This study aimed to describe successful cryopreservation of sperm from maned wolves (Chrysocyon brachyurus). Three ejaculates from 2 maned wolves were collected by digital manipulation of the penis and evaluated subjectively, centrifuged and frozen in BotuCrio® (Botupharma, Botucatu, Brazil) or Tris–yolk egg extender. Spermatozoa were thawed at 37ºC/30s or 70ºC/4s and evaluated for kinetics, morphology, plasma and acrosome membrane integrity, mitochondrial potential, hydrogen peroxide, superoxide anion and lipid peroxidation. From 5 thawed samples, two had sperm total motility >55% (56.0% and 64.0%) and progressive motility ~35% (35% and 40%), both frozen with Tris–yolk egg. Plasma and acrosome membrane integrity decreased and percentage of sperm defects increased post-thawing. We concluded that is possible to freeze spermatozoa from maned wolves using semen collection and processing methods applied for domestic dogs.  相似文献

This experiment was designed to evaluate the effects of different concentrate crude protein (CP) concentration on performance, metabolism and efficiency of N utilization (ENU) on early-lactation dairy cows grazing intensively managed tropical grass. Thirty cows were used in a ten replicated 3 × 3 Latin square design. The treatments consisted of three levels of concentrate CP: 7.9%, 15.4%, and 20.5% offered at a rate of 1 kg (as-fed basis)/3 kg of milk. The cows fed low and medium CP had negative balance of rumen degradable protein and metabolizable protein. Increasing CP tended to linearly increase DMI, 3.5% FCM and milk casein, and linearly increased the yields of milk fat and protein. Increasing CP linearly increased the intake of N, the concentration of rumen NH3–N, and the losses of N in milk, urine, and feces. Increasing dietary CP linearly increased the molar proportion of butyrate but had no effect on the other rumen VFAs and no effect on microbial yield. In conclusion, feeding a concentrate with 20.5% of CP to early-lactation dairy cows grazing tropical grasses, leading to a 17.8% CP diet, tended to increase DMI, increased the yield of 3.5% FCM and the milk N excretion, and decreased ENU by 32%.  相似文献

Carcass characteristic and meat quality from bulls and Nellore steers (n?=?64 total) subjected to different grazing heights (15, 25, 35, and 45 cm) were evaluated isolatedly, under continuous grazing and variable load, in pastures of Convert grass. The experimental period was from May 2015 to June 2017, in an area of 16 ha, divided into 16 paddocks of 1 ha. The experimental design was randomized blocks with four replications. Each paddock was grazed by three animals and regulators, used to adjust grazing heights. The final slaughter weight, hot carcass, and crude protein in the meat of steers were higher when the pastures were managed at 42 cm. When the pasture was managed between 25 and 35 cm in height, greater fat thickness, marbling, muscle:bone and muscle+fat:bone ratio and lower color* of the meat and percentage of bone were found. For the steers, the height of 40 cm provided higher fat thickness and marbling in the meat. The loss during thawing in meat was greater at 28 cm in height. The heights of grazing alter the carcass characteristics and meat of bulls and steers.

Carlaviruses are plant-infecting viruses with flexuous filamentous particles of approximately 650 nm in length and a positive single-stranded hexacistronic RNA molecule as the genome. In this study, we analysed 14 samples of brassicas plants, reportedly affected by carlaviruses, that were collected in distant and edaphoclimatic distinct regions in Brazil. The genomes of four viral isolates detected in leaf kale (Brassica oleracea var. acephala) plants displayed the typical genomic organization of carlaviruses, subdivided into two contrasting identity profiles. Isolates T25, T107, and T110 showed a higher sequence identity among them (c.93%) than with T90 (c.67%), which had the highest nucleotide sequence identity (85.2%) with the only documented genomic fragment, approximately 1 kb of the RNA 3′-end, of cole latent virus (CoLV). Identity values among the other three isolates and CoLV were consistently lower than 78.6%. Nucleotide and amino acid sequence identity values of the replicase cistron of the isolates T25 and T90 are below the threshold for species demarcation in the genus Carlavirus. Upon mechanical transmission, these two isolates induced different symptoms in brassicas and solanaceous plants. Overall, data indicated that these viruses belong to two related but distinct species of carlaviruses. While T90 was recognized as an isolate of CoLV, the remaining isolates were considered members of a new tentative carlavirus named Cole mild mosaic virus (CoMMV). Current work provides convincing support for the taxonomic status of the species Cole latent virus, enlarges the known diversity of carlaviruses, and makes available new molecular tools to improve surveys of brassica-infecting viruses.  相似文献

