An obscure sesquiterpene, rotundone, has been identified as a hitherto unrecognized important aroma impact compound with a strong spicy, peppercorn aroma. Excellent correlations were observed between the concentration of rotundone and the mean 'black pepper' aroma intensity rated by sensory panels for both grape and wine samples, indicating that rotundone is a major contributor to peppery characters in Shiraz grapes and wine (and to a lesser extent in wine of other varieties). Approximately 80% of a sensory panel were very sensitive to the aroma of rotundone (aroma detection threshold levels of 16 ng/L in red wine and 8 ng/L in water). Above these concentrations, these panelists described the spiked samples as more 'peppery' and 'spicy'. However, approximately 20% of panelists could not detect this compound at the highest concentration tested (4000 ng/L), even in water. Thus, the sensory experiences of two consumers enjoying the same glass of Shiraz wine might be very different. Rotundone was found in much higher amounts in other common herbs and spices, especially black and white peppercorns, where it was present at approximately 10000 times the level found in very 'peppery' wine. Rotundone is the first compound found in black or white peppercorns that has a distinctive peppery aroma. Rotundone has an odor activity value in pepper on the order of 50000-250000 and is, on this criterion, by far the most powerful aroma compound yet found in that most important spice.  相似文献

Hydromorphic soils should exhibit higher climate change feedback potentials than well aerated soils since soil organic matter (SOM) losses in them are predicted to be much larger than those of well aerated soils. To evaluate a combined feedback relationship between groundwater level (GWL) and total greenhouse gas (GHG) emission, a greenhouse microcosm experiment was performed by exposing three hydromorphic forest soil types that differed in carbon content to three water levels (?40, ?20 and ?5 cm) while plants were excluded. Net GHG fluxes were measured continuously. GHG concentrations plus oxygen were measured in soil air and soil water at different depths. In this study, soil type hardly affected GHG emissions but GWL did. CO2 emissions peaked at GWL of ?40 cm and declined on average to 65 and 33% during GWL at ?20 and ?5 cm, respectively. CH4 emissions showed the opposite pattern having the highest emission rates at GWL of ?5 cm and compared to that on average only ?3 and ?8% during GWL at ?20 and ?40 cm, respectively. The highest mean N2O emissions were detected at the intermediate GWL of ?20 cm, whereas it is reduced on average to 18% for GWL at ?40 cm and at ?5 cm. The highest greenhouse gas emissions (in CO2 equivalents) were calculated for GWL at ?20 cm. During GWL at ?40 cm, CO2 equivalent fluxes were only insignificantly lower. CO2 equivalent fluxes reduced explicitly in mean to 35% with GWL at ?5 cm. The outcome emphasizes that anaerobic SOM decomposition apparently produces a lower warming potential than aerobic SOM decomposition. Undoubtedly, hydromorphic soils have to be considered for climate–carbon feedback scenarios.  相似文献

Current theory expects that fungi, on the one hand, are spatially ubiquitous but, on the other, are more susceptible than bacteria to disturbance such as land use change due to dispersal limitations. This study examined the relative importance of location and land use effects in determining soil fungal community composition in south-eastern Australia. We use terminal restriction fragment length polymorphism (T-RFLP; primer pair ITS1-F–ITS4) and multivariate statistical methods (NMDS ordinations, ANOSIM tests) to compare relative similarities of soil fungal communities from nine sites encompassing three locations (ca 50–200 km apart) and four land uses (native eucalypt forest, Pinus radiata plantation, Eucalyptus globulus plantation, and unimproved pasture). Location effects were generally weak (e.g. ANOSIM test statistic R  0.49) and were, in part, attributed to minor differences in soil texture. By contrast, we found clear and consistent evidence of land use effects on soil fungal community composition (R  0.95). That is, soils from sites of the same land use grouped together in NMDS ordinations of fungal composition despite geographic separations of up to ca 175 km (native eucalypt forests) and 215 km (P. radiata plantations). In addition, different land uses from the same location were clearly separate in NMDS ordinations, despite, in one case, being just 180 m apart and having similar land use histories (i.e. P. radiata versus E. globulus plantation both established on pasture in the previous decade). Given negligible management of all sites beyond the early establishment phase, we attribute much of the land use effects to changes in dominant plant species based on consistent evidence elsewhere of strong specificity in pine and eucalypt mycorrhizal associations. In addition, weak to moderate correlations between soil fungal community composition and soil chemical variables (e.g. Spearman rank correlation coefficients for individual variables of 0.08–0.32), indicated a minor contributing role of vegetation-mediated changes in litter and soil chemistry. Our data provide evidence of considerable plasticity in soil fungal community composition over time spans as short as 6–11 years. This suggests that – at least within geographic zones characterised by more-or-less contiguous forest cover – soil fungal community composition depends most on availability of suitable habitat because dispersal propagules are readily available for colonisation after land use change.  相似文献

