in Central Europe with comparable soil and climate conditions and thus, have widespread application.  相似文献

Innate immune responses are vital for pathogen defense but can result in septic shock when excessive. A key mediator of septic shock is tumor necrosis factor-α (TNFα), which is shed from the plasma membrane after cleavage by the TNFα convertase (TACE). We report that the rhomboid family member iRhom2 interacted with TACE and regulated TNFα shedding. iRhom2 was critical for TACE maturation and trafficking to the cell surface in hematopoietic cells. Gene-targeted iRhom2-deficient mice showed reduced serum TNFα in response to lipopolysaccharide (LPS) and could survive a lethal LPS dose. Furthermore, iRhom2-deficient mice failed to control the replication of Listeria monocytogenes. Our study has identified iRhom2 as a regulator of innate immunity that may be an important target for modulating sepsis and pathogen defense.  相似文献