Pigeons (Columba livia) cohabit with humans in urban and rural areas, representing a public health problem since microorganisms are transmitted through the inhalation of dust from their dry feces (chlamydiosis) and through ingestion of their undercooked or poorly refrigerated meat (toxoplasmosis). This study aimed to evaluate the presence of Chlamydophila psittaci and Toxoplasma gondii in pigeons from four cities in São Paulo State, Brazil. C. psittaci was evaluated through hemi-nested polymerase chain reaction (hnPCR) using cloacal and tracheal swabs, whereas T. gondii specific antibodies were assessed by means of modified agglutination test (MAT), mouse brain and muscle bioassay, and polymerase chain reaction (PCR). To confirm the infection in mice, T. gondii antibodies were assayed by using indirect fluorescent antibody test (IFAT). Considering C. psittaci, 40/238 (16.8%; 95%CI 12.6–22.1%) samples were positive according to hnPCR, especially for the cities of São Paulo (42.5%) and Bauru (35%). As regards T. gondii, 12/238 (5%; 95%CI 2.9–8.6%) serum samples were positive according to MAT. Of these, five samples had titer equal to 1:8; six samples, 1:16; and one sample, 1:32. Bioassay, IFAT and PCR were negative for mouse toxoplasmosis. The absence of T. gondii antibodies suggests that pigeons may be infected with a low concentration of the agent, not detected by the antigen test. Thus, C. psittaci represents an actual problem concerning bird health.  相似文献

In the dry regions of Chile, prolific flowering from forest plantation is particularly advantageous for honey production, in order to supplement the erratic flowering in native plants. Eucalyptus cladocalyx is a species suitable for areas with low water availability and their flowers provide a reliable source for the production of honey. The aim of this study was to examine the heritability of flowering intensity in 49 open-pollinated families of E. cladocalyx in southern Atacama Desert, Chile, with the view to the selection for prolific flowering, but with minimal impact on precocious flowering. The Bayesian variance component estimation model was assumed using the Gibbs sampling algorithm. Threshold models were fitted to flowering data (bi-character model). Flowering intensity was found to be highly heritable (posterior mean: h 2 = 0.48; and credible interval: 0.41–0.56). The posterior mean of the genetic correlation between flowering precocity and intensity was positive (r = 0.45) and according to the credible interval (0.341–0.542), it was significantly different from zero, indicating that selection on breeding values of early flowering at age three, would have significant and positive impact on flowering intensity 5 years later (or in 8-year-old trees). These results are important for the start of a small-scale breeding program for the species in southern Atacama Desert. The genetic variability found in these breeding populations may be used for breeding purposes in regions where arid environmental conditions are limiting to the establishment of eucalypts trees.  相似文献

  1. The extent and intensity of impacts of multiple new dams in the Amazon basin on specific biological groups are potentially large, but still uncertain and need to be better understood.
  2. It is known that river disruption and regulation by dams may affect sediment supplies, river channel migration, floodplain dynamics, and, as a major adverse consequence, are likely to decrease or even suppress ecological connectivity among populations of aquatic organisms and organisms dependent upon seasonally flooded environments.
  3. This article complements our previous results by assessing the relationships between dams, our Dam Environmental Vulnerability Index (DEVI), and the biotic environments threatened by the effects of dams. Because of the cartographic representation of DEVI, it is a useful tool to compare the potential hydrophysical impacts of proposed dams in the Amazon basin with the spatial distribution of biological diversity. As the impact of Amazonian dams on the biota of both rivers and periodically flooded riparian environments is severe, DEVIs from different Amazonian tributary basins are contrasted with patterns of diversity and distribution of fish, flooded forest trees and bird species.
  4. There is a consistent relationship between higher DEVI values and the patterns of higher species richness and endemism in all three biological groups. An assessment of vulnerability at the scale of tributary basins, the assessment of biodiversity patterns related to DEVI, and the analysis of teleconnections at basin scale, demonstrate that recent construction of dams is affecting the biota of the Amazon basin.
  5. The evidence presented here predicts that, if currently planned dams are built without considering the balance between energy production and environmental conservation, their cumulative effects will increase drastically and represent a major threat to Amazonian biodiversity.

Derelict fishing gear is a highly visible source of marine plastic pollution, causing mortality and ecosystem degradation with uncertain long-term consequences. The quantity of derelict gear entering the oceans remains unknown because of heterogeneity in fishing gear and effort, as well as inadequate monitoring. Prior studies have been limited in scope to specific fisheries and regions, and large-scale estimates lack an empirical basis. It is critically important for decision makers to have credible information in order to design effective remediation efforts. We estimated the amount of industrial fishing effort and the associated plastic debris entering the ocean globally each year from lost fishing gear. Using remote observations of fishing vessel activity paired with technical fishing gear models, we generated a bounding estimate for gear operation and loss worldwide in 2018. We estimate that industrial trawl, purse-seine and pelagic longline fisheries operated 2.1 Mt of plastic gear over 2018 to obtain 49.7 Mt of retained and discarded catch, representing 74% of industrial marine capture globally. The median estimate for plastic gear lost during the use of these gear types was 48.4 kt (95% confidence interval: 28.4–99.5 kt). This estimate excludes abandoned and discarded gear. Improved observation, especially of small-scale fisheries, is needed to better understand the sources of derelict gear. These findings serve as a benchmark for future monitoring and management efforts to reduce derelict gear in the global ocean.  相似文献