In mammals, insulin regulates S6K1, a key enzyme involved in the control of protein synthesis, via the well-documented phosphoinositide-3'kinase (PI3K) pathway. Conversely, S6K1 is activated by insulin in avian muscle despite the relative insulin insensitivity of the PI3K pathway in this tissue. Mitogen-activated protein kinase (MAPK) cascade is another insulin sensitive pathway. The aim of this study was to explore the potential involvement of the ERK1/2 MAPK pathway in the control of p70 S6 kinase (S6K1) in avian species. Firstly, we characterized ERK1/2 MAPK in various chicken tissues. ERK2 was the only isoform detected in avian species whatever the tissue studied. We also showed that ERK2 is activated in vivo by insulin in chicken muscle. The regulation and the role of ERK2 in insulin signaling were next investigated in chicken hepatoma cells (LMH) and primary myoblasts. Insulin stimulation led to ERK2 and S6K1 phosphorylation, and concomitantly increased kinase activity. U0126, an inhibitor of the ERK MAPK pathway, completely abolished insulin-induced S6K1 phosphorylation and activity in chicken myoblasts, whereas its effect was only partial in LMH cells. In conclusion, these results show that ERK1/2 MAPK is involved in the control of S6K1 by insulin in chicken cells, particularly myoblasts.  相似文献

Objective

To compare the effects of MK-467 during isoflurane anaesthesia combined with xylazine or dexmedetomidine on global and gastrointestinal perfusion parameters.

Study design

Prospective, randomized experimental trial.

Animals

A total of 15 warmblood horses.

Methods

Horses were divided into two groups for administration of either dexmedetomidine (D) or xylazine (X) for premedication (D: 3.5 μg kg?1; X: 0.5 mg kg?1) and as constant rate infusion during isoflurane anaesthesia (D: 7 μg kg?1 hour?1; X: 1 mg kg?1 hour?1). During anaesthesia, heart rate, mean arterial blood pressure (MAP), systemic vascular resistance index (SVRI) and cardiac index (CI) were measured. Microperfusion of the colon, jejunum and stomach was measured using laser Doppler flowmetry. After 2 hours of stabilization, MK-467 (250 μg kg?1) was administered, and measurements were continued for another 90 minutes. For statistical analysis, the permutation test and Wilcoxon rank-sum test were used (p < 0.05).

Results

There were no differences in baseline measurements between groups. The MK-467 bolus resulted in a significant decrease in MAP (D: –58%; X: –48%) and SVRI (D: –68%; X: –65%) lasting longer in group D (90 minutes) compared to group X (60 minutes). While CI increased (D: +31%; X: +35%), microperfusion was reduced in the colon (D: –44%; X: –34%), jejunum (D: –26%; X: –33%) and stomach (D: –37%; X: –35%).

Conclusions and clinical relevance

Alpha-2-agonist induced vasoconstriction was reversed by the MK-467 dose used, resulting in hypotension and rise in CI. Gastrointestinal microperfusion decreased, probably as a result of insufficient perfusion pressure. An infusion rate for MK-467 as well as an ideal agonist/antagonist ratio should be determined.  相似文献

The effects of two organic amendments, a municipal solid waste compost and a composted straw, on [U-ring- ]atrazine degradation pathways in a loamy soil (Grignon, Yvelines, France) were studied during laboratory incubations under controlled conditions. Three month conditionings were conducted under sterile or non-sterile conditions, with soil plus atrazine, organic amendment plus atrazine, or soil plus organic amendment. Then either an organic amendment or soil or atrazine was added, respectively, to these three treatments and incubated for an additional 3 months under non-sterile conditions. Both organic amendments modified the behaviour of atrazine in soil but via different processes. The addition of municipal compost increased atrazine sorption and decreased its availability for degradation by soil microorganisms. The effect of the composted straw was mainly related to its high enzymatic activity, which appeared to be responsible for the production of large amounts of hydroxyatrazine. This hydroxylation favoured the opening of the triazine ring and its subsequent mineralization in the soil. At the end of the incubations, less atrazine was mineralized in the presence of the two types of organic amendment, which both increased the formation of non-extractable residues of atrazine. The addition of municipal compost preserved larger amounts of extractable atrazine, while the addition of composted straw enhanced hydroxyatrazine production. In all cases, the greatest effects were found when atrazine was directly in contact with the organic amendment during conditioning.  相似文献
